    Building Community at the Library With Coffee and Conversation

    As communities and individuals struggle with houselessness and housing insecurity, library staff must adapt in order to help our patrons fulfill needs. In 2013, Multnomah County Library (MCL) and the Multnomah County Department of County Human Services surveyed library patrons about library use. The survey identified that about 18.6 percent of visitors to Central Library—the county system’s main branch, located in downtown Portland— are people experiencing houselessness. These patrons visit the library more frequently than patrons who are not experiencing houselessness, and they tend to stay at the library longer than the latter group. Because patrons experiencing houselessness may have negative experiences interacting with staff in businesses, government offices, and other public institutions like libraries, it’s important to start any engagement by building relationships. Staff members need to help these patrons find resources and library materials, assist them with account issues, and show them that the library is the right place for them by making them feel welcome. In order to address this need, staff looked for a way to build relationships between staff and patrons and to create a welcoming environment in the library

    Sequenced copolymers with controlled molecular weights prepared via entropy-driven ring-opening metathesis polymerization

    A segmer-based approach towards monomer synthesis was successfully carried out to produce a series of linear α,ω-diolefinic species with varying linker lengths. Ring-closing metathesis (RCM) using a cis-selective ruthenium catalyst yielded a large strainless macrocycle that then underwent entropy-driven ring-opening metathesis polymerization (ED-ROMP). When compared to the same polymerization carried out with the complementary trans-monomer, the ED-ROMP of cis-monomer demonstrated a significantly higher degree of molecular weight control. The narrow dispersity exhibited by this process suggests that a new regime of selectivity enhanced ED-ROMP (SEED-ROMP) has been achieved. Reaction kinetics for this process were probed and the process was compared to alternative polymerization methods. Finally, the high degree of control was exemplified in a series of chain extension and block copolymerization experiments. Ultimately, SEED-ROMP represents a novel method to obtain well-defined copolymers of precise molecular weights

    Structure-Activity Relationships Analysis of (+)-Discodermolide and the Synthesis of Precursors for Further Analogue Studies

    In 1990, the natural product discodermolide was isolated from a marine sponge and later found to have exceptional antiproliferative activity in cancer cells. Further studies showed discodermolide to stabilize microtubules via the taxoid binding site on beta-tubulin. Unlike paclitaxel, it is not a substrate for P-glycoprotein transport, is potent in paclitaxel-resistant cell lines with beta-tubulin mutations, exhibits better water solubility, and can act synergistically when combined with paclitaxel treatment. For this reason, discodermolide has been the focus of many synthetic and biological studies, culminating in a 60 gram-scale synthesis and Phase I/II clinical trials carried out by Novartis in 2004. Despite the great level of interest from the scientific community, much about this compound’s behavior in vivo is still unknown. An extended analysis of known structure-activity relationships for discodermolide is presented herein. Also reported are the syntheses of a series of discodermolide fragments, which are designed to enable the formation of a novel analogue library via late-stage multicomponent reactions. Fragments to be synthesized prior to coupling are streamlined alternatives to the discodermolide framework that have shown promise in prior analogue and HQSAR studies. Overall, these modifications have been designed to improve the efficiency of synthetic efforts and to represent a wholly unique series of analogues for biological study

    A systematic review of the feasibility, acceptability, and efficacy of online supportive care interventions targeting men with a history of prostate cancer

    © 2019, The Author(s). Purpose: To examine the feasibility, acceptability, and efficacy of online supportive care interventions targeting prostate cancer survivors (PCS). Methods: Studies were identified through structured searches of PubMed, Embase and PsycINFO databases, and bibliographic review. Inclusion criteria were (1) examined feasibility, acceptability, or efficacy of an online intervention designed to improve supportive care outcomes for PCS; (2) presented outcome data collected from PCS separately (if mixed cancer); and (3) evaluated efficacy outcomes using randomized controlled trial (RCT) design. Results: Sixteen studies met inclusion criteria; ten were classified as RCTs. Overall, 2446 men (average age 64years) were included. Studies reported on the following outcomes: feasibility and acceptability of an online intervention (e.g., patient support, online medical record/follow-ups, or decision aids); reducing decisional conflict/distress; improving cancer-related distress and health-related quality of life; and satisfaction with cancer care. Conclusion: We found good preliminary evidence for online supportive care among PCS, but little high level evidence. Generally, the samples were small and unrepresentative. Further, inadequate acceptability measures made it difficult to determine actual PCS acceptability and satisfaction, and lack of control groups precluded strong conclusions regarding efficacy. Translation also appears minimal; few interventions are still publicly available. Larger trials with appropriate control groups and greater emphasis on translation of effective interventions is recommended. Implications for Cancer Survivors: Prostate cancer survivors have a variety of unmet supportive care needs. Using online delivery to improve the reach of high-quality supportive care programs could have a positive impact on health-related quality of life among PCS

