    Comprehensive systematic review summary: Treatment of tics in people with Tourette syndrome and chronic tic disorders

    Objective To systematically evaluate the efficacy of treatments for tics and the risks associated with their use. Methods This project followed the methodologies outlined in the 2011 edition of the American Academy of Neurology\u27s guideline development process manual. We included systematic reviews and randomized controlled trials on the treatment of tics that included at least 20 participants (10 participants if a crossover trial), except for neurostimulation trials, for which no minimum sample size was required. To obtain additional information on drug safety, we included cohort studies or case series that specifically evaluated adverse drug effects in individuals with tics. Results There was high confidence that the Comprehensive Behavioral Intervention for Tics was more likely than psychoeducation and supportive therapy to reduce tics. There was moderate confidence that haloperidol, risperidone, aripiprazole, tiapride, clonidine, onabotulinumtoxinA injections, 5-ling granule, Ningdong granule, and deep brain stimulation of the globus pallidus were probably more likely than placebo to reduce tics. There was low confidence that pimozide, ziprasidone, metoclopramide, guanfacine, topiramate, and tetrahydrocannabinol were possibly more likely than placebo to reduce tics. Evidence of harm associated with various treatments was also demonstrated, including weight gain, drug-induced movement disorders, elevated prolactin levels, sedation, and effects on heart rate, blood pressure, and ECGs. Conclusions There is evidence to support the efficacy of various medical, behavioral, and neurostimulation interventions for the treatment of tics. Both the efficacy and harms associated with interventions must be considered in making treatment recommendations

    Practice Guideline Recommendations Summary: Treatment of Tics in People with Tourette Syndrome and Chronic Tic Disorders

    Objective To make recommendations on the assessment and management of tics in people with Tourette syndrome and chronic tic disorders. Methods A multidisciplinary panel consisting of 9 physicians, 2 psychologists, and 2 patient representatives developed practice recommendations, integrating findings from a systematic review and following an Institute of Medicine–compliant process to ensure transparency and patient engagement. Recommendations were supported by structured rationales, integrating evidence from the systematic review, related evidence, principles of care, and inferences from evidence. Results Forty-six recommendations were made regarding the assessment and management of tics in individuals with Tourette syndrome and chronic tic disorders. These include counseling recommendations on the natural history of tic disorders, psychoeducation for teachers and peers, assessment for comorbid disorders, and periodic reassessment of the need for ongoing therapy. Treatment options should be individualized, and the choice should be the result of a collaborative decision among patient, caregiver, and clinician, during which the benefits and harms of individual treatments as well as the presence of comorbid disorders are considered. Treatment options include watchful waiting, the Comprehensive Behavioral Intervention for Tics, and medication; recommendations are provided on how to offer and monitor these therapies. Recommendations on the assessment for and use of deep brain stimulation in adults with severe, treatment-refractory tics are provided as well as suggestions for future research

    Polylactic acid as a promising sustainable plastic packaging for edible oils

    The influence of renewable packaging materials on the oxidative stability of sunflower oil was investigated to evaluate whether they could be used as alternatives to conventional plastics. Two renewable bottle materials, polylactic acid (PLA) and bio-polyethylene (Green-PE) were compared to conventional plastics consisting of virgin and recycled polyethylene terephthalate (PET, r-PET) and regular polyethylene (PE), in a storage study over a period of 56 days. The results showed that the progress of lipid oxidation in PLA was similar to PET and r-PET until day 28, while it was significantly increased in PE and Green-PE. Benzene was detected as the only migration compound in the oil stored in PET and r-PET, with concentrations of 0.153 ± 0.027 µg/g and 0.187 ± 0.024 µg/g after 56 days of storage. The study concluded that PLA could be used as an alternative packaging material for edible oils to replace PET.This work was kindly funded by the Austrian Science Fund (FWF): P 34512. J.     Influence of polymorphism within the heme oxygenase-I promoter on overall survival and transplantation-related mortality after allogeneic stem cell transplantation

