What limits supercurrents in high temperature superconductors? A microscopic model of cuprate grain boundaries
The interface properties of high-temperature cuprate superconductors have
been of interest for many years, and play an essential role in Josephson
junctions, superconducting cables, and microwave electronics. In particular,
the maximum critical current achievable in high-Tc wires and tapes is well
known to be limited by the presence of grain boundaries, regions of mismatch
between crystallites with misoriented crystalline axes. In studies of single,
artificially fabricated grain boundaries the striking observation has been made
that the critical current Jc of a grain boundary junction depends exponentially
on the misorientation angle. Until now microscopic understanding of this
apparently universal behavior has been lacking. We present here the results of
a microscopic evaluation based on a construction of fully 3D YBCO grain
boundaries by molecular dynamics. With these structures, we calculate an
effective tight-binding Hamiltonian for the d-wave superconductor with a grain
boundary. The critical current is then shown to follow an exponential
suppression with grain boundary angle. We identify the buildup of charge
inhomogeneities as the dominant mechanism for the suppression of the

Protocol for a randomised, double-blind, placebo-controlled study of grass allergen immunotherapy tablet for seasonal allergic rhinitis: time course of nasal, cutaneous and immunological outcomes
BACKGROUND: Seasonal Allergic Rhinitis is characterised by inflammation of the nasal mucosa upon exposure to common aeroallergens, affecting up to 20-25 % of the population. For those patients whose symptoms are not controlled by standard medical treatment, allergen specific immunotherapy is a therapeutic alternative. Although several studies have shown changes in immunologic responses as well as long term tolerance following treatment with a sublingual allergy immunotherapy tablet, a detailed time course of the early mechanistic changes of local and systemic T and B cell responses and the effects on B cell repertoire in the nasal mucosa have not been fully examined. METHODS/DESIGN: This is a randomized, double-blind, single-centre, placebo controlled, two arm time course study based in the United Kingdom comparing sublingual allergy immunotherapy tablet (GRAZAX(®), ALK-Abello Horsholm, Denmark) plus standard treatment with placebo plus standard treatment. Up to 50 moderate to severe grass pollen allergic participants will be enrolled to ensure randomisation of at least 44. Further, we shall enrol 20 non-atopic volunteers. Screening will be completed before eligible atopic participants are randomised to one of the two treatment arms in a 1 to 1 ratio. The primary endpoint will be the total nasal symptom score assessed over 60 min following grass pollen nasal allergen challenge after 12 months of treatment. Clinical assessments and/or mechanistic analyses on blood, nasal fluid, brushing and biopsies will be performed at baseline at 1, 2, 3, 4 (coinciding with the peak pollen season), 6 and 12 months of treatment. After 12 months of treatment, unblinding will take place. Those atopic participants receiving active treatment will continue therapy for another 12 months followed by a post treatment phase of 12 months. Assessments and collection of biologic samples from these participants will take place again at 24 and at 36 months from the start of treatment. The 20 healthy, non-atopic controls will undergo screening and one visit only coinciding with the 12 month visit for the atopic participants. DISCUSSION: The trial will end in April 2017. Substellar and low-mass dwarf identification with near-infrared imaging space observatories
AIMS: We aim to evaluate the near-infrared colors of brown dwarfs as observed
with four major infrared imaging space observatories: the Hubble Space
Telescope (HST), the James Webb Space Telescope (JWST), the Euclid mission, and
the WFIRST telescope.
METHODS: We used the SPLAT SPEX/ISPEX spectroscopic library to map out the
colors of the M-, L-, and T-type dwarfs. We have identified which color-color
combination is optimal for identifying broad type and which single color is
optimal to then identify the subtype (e.g., T0-9). We evaluated each
observatory separately as well as the narrow-field (HST and JWST) and
wide-field (Euclid and WFIRST) combinations.
RESULTS: The Euclid filters perform poorly typing brown dwarfs and WFIRST
performs only marginally better, despite a wider selection of filters. WFIRST's
W146 and F062 combined with Euclid's Y-band discriminates somewhat better
between broad brown dwarf categories. However, subtyping with any combination
of Euclid and WFIRST observations remains uncertain due to the lack of medium
or narrow-band filters. We argue that a medium band added to the WFIRST filter
selection would greatly improve its ability to preselect brown dwarfs its
imaging surveys.
CONCLUSIONS: The HST filters used in high-redshift searches are close to
optimal to identify broad stellar type. However, the addition of F127M to the
commonly used broad filter sets would allow for unambiguous subtyping. An
improvement over HST is one of two broad and medium filter combinations on
JWST: pairing F140M with either F150W or F162M discriminates very well between
subtypes
Two-Point Functions and S-Parameter in QCD-like Theories
We calculated the vector, axial-vector, scalar and pseudo-scalar two-point
functions up to two-loop level in the low-energy effective field theory for
three different QCD-like theories. In addition we also calculated the
pseudo-scalar decay constant . The QCD-like theories we used are those
with fermions in a complex, real or pseudo-real representation with in general
n flavours. These case correspond to global symmetry breaking pattern of
, or .

