    In Vitro Toxicity of Asparagus Saponins in Distinct Multidrug-Resistant Colon Cancer Cells

    Colorectal cancer is the third most common cancer in the world. Many efforts have focused on finding natural molecules with potential chemo-preventive activity due to their low toxicity compared to synthetic drugs. However, comprehensive information on the bioactive fractions and components is still missing. In this study, we developed a method for the quantitative separation and isolation of saponins from asparagus genotypes consisting of an adsorption chromatography and subsequent liquid chromatographic separation on a reversed-phase column. The saponins isolated were tested for their cytotoxic activity against human colon cancer cell lines, which could develop cross-resistance to a wide variety of chemotherapeutic drugs. Our results showed that Huétor-Tájar asparagus saponins (HTSAP), mainly protodioscin and HTSAP-10 have higher cytotoxic activity than HTSAP-1, HTSAP-6, and HTSAP-8.     Quality Characteristics and Antioxidant Properties of Muffins Enriched with Date Fruit (Phoenix Dactylifera L.) Fiber Concentrates

Fiber concentrates (DFC) can be obtained from these fruits after a steam pretreatment. DFCs were evaluated as a source of antioxidant dietary fiber for bakery products. Muffins were prepared with 2.5 and 5% flour substitution with DFCs obtained by treatments at 165 and 180C. The DFC-doughs presented a similar yield to the control but the muffins reached a lower volume. The density increase did not imply an increase in texture. In fact, the muffins with DFC-165 were the softest tested, although they had lower cohesiveness and springiness. The proximate composition was similar among samples. The DFC-muffins had higher antioxidant capacity than the control, and obtained good scores in the sensory evaluation. DFC-165 is a valuable ingredient for baked goods, but its effect on fat rancidity and staling delays should be confirmed.This research was supported by the Banq Islamique de Développement BID (Saudi Arabia REF. 36/11201707).     Saponins from edible spears of wild asparagus inhibit AKT, p70S6K, and ERK signalling, and induce apoptosis through G0/G1 cell cycle arrest in human colon cancer HCT-116 cells

    41 Páginas, 6 Figuras, 1 TablaThe effects of steroidal saponins from edible spears of wild triguero Huetor-Tajar asparagus on some of the oncogenic molecular pathways that are affected in human colon cancer cells were investigated. Reverse-phase chromatography and a new HPLC-MS method were used to respectively isolate and analyse the composition of the steroidal saponins. They were resistant to simulated digestion and, when in contact with HTC-116 human colon carcinoma cells, interfered with extracellular signal-regulated kinase (ERK), S6 kinase (p70S6K, mTOR), and RAC-alpha serine/threonine-protein kinase (AKT) pathways by a downregulation of these proteins. The expressions of cyclins D, E, and A were also decreased, leading to G0/G1 cell cycle arrest. In addition, these steroidal saponins induced typical features of apoptosis by the promotion of caspase-3 activity, poly(ADP-ribose) polymerase 1 (PARP-1) cleavage, and DNA fragmentation.     Autologous Platelet-Rich Plasma in the Treatment of Perianal Fistula in Crohn’s Disease

    [Aim] To assess clinical healing in patients with perianal Crohn’s disease with local intrafistular injection of autologous platelet-rich plasma.[Method] The pilot study was conducted at a single centre between January 2013 and December 2015. Autologous platelet-rich plasma was prepared in platelet-rich and platelet-poor fractions for local intrafistular injection in patients with proven, established perianal Crohn’s disease. Patients were permitted biological therapies, and the Perianal Crohn’s Disease Activity Index was recorded. Patients were followed for 48 weeks for clinical signs of healing (complete, partial or non-healing), monitoring fistula drainage, closure and epithelialization.[Results] The study included 29 patients (19 males; mean age 38 ± 12.8 years) with four exclusions in the operating room because surgery was not indicated and four lost to follow-up. Five adverse events were recorded, with two requiring the drainage of abscess collections. Of the 21 patients assessable at 24 weeks, there was complete healing, partial healing and non-healing in 7 (33.3%), 8 (38.1%) and 6 (28.6%) patients, respectively. By 48 weeks, there was complete healing, partial healing and non-healing in 6 (40%), 6 (40%) and 3 (20%) patients, respectively, with a reduction in the number of visible external fistula openings at both time points (P = 0.021). By the end of the study, there was a higher trend of healing if biological therapies were continued (85.7% with biologics vs. 75% without, P = 0.527), but there were no statistically significant differences and no differences in the Perianal Crohn’s Disease Activity Index.[Conclusion]Autologous platelet-rich plasma is safe in patients with perianal Crohn’s disease, with an acceptable healing rate over a medium-term follow-up, particularly if biological therapies are used concomitantly

