    As host of the Olympic Games, China seeks to increase national economic and socialdevelopment and "display to the world a new image of China", and presents the Games as an opportunity to foster democracy, improve human rights and integrate China with the rest of the world. In its Olympic Action Plan promulgated in 2002, China outlined the phases of construction in the run up to the 2008 Games, and the standards to which it would hold itself in the governance and construction of venues, impact on Beijing's environment, increasing social and economic development and providing China's citizenry with greater access to information and technology.The goals and specific commitments that the government has adopted not only have implications for the smooth and successfuloperation of the Olympic Games, but also have the potential to impact on a number of China's international obligations, including its human rights obligations.Despite human rights-related commitments as diverse as transparency and accountability, access to information and freedom of the press, poverty alleviation, an improved standard of living for all people, and compensation for evictions and health issues, the record to date raises serious compliance issues

    Use It and Lose It? Behavioral and Energetic Influences on Lizard Tail Autotomy

    In many lizard species, the ability to autotomize (lose) the tail allows for immediate survival in the face of predation. However, tails are often used in communication and energy storage, and so tail autotomy can thus decrease an individual’s overall fitness. In this study, I examine this tradeoff in species that use the tail differently. I predict less frequent tail autotomy in species that use the tail for social display or energy storage, as a full tail is particularly valuable. In species that primarily display the tail in an anti-predatory context, I predict the frequency of tail loss will be higher, as the tail is made especially vulnerable to autotomization. I studied seven lizard species that vary in tail use: greater earless lizards (Cophosaurus texanus) and curly tail lizards (Leiocephalus carinatus) use tails frequently in display; Mediterranean house geckos (Hemidactylus frenatus) and crested anoles (Anolis cristatellus) use tails occasionally; and green anoles (Anolis carolinensus), Texas spiny lizards (Sceloporus olivaceus), and Texas spotted whiptails (Aspidoscelis gularis) almost never include tail movements in display. I observed lizards of each species in the field to quantify the use of the tail in social contexts, and performed predator simulation trials to quantify tail use in anti-predatory contexts. I calculated the energetic content of the tail using bomb calorimetry. I found that the frequency of tail autotomy varies from 20-60 % of individuals across the seven species, and lizards that use their tail in a social context also tend to do so in an anti-predatory context. Finally, I found that total energy content in the tail is positively associated with tail loss frequency across species. By studying lizard tail loss, we can better understand the evolutionary trade-offs involved in balancing the benefit of predator evasion with the cost of tail loss in social and energetic utility

    Office bladder distention with Electromotive Drug Administration (EMDA) is equivalent to distention under General Anesthesia (GA)

    BACKGROUND: Bladder distention is commonly used in diagnosis and treatment of interstitial cystitis (IC). Traditionally performed in the operating room under general or spinal anesthesia (GA), it is expensive and associated with short term morbidity. Office bladder distention using electromotive drug administration (EMDA) has been suggested as an alternative that is well tolerated by patients. We report the first comparative findings of patients undergoing both office distention with EMDA and distention in the operating room (OR) with GA. METHODS: This retrospective chart review identified 11 patients participating in two protocols of EMDA bladder distention who also underwent bladder distention under GA either prior to or after the EMDA procedure. RESULTS: The median absolute difference in bladder capacity between GA and EMDA was only 25 cc; the median percent difference was 5%. Cystoscopic findings, while not prospectively compiled, appear to have been similar. CONCLUSION: This study represents the first comparison between distention with EMDA versus GA and confirms the technical feasibility of performing bladder distention in an office setting. The distention capacity achieved in the office was nearly identical to that in the OR and the cystoscopic findings very similar. Further investigation into the comparative morbidity, cost, and other outcome measures is warranted to define the ultimate role of EMDA bladder distention in the clinical evaluation and care of patients with IC

    Skeletal muscle mitochondrial dysfunction in contemporary antiretroviral therapy: a single cell analysis

    OBJECTIVE: To quantify mitochondrial function in skeletal muscle of people treated with contemporary antiretroviral therapy. DESIGN: Cross-sectional observational study. METHODS: Quantitative multiplex immunofluorescence was performed to determine mitochondrial mass and respiratory chain complex abundance in individual myofibres from tibialis anterior biopsies. Individual myofibres were captured by laser microdissection and mitochondrial DNA (mtDNA) content and large-scale deletions were measured by real-time PCR. RESULTS: Forty five antiretroviral therapy (ART) treated people with HIV (PWH, mean age 58 years, mean duration of ART 125 months) were compared with 15 HIV negative age-matched controls. Mitochondrial complex I (CI) deficiency was observed at higher proportional levels in PWH than negative controls (P = 0.008). Myofibre mitochondrial mass did not differ by HIV status.No ART class was significantly associated with mitochondrial deficiency, including prior exposure to historical NRTIs (nucleoside analogue reverse transcriptase inhibitors) associated with systemic mitochondrial toxicity.To exclude an effect of untreated HIV, we also studied skeletal muscle from 13 ART-naïve PWH (mean age 37). These showed negligible CI defects, as well as comparable myofibre mitochondrial mass to ART-treated PWH.Most CI-deficient myofibres contained mtDNA deletions. No mtDNA depletion was detected. CONCLUSION: Here, we show that PWH treated with contemporary ART have mitochondrial dysfunction in skeletal muscle, exceeding that expected due to age alone. Surprisingly, this was not mediated by prior exposure to mitochondrially toxic NRTIs, suggesting novel mechanisms of mitochondrial dysfunction in contemporary ART-treated PWH. These findings are relevant for better understanding successful ageing in PWH

