    Galectin-3-mediated glial crosstalk drives oligodendrocyte differentiation and (Re)myelination

    Galectin-3 (Gal-3) is the only chimeric protein in the galectin family. Gal-3 structure comprises unusual tandem repeats of proline and glycine-rich short stretches bound to a carbohydrate-recognition domain (CRD). The present review summarizes Gal-3 functions in the extracellular and intracellular space, its regulation and its internalization and secretion, with a focus on the current knowledge of Gal-3 role in central nervous system (CNS) health and disease, particularly oligodendrocyte (OLG) differentiation, myelination and remyelination in experimental models of multiple sclerosis (MS). During myelination, microglia-expressed Gal-3 promotes OLG differentiation by binding glycoconjugates present only on the cell surface of OLG precursor cells (OPC). During remyelination, microglia-expressed Gal-3 favors an M2 microglial phenotype, hence fostering myelin debris phagocytosis through TREM-2b phagocytic receptor and OLG differentiation. Gal-3 is necessary for myelin integrity and function, as evidenced by myelin ultrastructural and behavioral studies from LGALS3-/- mice. Mechanistically, Gal-3 enhances actin assembly and reduces Erk 1/2 activation, leading to early OLG branching. Gal-3 later induces Akt activation and increases MBP expression, promoting gelsolin release and actin disassembly and thus regulating OLG final differentiation. Altogether, findings indicate that Gal-3 mediates the glial crosstalk driving OLG differentiation and (re)myelination and may be regarded as a target in the design of future therapies for a variety of demyelinating diseases.Fil: Thomas, Laura.     Galectin-1 in myelin repair

    Galectin-1 (Gal-1) is a member of a highly conserved family of animal lectins which binds to the common disaccharide [Galβ(1-4)-GlcNAc] on both N- and O-glycans decorating cell surface glycoconjugates. Current evidence supports a role for Gal-1 in the pathophysiology of multiple sclerosis (MS), one of the most prevalent chronic inflammatory diseases, as approximately one third of MS patients generate high titres of anti-Gal-1 antibodies. Four different lesion types have been described in MS: pattern-1 and -2 lesions are thought to be mediated by the autoimmune response, while pattern-3 and -4 lesions are considered primary oligodendropathy. The first two types are experimentally simulated in mice by experimental autoimmune encephalomyelitis (EAE), while the second two are mimicked by toxic models such as cuprizone (CPZ) or lysolecithin (LPC) administration. Studies in EAE models have demonstrated that Gal-1 is highly expressed in the acute phase of the disease and that its deficiency leads to severe inflammation-induced neurodegeneration [1].     Extraction of proton form factors in the timelike region from unpolarized e+e- --> p pbar events

    We have performed numerical simulations of the unpolarized e+e- --> p pbar process in kinematic conditions under discussion for a possible upgrade of the existing DAFNE facility. By fitting the cross section angular distribution with a typical Born expression, we can extract information on the ratio |G_E/G_M| of the proton electromagnetic form factors in the timelike region within a 5-10% uncertainty. We have explored also non-Born contributions to the cross section by introducing a further component in the angular fit, which is related to two-photon exchange diagrams. We show that these corrections can be identified if larger than 5% of the Born contribution, and if relative phases of the complex form factors do not produce severe cancellations.Comment: 10 pages in RevTeX style; 8 figures in eps forma