What do -ray bursts look like?
There have been great and rapid progresses in the field of -ray
bursts (denoted as GRBs) since BeppoSAX and other telescopes discovered their
afterglows in 1997. Here, we will first give a brief review on the
observational facts of GRBs and direct understanding from these facts, which
lead to the standard fireball model. The dynamical evolution of the fireball is
discussed, especially a generic model is proposed to describe the whole
dynamical evolution of GRB remnant from highly radiative to adiabatic, and from
ultra-relativistic to non-relativistic phase. Then, Various deviations from the
standard model are discussed to give new information about GRBs and their
environment. In order to relax the energy crisis, the beaming effects and their

Long-term recurrence and complication rates after incisional hernia repair with the open onlay technique
<p>Abstract</p> <p>Background</p> <p>Incisional hernia after abdominal surgery is a well-known complication. Controversy still exists with respect to the choice of hernia repair technique. The objective of this study was to evaluate the long-term recurrence rate as well as surgical complications in a consecutive group of patients undergoing open repair using an onlay mesh technique.</p> <p>Methods</p> <p>Consecutive patients undergoing open incisional hernia repair with onlay-technique between 01/05/1995 and 01/09/2007 at a single institution were included in the study. For follow-up patients were contacted by telephone, and answered a questionnaire containing questions related to the primary operation, the hernia and general risk factors. Patients were examined by a consultant surgeon in the outpatient clinic or in the patient's home if there was suspicion of an incisional hernia recurrence.</p> <p>Results</p> <p>The study included 56 patients with 100% follow-up. The median follow-up was 35 months (range 4–151). Recurrent incisional hernia was found in 8 of 56 patients (15%, 95% CI: 6–24). The overall complication rate was 13% (95% CI, 4–22). All complications were minor and needed no hospital admission.</p> <p>Conclusion</p> <p>This study with a long follow-up showed low recurrence and complication rates in patients undergoing incisional hernia repair with the open onlay technique.</p
An instability criterion for nonlinear standing waves on nonzero backgrounds
A nonlinear Schr\"odinger equation with repulsive (defocusing) nonlinearity
is considered. As an example, a system with a spatially varying coefficient of
the nonlinear term is studied. The nonlinearity is chosen to be repelling
except on a finite interval. Localized standing wave solutions on a non-zero
background, e.g., dark solitons trapped by the inhomogeneity, are identified
and studied. A novel instability criterion for such states is established
through a topological argument. This allows instability to be determined
quickly in many cases by considering simple geometric properties of the
standing waves as viewed in the composite phase plane. Numerical calculations

Allergen-specific IgG+ memory B cells are temporally linked to IgE memory responses
BACKGROUND: Immunoglobulin E (IgE) are least abundant, tightly regulated and IgE producing B cells are rare. The cellular origin and evolution of IgE responses are poorly understood. OBJECTIVE: To investigate the cellular and clonal origin of IgE memory responses following mucosal allergen exposure by sublingual immunotherapy (SLIT). METHODS: In a randomized double-blind, placebo-controlled, time-course SLIT study, peripheral blood mononuclear cells (PBMCs) and nasal biopsies were collected from forty adults with seasonal allergic rhinitis at baseline, 4, 8, 16, 28 and 52 weeks. RNA was extracted from PBMCs, sorted B cells and nasal biopsies for VH repertoire sequencing. Moreover, monoclonal antibodies were derived from single B cell transcriptomes. RESULTS: Combining VH repertoire sequencing and single cell transcriptomics yielded direct evidence of a parallel boost of two clonally and functionally related B cell subsets of short-lived IgE+ plasmablasts and IgG+ memory B cells (termed IgGE). Mucosal grass pollen allergen exposure by SLIT resulted in highly diverse IgE and IgGE repertoires. These were extensively mutated and appeared relative stable as per heavy chain isotype, somatic hypermutations and clonal composition. Single IgGE + memory B cell and IgE+ pre-plasmablast transcriptomes encoded antibodies that were specific for major grass pollen allergens and were able to elicit basophil activation at very low allergen concentrations. CONCLUSION: For the first time, we have shown that upon mucosal allergen exposure, human IgE memory resides in allergen-specific IgG+ memory B cells. These rapidly switch isotype and expand into short-lived IgE+ plasmablasts and serve as a potential target for therapeutic intervention
Neoantigen-reactive CD8+ T cells affect clinical outcome of adoptive transfer with tumor-infiltrating lymphocytes in melanoma
BACKGROUND: Neoantigen-driven recognition and T cell-mediated killing contribute to tumor clearance following adoptive cell therapy (ACT) with Tumor-Infiltrating Lymphocytes (TILs). Yet, how diversity, frequency, and persistence of expanded neoepitope-specific CD8+ T cells derived from TIL infusion products affect patient outcome is not fully determined. METHODS: Using barcoded pMHC multimers, we provide a comprehensive mapping of CD8+ T cells recognizing neoepitopes in TIL infusion products and blood samples from 26 metastatic mela-noma patients who received ACT. RESULTS: We identified 106 neoepitopes within TIL infusion products corresponding to 1.8% of all predicted neoepitopes. We observed neoepitope-specific recognition to be virtually devoid in TIL infusion products given to patients with progressive disease outcome. Moreover, we found that the frequency of neoepitope-specific CD8+ T cells in TIL infusion products correlated with in-creased survival, and that detection of engrafted CD8+ T cells in post-treatment (i.e. originating from the TIL infusion product) were unique to responders of TIL-ACT. Finally, we found that a transcriptional signature for lymphocyte activity within the tumor microenvironment was associated with a higher frequency of neoepitope-specific CD8+ T cells in the infusion product. CONCLUSIONS: These data support previous case studies of neoepitope-specific CD8+ T cells in melanoma, and indicate that successful TIL-ACT is associated with an expansion of neoepitope-specific CD8+ T cells.