    AbstractAside from major and minor histocompatibility antigens, genetic polymorphisms of various donor and host genes have been found to be risk factors for graft-versus-host disease and transplantation-related mortality (TRM). The heme oxygenase I (HO-I) protein has been implicated in regulating inflammatory response and has been described as a “protective gene” in solid organ transplantation. In humans, the promoter region displays length polymorphism due to a variable number of GT repeats. Individuals exhibiting 29 or fewer GT repeats express higher levels of HO-I on cellular stress compared with individuals with 30 or more GT repeats. We retrospectively analyzed length polymorphisms of 92 donor–host pairs undergoing allogeneic stem cell transplantation. Our findings demonstrate that mainly donor polymorphism leading to high expression of HO-1 (<30 GT repeats) on stress signals is associated with reduced overall survival, and that TRM is significantly increased in this group. This reduction in survival was most prominent when unrelated donors were used. Polymorphisms of the recipient HO-1 genes did not influence posttransplantation outcomes. We conclude that HO-1 polymorphism represents a new genetic risk factor for TRM and overall survival

    Single molecule MATAC-seq reveals key determinants of DNA replication origin efficiency

    Stochastic origin activation gives rise to significant cell-to-cell variability in the pattern of genome replication. The molecular basis for heterogeneity in efficiency and timing of individual origins is a long-standing question. Here, we developed Methylation Accessibility of TArgeted Chromatin domain Sequencing (MATAC-Seq) to determine single-molecule chromatin accessibility of four specific genomic loci. MATAC-Seq relies on preferential modification of accessible DNA by methyltransferases combined with Nanopore-Sequencing for direct readout of methylated DNA-bases. Applying MATAC-Seq to selected early-efficient and late-inefficient yeast replication origins revealed large heterogeneity of chromatin states. Disruption of INO80 or ISW2 chromatin remodeling complexes leads to changes at individual nucleosomal positions that correlate with changes in their replication efficiency. We found a chromatin state with an accessible nucleosome-free region in combination with well-positioned +1 and +2 nucleosomes as a strong predictor for efficient origin activation. Thus, MATAC-Seq identifies the large spectrum of alternative chromatin states that co-exist on a given locus previously masked in population-based experiments and provides a mechanistic basis for origin activation heterogeneity during eukaryotic DNA replication. Consequently, our single-molecule chromatin accessibility assay will be ideal to define single-molecule heterogeneity across many fundamental biological processes such as transcription, replication, or DNA repair in vitro and ex vivo

    Zukunft Bio 2030: Umsetzung von ausgewählten Maßnahmen zur Stärkung der biologischen Landwirtschaft in Österreich

    Für die Studie „Zukunft Bio 2030“ wurde aufbauend auf der abgeschlossenen Studie „Stärkung der biologischen Landwirtschaft in Österreich bis 2030“ (Kummer et al. 2021) vier Maßnahmen ausgewählt und weitergehend bearbeitet. Das Ziel war es, für diese Maßnahmen erste Umsetzungsschritte einzuleiten. In der Studie wurden folgende Bereiche bearbeitet: • Formulierung von konkreten Vorschlägen zur besseren Vernetzung von Forschung, Beratung und Bildung und zur weiteren Entwicklung von Wissenssystemen zur biologischen Landwirtschaft; • Erstellung eines Konzeptes zur Ernährungsbildung über biologische Lebensmittel für Lehrlinge des Lebensmitteleinzelhandels (LEH) in Zusammenarbeit mit dem LEH; • Erstellung eines Umsetzungskonzepts zum Ausbau des Bio-Anteils in der öffentlichen Verpflegung einer Bundeseinrichtung; • Begleitung der Umsetzung einer rechtsverbindlichen Bio-Kontrolle und Bio-Zertifizierung in der Gastronomie und Hotellerie durch die Vernetzung der maßgeblichen Akteur:innen in einem moderierten Prozess. Im Zuge der Studie wurden insgesamt 93 Expert:innen bzw. Stakeholder in Form von Interviews und Gruppendiskussionen eingebunden. Die Ergebnisse der Studie wurden dem Auftraggeber (Bundesministerium für Land- und Forstwirtschaft, Regionen und Wasserwirtschaft) präsentiert und flossen teilweise in das Bio-Aktionsprogramm 2023+ ein

    The James Webb Space Telescope Mission

    Twenty-six years ago a small committee report, building on earlier studies, expounded a compelling and poetic vision for the future of astronomy, calling for an infrared-optimized space telescope with an aperture of at least 4m4m. With the support of their governments in the US, Europe, and Canada, 20,000 people realized that vision as the 6.5m6.5m James Webb Space Telescope. A generation of astronomers will celebrate their accomplishments for the life of the mission, potentially as long as 20 years, and beyond. This report and the scientific discoveries that follow are extended thank-you notes to the 20,000 team members. The telescope is working perfectly, with much better image quality than expected. In this and accompanying papers, we give a brief history, describe the observatory, outline its objectives and current observing program, and discuss the inventions and people who made it possible.