    Assessing the Effect of High Performance Computing Capabilities on Academic Research Output

    This paper uses nonparametric methods and some new results on hypothesis testing with nonparametric efficiency estimators and applies these to analyze the effect of locally-available high performance computing (HPC) resources on universities efficiency in producing research and other outputs. We find that locally-available HPC resources enhance the technical efficiency of research output in Chemistry, Civil Engineering, Physics, and History, but not in Computer Science, Economics, nor English; we find mixed results for Biology. Out research results provide a critical first step in a quantitative economic model for investments in HPC

    Intact implicit processing of facial threat cues in schizophrenia

    An emerging body of research suggests that people with schizophrenia retain the ability to implicitly perceive facial affect, despite well-documented difficulty explicitly identifying emotional expressions. It remains unclear, however, whether such functional implicit processing extends beyond emotion to other socially relevant facial cues. Here, we constructed two novel versions of the Affect Misattribution Procedure, a paradigm in which affective responses to primes are projected onto neutral targets. The first version included three face primes previously validated to elicit varying inferences of threat from healthy individuals via emotion-independent structural modification (e.g., nose and eye size). The second version included the threat-relevant emotional primes of angry, neutral, and happy faces. Data from 126 participants with schizophrenia and 84 healthy controls revealed that although performing more poorly on an assessment of explicit emotion recognition, patients showed normative implicit threat processing for both non-emotional and emotional facial cues. Collectively, these results support recent hypotheses postulating that the initial perception of salient facial information remains intact in schizophrenia, but that deficits arise at subsequent stages of contextual integration and appraisal. Such a breakdown in the stream of face processing has important implications for mechanistic models of social cognitive impairment in schizophrenia and treatment strategies aiming to improve functional outcome

    Deficiency of plasminogen activator inhibitor‐2 results in accelerated tumor growth

    BackgroundUpregulation of the plasminogen activation system, including urokinase plasminogen activator (uPA), has been observed in many malignancies, suggesting that co‐opting the PA system is a common method by which tumor cells accomplish extracellular matrix proteolysis. PAI‐2, a serine protease inhibitor, produced from the SERPINB2 gene, inhibits circulating and extracellular matrix‐tethered uPA. Decreased SERPINB2 expression has been associated with increased tumor invasiveness and metastasis for several types of cancer. PAI‐2 deficiency has not been reported in humans and PAI‐2‐deficient (SerpinB2−/−) mice exhibit no apparent abnormalities.ObjectivesWe investigated the role of PAI‐2 deficiency on tumor growth and metastasis.MethodsTo explore the long‐term impact of PAI‐2 deficiency, a cohort of SerpinB2−/− mice were aged to >18 months, with spontaneous malignancies observed in 4/9 animals, all of apparently vascular origin. To further investigate the role of PAI‐2 deficiency in malignancy, SerpinB2−/− and wild‐type control mice were injected with either B16 melanoma or Lewis lung carcinoma tumor cells, with markedly accelerated tumor growth observed in SerpinB2−/− mice for both cell lines. To determine the relative contributions of PAI‐2 from hematopoietic or nonhematopoietically derived sources, bone marrow transplants between wild‐type C57BL/6J and SerpinB2−/− mice were performed.Results and ConclusionsOur results suggest that PAI‐2 deficiency increases susceptibility to spontaneous tumorigenesis in the mouse, and demonstrate that SerpinB2 expression derived from a nonhematopoietic compartment is a key host factor in the regulation of tumor growth in both the B16 melanoma and Lewis lung carcinoma models.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/163438/2/jth15054_am.pdfhttp://deepblue.lib.umich.edu/bitstream/2027.42/163438/1/jth15054.pd

    Analysis of Iophenoxic Acid Analogues in Small Indian Mongoose (\u3ci\u3eHerpestes Auropunctatus\u3c/i\u3e) Sera for Use as an Oral Rabies Vaccination Biological Marker

    The small Indian mongoose (Herpestes auropunctatus) is a reservoir of rabies virus (RABV) in Puerto Rico and comprises over 70% of animal rabies cases reported annually. The control of RABV circulation in wildlife reservoirs is typically accomplished by a strategy of oral rabies vaccination (ORV). Currently no wildlife ORV program exists in Puerto Rico. Research into oral rabies vaccines and various bait types for mongooses has been conducted with promising results. Monitoring the success of ORV relies on estimating bait uptake by target species, which typically involves evaluating a change in RABV neutralizing antibodies (RVNA) post vaccination. This strategy may be difficult to interpret in areas with an active wildlife ORV program or in areas where RABV is enzootic and background levels of RVNA are present in reservoir species. In such situations, a biomarker incorporated with the vaccine or the bait matrix may be useful. We offered 16 captive mongooses placebo ORV baits containing ethyl-iophenoxic acid (et-IPA) in concentrations of 0.4% and 1% inside the bait and 0.14% in the external bait matrix. We also offered 12 captive mongooses ORV baits containing methyl-iophenoxic acid (me-IPA) in concentrations of 0.035%, 0.07% and 0.14% in the external bait matrix. We collected a serum sample prior to bait offering and then weekly for up to eight weeks post offering. We extracted Iophenoxic acids from sera into acetonitrile and quantified using liquid chromatography/mass spectrometry. We analyzed sera for et-IPA or me-IPA by liquid chromatography-mass spectrometry. We found adequate marking ability for at least eight and four weeks for et- and me-IPA, respectively. Both IPA derivatives could be suitable for field evaluation of ORV bait uptake in mongooses. Due to the longevity of the marker in mongoose sera, care must be taken to not confound results by using the same IPA derivative during consecutive evaluations