    Axonal regeneration in spinal cord injury: a key role of galectin-1

    Al producirse una lesión de médula espinal (LME), un sinnúmero de proteínas inhibidoras de la regeneración axonal ocupan el sitio de lesión en forma secuencial. La primer proteína en llegar al mismo se conoce como semaforina 3A (Sema3A), siendo además una de las más potentes por su acción de inhibir la regeneración axonal. A nivel mecanístico la unión de esta proteína al complejo-receptor neuronal neuropilin-1 (NRP-1)/PlexinA4 evita que se produzca regeneración axonal. En este trabajo de revisión se discutirá la acción de galectin-1 (Gal-1), una proteína endógena de unión a glicanos, que selectivamente se une al complejo-receptor NRP-1/PlexinA4 de las neuronas lesionadas a través de un mecanismo dependiente de interacciones lectina-glicano, interrumpiendo la señalización generada por Sema3A y permitiendo de esta manera la regeneración axonal y recuperación locomotora luego de producirse la LME. Mientras ambas formas de Gal-1 (monomérica y dimérica) contribuyen a la inactivación de la microglia, solo la forma dimérica de Gal-1 es capaz deunirse al complejo-receptor NRP-1/PlexinA4 y promover regeneración axonal. Por lo tanto, Gal-1 dimérica produce recuperación de las lesiones espinales interfiriendo en la señalización de Sema3A a través de la unión al complejoreceptor NRP-1/PlexinA4, sugiriendo el uso de esta lectina en su forma dimérica para el tratamiento de pacientes con LME.When spinal cord injury (SCI) occurs, a great number of inhibitors of axonal regeneration consecutively invade the injured site. The first protein to reach the lesion is known as semaphorin 3A (Sema3A), which serves as a powerful inhibitor of axonal regeneration. Mechanistically binding of Sem3A to the neuronal receptor complex neuropilin-1 (NRP1) / PlexinA4 prevents axonal regeneration. In this special article we review the effects of galectin-1 (Gal-1), an endogenous glycan-binding protein, abundantly present at inflammation and injury sites. Notably, Gal1 adheres selectively to the NRP-1/PlexinA4 receptor complex in injured neurons through glycan-dependent mechanisms, interrupts the Sema3A pathway and contributes to axonal regeneration and locomotor recovery after SCI. While both the monomeric and dimeric forms of Gal-1 contribute to ’switch-off’ classically-activated microglia, only dimeric Gal-1 binds to the NRP-1/PlexinA4 receptor complex and promotes axonal regeneration. Thus, dimeric Gal-1 promotes functional recovery of spinal lesions by interfering with inhibitory signals triggered by Sema3A adhering to the NRP-1/PlexinA4 complex, supporting the use of dimeric Gal-1 for the treatment of SCI patients.Fil: Quintá, Héctor Ramiro.     Neuropsychiatric disturbances in atypical Parkinsonian disorders

    Multiple system atrophy (MSA), progressive supranuclear palsy (PSP), and corticobasal degeneration (CBD) are the most common atypical parkinsonisms. These disorders are characterized by varying combinations of autonomic, cerebellar and pyramidal system, and cognitive dysfunctions. In this paper, we reviewed the evidence available on the presence and type of neuropsychiatric disturbances in MSA, PSP, and CBD. A MedLine, Excerpta Medica, PsycLit, PsycInfo, and Index Medicus search was performed to identify all articles published on this topic between 1965 and 2018. Neuropsychiatric disturbances including depression, anxiety, agitation, and behavioral abnormalities have been frequently described in these disorders, with depression as the most frequent disturbance. MSA patients show a higher frequency of depressive disorders when compared to healthy controls. An increased frequency of anxiety disorders has also been reported in some patients, and no studies have investigated apathy. PSP patients may have depression, apathy, disinhibition, and to a lesser extent, anxiety and agitation. In CBD, neuropsychiatric disorders are similar to those present in PSP. Hallucinations and delusions are rarely reported in these disorders. Neuropsychiatric symptoms in MSA, PSP, and CBD do not appear to be related to the severity of motor dysfunction and are one of the main factors that determine a low quality of life. The results suggest that neuropsychiatric disturbances should always be assessed in patients with atypical parkinsonisms

    Inhalation of growth factors and apo-transferrin to protect and repair the hypoxic-ischemic brain

    Hypoxic-ischemic brain damage is a major contributor to chronic neurological dysfunction and acute mortality in infants as well as in adults. In this review, we summarize recent publications demonstrating that the intranasal administration (INA) of apo-transferrin (aTf) and different growth factors provides neuroprotection to the mouse and rat brain after a hypoxic-ischemic event. The intranasal delivery of growth factors such as insulin-like growth factor-1 (IGF-1) and vascular endothelial growth factor (VEGF) has been found to improve neurological function and reduce infarct size in adult rats after a hypoxic-ischemic event. On the other hand, INA of aTf and epidermal growth factor (EGF) were effective in reducing white matter damage and inflammation and in promoting the proliferation and survival of oligodendroglial progenitor cells (OPCs) in a model of hypoxic-ischemic encephalopathy. Therefore, data summarized in this review suggest that INA of growth factors and aTf can be used in combination in clinical treatment in order to protect and repair the hypoxic-ischemic brain.Fil: Guardia Clausi, Mariano.     Geochemistry of a Late Quaternary loess-paleosol sequence in central Argentina: Implications for weathering, sedimentary recycling and provenance