    Date Seed: Rich Source of Antioxidant Phenolics Obtained by Hydrothermal Treatments

    peer reviewedThe growing interest in natural compounds is helping to improve the management of agro-industrial by-products such as the date seed as sources of such compounds. In this work, the application of a hydrothermal treatment at 160 and 180 °C for 60 min was studied to achieve the solubilization of its phenolic components and sugars in order to obtain biologically active extracts. The percentage of phenols and total sugars in the final extracts were very similar, at 45 and 25% for the 160 and 180 °C treatments, respectively. The treatment at a higher temperature allowed greater solubilization of other components. The antioxidant activity was measured as free-radical scavenging capacity. For the DPPH• method, expressed as EC50, the results were 0.34 and 0.37 mg/L, the TEAC values for the ABTS• method were 6.61 and 3.28 mg/g dried extract, and the values obtained by the ORAC method were 12.82 and 9.91 mmol Trolox/g dried extract, for 160 and 180 °C, respectively. All these values are higher than those of other plant extracts and extracts obtained using the whole date.     Recombinant human leptin treatment in genetic lipodystrophic syndromes: the long-term Spanish experience

    Lipodystrophies are a group of diseases mainly characterized by a loss of adipose tissue and frequently associated with insulin resistance, hypertriglyceridemia, and hepatic steatosis. In uncommon lipodystrophies, these complications frequently are difficult to control with conventional therapeutic approaches. This retrospective study addressed the effectiveness of recombinant methionyl leptin (metreleptin) for improving glucose metabolism, lipid profile, and hepatic steatosis in patients with genetic lipodystrophic syndromes. We studied nine patients (five females and four males) with genetic lipodystrophies [seven with Berardinelli-Seip syndrome, one with atypical progeroid syndrome, and one with type 2 familial partial lipodystrophy (FPLD)]. Six patients were children under age 9 years, and all patients had baseline triglycerides levels >2.26 mmol/L and hepatic steatosis; six had poorly controlled diabetes mellitus. Metreleptin was self-administered subcutaneously daily at a final dose that ranged between 0.05 and 0.24 mg/(kg day) [median: 0.08 mg/(kg day)] according to the body weight. The duration of treatment ranged from 9 months to 5 years, 9 months (median: 3 years). Plasma glucose, hemoglobin A1c (Hb A1c), lipid profile, plasma insulin and leptin, and hepatic enzymes were evaluated at baseline and at least every 6 months. Except for the patient with FPLD, metreleptin replacement significantly improved metabolic control (Hb A1c: from 10.4 to 7.1 %, p < 0.05). Plasma triglycerides were reduced 76 % on average, and hepatic enzymes decreased more than 65 %. This study extends knowledge about metreleptin replacement in genetic lipodystrophies, bearing out its effectiveness for long periods of time

    Los socios de la RSEHN y el desarrollo de las colecciones científicas del MNCN

    Valencia, del 8 al 11 de septiembre de 2021. El tema principal tuvo como lema: “La huella Humana en la Naturaleza”.Las colecciones científicas son una infraestructura de investigación única e irremplazable para numerosas áreas de la ciencia. En la actualidad, se estima que en el Museo Nacional de Ciencias Naturales se conservan 10 millones de especímenes, lo que suponen casi la mitad de todos los conservados en España. Esta infraestructura científica o este tesoro, como puede ser llamado, se ha reunido principalmente a lo largo del último siglo, y se debe al trabajo conjunto de muchos especialistas, estudiosos e interesados en diferentes disciplinas de las ciencias naturales. En esta ponencia se quiere poner en valor la aportación de los socios de la RSEHN en el incremento y desarrollo de las colecciones científicas del MNCN. Se ha realizado un análisis preliminar de los fondos de las diferentes colecciones del MNCN y de los ingresos efectuados por los socios de RSEHN desde su fundación (1871) hasta el momento en que abandona el MNCN (1971). Se realizó en cada una de ellas una consulta de los diferentes colectores y personas que aportaron especímenes y que aparecen en las bases de datos, cruzándola con la base de datos de socios durante dicho periodo. Se ha contabilizado el número de especímenes, número de tipos y taxones correspondientes a éstos. La cifra de socios total supera el centenar. El número de ejemplares ingresados por estos socios, entre esas fechas, se estima que superan el millón y medio, lo que supone al menos un 15% del total actual estimado. Hay que tener en cuenta que las colecciones no se encuentran informatizadas al 100%