    Impoverished encoding of speaker identity in spontaneous laughter

    Our ability to perceive person identity from other human voices has been described as prodigious. However, emerging evidence points to limitations in this skill. In this study, we investigated the recent and striking finding that identity perception from spontaneous laughter - a frequently occurring and important social signal in human vocal communication - is significantly impaired relative to identity perception from volitional (acted) laughter. We report the findings of an experiment in which listeners made speaker discrimination judgements from pairs of volitional and spontaneous laughter samples. The experimental design employed a range of different conditions, designed to disentangle the effects of laughter production mode versus perceptual features on the extraction of speaker identity. We find that the major driving factor of reduced accuracy for spontaneous laughter is not its perceived emotional quality but rather its distinct production mode, which is phylogenetically homologous with other primates. These results suggest that identity-related information is less successfully encoded in spontaneously produced (laughter) vocalisations. We therefore propose that claims for a limitless human capacity to process identity-related information from voices may be linked to the evolution of volitional vocal control and the emergence of articulate speech

    Preferential, enhanced breast cancer cell migration on biomimetic electrospun nanofiber ‘cell highways’

    BACKGROUND: Aggressive metastatic breast cancer cells seemingly evade surgical resection and current therapies, leading to colonization in distant organs and tissues and poor patient prognosis. Therefore, high-throughput in vitro tools allowing rapid, accurate, and novel anti-metastatic drug screening are grossly overdue. Conversely, aligned nanofiber constitutes a prominent component of the late-stage breast tumor margin extracellular matrix. This parallel suggests that the use of a synthetic ECM in the form of a nanoscale model could provide a convenient means of testing the migration potentials of cancer cells to achieve a long-term goal of providing clinicians an in vitro platform technology to test the efficacy of novel experimental anti-metastatic compounds. METHODS: Electrospinning produces highly aligned, cell-adhesive nanofiber matrices by applying a strong electric field to a polymer-containing solution. The resulting fibrous microstructure and morphology closely resembles in vivo tumor microenvironments suggesting their use in analysis of migratory potentials of metastatic cancer cells. Additionally, a novel interface with a gel-based delivery system creates CXCL12 chemotactic gradients to enhance CXCR4-expressing cell migration. RESULTS: Cellular dispersions of MCF-10A normal mammary epithelial cells or human breast cancer cells (MCF-7 and MDA-MB-231) seeded on randomly-oriented nanofiber exhibited no significant differences in total or net distance traveled as a result of the underlying topography. Cells traveled ~2-5 fold greater distances on aligned fiber. Highly-sensitive MDA-MB-231 cells displayed an 82% increase in net distance traversed in the presence of a CXCL12 gradient. In contrast, MCF-7 cells exhibited only 31% increase and MCF-10A cells showed no statistical difference versus control or vehicle conditions. MCF-10A cells displayed little sensitivity to CXCL12 gradients, while MCF-7 cells displayed early sensitivity when CXCL12 concentrations were higher. MDA-MB-231 cells displayed low relative expression levels of CXCR4, but high sensitivity resulting in 55-fold increase at late time points due to CXCL12 gradient dissipation. CONCLUSIONS: This model could create clinical impact as an in vitro diagnostic tool for rapid assessment of tumor needle biopsies to confirm metastatic tumors, their invasiveness, and allow high-throughput drug screening providing rapid development of personalized therapies.