    A loess-paleosol sequence in the Pampean Plain, central Argentina was analyzed geochemically, in order to evaluate the weathering signature and sedimentary recycling as well as to analyze the origin of these loessic sediments. Corralito I sequence is composed of three paleosols and a buried soil separated by loess layers. The loess-paleosol sequence shows a similar geochemical composition throughout, suggesting that pedogenesis has not been intense enough to mask the chemical signature of the parent material. Weathering indices (CIA, CIW, PIA and LWI), elemental ratios (Rb/Sr, CaO/TiO2 and Na2O/TiO2), and the A-CN-K ternary diagram indicate incipient chemical alteration for this sequence, compatible with a weathering-limited regime. As expected, paleosols (CIAs ~60)exhibit a slightly higher chemical alteration than loess layers (CIAs ~58). Mass balance calculations reveal the losses of some major oxides (i.e., CaO, Na2O)and Sr in the buried soil and paleosols compared to its loess mantles, reflecting the weathering of plagioclase. Besides, the losses of K2O and Ba in the buried soil and paleosols I and II, when compared to its loess layers, suggest the chemical alteration of K-feldspar. The chemical alteration of volcanic glass is evidenced by the relative losses of As in almost all levels. Conversely, the relative gains of Fe2O3 and MgO reveal that pyroxenes and amphiboles or Fe oxy-hydroxides are concentrated in paleosols and the buried soil compared to its loess mantles. The geochemical approach used to constrain the origin of these Pampean Plain loessic sediments indicates they derive mainly from the Andean region. Conversely, the Pampean ranges, the Paraná River basin and Uruguayan Precambrian outcrops, which were pointed out as minor local sources, do not contribute significantly to the loess deposits. This work also shows that Corralito I loess-paleosol sequence and other loess samples from Argentina do not evidence sedimentary recycling, suggesting that these Pampean Plain loess deposits are mainly composed by young materials derived from undifferentiated volcanic rocks.Fil: Campodonico, Verena Agustina.     Geoquímica de una secuencia de loess-paleosuelos del cuaternario tardío en el centro de Argentina

    A loess-paleosol sequence in central Argentina was geochemically analyzed. Corralito I sequence (32°00‘7” S, 64°11’08” W, 469 m a.s.l) is composed by three paleosols and a buried soil separated by loess layers. Weathering indices (CIA, CIW, LWI), elemental ratios (ΣBases/Al2O3, Rb/Sr, CaO/TiO2, Na2O/TiO2), and mean annual paleo-precipitation indices (MAPPs) were calculated in order to obtain a weathering insight and to probe geochemical paleoclimatic proxies. The results indicate an incipient weathering degree throughout the sequence, without significant geochemical differences between paleosols and the loess mantles. Moreover, geochemical paleoclimatic proxies (MAPPs) do not detect a significant paleoprecipitation variability for the time period recorded in the studied loess-paleosol sequence.Centro de Investigaciones Geológica

    Prevalence and risk factors for thermotolerant species of Campylobacter in poultry meat at retail in Europe

    Abstract The thermotolerant species Campylobacter jejuni, Campylobacter coli, Campylobacter lari and Campylobacter upsaliensis are the causative agents of the human illness called campylobacteriosis. This infection represents a threat for the health of consumers in Europe. It is well known that poultry meat is an important food vehicle of Campylobacter infection. As emerged from the reported scientific literature published between 2006 and 2016, poultry meat sold at retail level in Europe represents an important source of the pathogen. The contamination level of poultry meat sold at retail can vary depending on pre- and post-harvest factors. Among the pre-harvest measures, strict biosecurity practices must be guaranteed; moreover, among post-harvest control measures scalding, chilling and removal of faecal residues can reduce the contamination level of Campylobacter. An additional issue is represented by increasing proportion of Campylobacter isolates resistant to tetracyclines, ciprofloxacin, and nalidixic acid, thus feeding a serious concern on the effectiveness of antibiotic treatment for human campylobacteriosis in a near future
