    Isolation and Identification of Bacteria-producing Cellulose from Tropical Fruit for Halal Capsule Application

    Bacterial cellulose (BC) is pure cellulose synthesized by various species of bacteria. Raising demands on bacterial cellulose is due to its pure and simpler structure. It has plenty of applications in various industries such as food, medical and cosmetics, rendering it a choice in halal industry application as a substitute for non-halal gelatine. However, challenges arise during the BC production such as high production cost and low volumetric yield. In this study, BC is studied to overcome the barriers to BC production. Isolation and identification of cellulose producing bacteria were carried out on eight different tropical fruit sources using Hestrin-Schramm media in static culture condition. Morphological and molecular identification by microscopic observation, gram staining, and 16S rRNA analysis were conducted to identify the characteristics and strain of the new isolates. Next, the selected colonies were challenged to grow in agitating condition using modified HS media. The effects of carbon concentration and agitation speed on the production of cellulose were investigated using on central composite design (CCD). Three new cellulose producing bacteria were successfully isolated and identified to be similar to Enterobacter sp. SJZ-5, Bacterium sp NLAE-zl-H356, and Bulkhoderia sp. RD_DACAR_02 through morphological and molecular analysis. The most potent strain which is similar to Enterobacter sp. SJZ-5 (named as Enterobacter sp.M003) has been chosen for BC optimization study for high BC production using modified HS media. Optimization of bacterial cellulose production using response surface methodology (RSM) with 13 runs indicated that the optimal production parameters were 17.5 g/L for carbon concentration at 277 rpm for agitation speed gave 1.7g/L cellulose. It is expected that the newly isolated bacteria will be able to provide an alternative to gelatine for halal capsule production, thus minimizing and replacing non-halal gelatine usage

Metode yang digunakan sebagai berikut : (1) Sosialisasi pembuatan sumur resapan biopori, (2) Pelatihan dan pendampingan pembuatan sumur resapan biopori, (3) Pemanenan dan pemanfaatan kompos hasil sumur resapan biopori. Setelah mengikuti pelatihan dan pendampingan dari Tim Pengabdian Masyarakat Polines, warga masyarakat di RT 04/ RW.02 Kel. Banyumanik Semarang dapat mencapai target sebagai berikut :Masyarakat mampu membuat sumur resapan biopori secara benar untuk mencegah banjir dan sekaligus memanfaatkan kompos dari hasil pemanenan biopori.     The purpose of this activity is to empower the community through making biopore infiltration wells as an effort to help prevent flooding and at the same time produce compost to improve the community's economy. The method used is as follows : (1) Socialization of making biopore infiltration wells, (2) Training and assistance in making biopore infiltration wells, (3) Harvesting and utilizing compost from biopore infiltration wells. After attending training and mentoring from the Polines Community Service Team, community members in RT 01/ RW.02 Kel. Tembalang Semarang can achieve the following targets: The community is able to properly construct biopore infiltration wells to prevent flooding and at the same time utilize compost from the harvesting of biopori. The outputs resulting from the assistance of this community service activity are: (1) The realization of biopore infiltration wells and (2) utilization of biopore compost to improve the community's economy.Keywords: Biopori Infiltration Hole, compost, preventing flooding

    Open tension free repair of inguinal hernias; the Lichtenstein technique

    BACKGROUND: Recurrences have been a significant problem following hernia repair. Prosthetic materials have been increasingly used in hernia repair to prevent recurrences. Their use has been associated with several advantages, such as less postoperative pain, rapid recovery, low recurrence rates. METHODS: In this retrospective study, 540 tension-free inguinal hernia repairs were performed between August 1994 and December 1999 in 510 patients, using a polypropylene mesh (Lichtenstein technique). The main outcome measure was early and late morbidity and especially recurrence. RESULTS: Inguinal hernia was indirect in 55 % of cases (297 patients), direct in 30 % (162 patients) and of the pantaloon (mixed) type in 15 % (81 patients). Mean patient age was 53.7 years (range, 18 – 85). Follow-up was completed in 407 patients (80 %) by clinical examination or phone call. The median follow-up period was 3.8 years (range, 1 – 6 years). Seroma and hematoma formation requiring drainage was observed in 6 and 2 patients, respectively, while transient testicular swelling occurred in 5 patients. We have not observed acute infection or abscess formation related to the presence of the foreign body (mesh). In two patients, however, a delayed rejection of the mesh occurred 10 months and 4 years following surgery. There was one recurrence of the hernia (in one of these patients with late mesh rejection) (recurrence rate = 0.2 %). Postoperative neuralgia was observed in 5 patients (1 %). CONCLUSION: Lichtenstein tension-free mesh inguinal hernia repair is a simple, safe, comfortable, effective method, with extremely low early and late morbidity and remarkably low recurrence rate and therefore it is our preferred method for hernia repair since 1994

    Comparison of sequencing methods and data processing pipelines for whole genome sequencing and minority single nucleotide variant (mSNV) analysis during an influenza A/H5N8 outbreak

    As high-throughput sequencing technologies are becoming more widely adopted for analysing pathogens in disease outbreaks there needs to be assurance that the different sequencing technologies and approaches to data analysis will yield reliable and comparable results. Conversely, understanding where agreement cannot be achieved provides insight into the limitations of these approaches and also allows efforts to be focused on areas of the process that need improvement. This manuscript describes the next-generation sequencing of three closely related viruses, each analysed using different sequencing strategies, sequencing instruments and data processing pipelines. In order to determine the comparability of consensus sequences and minority (sub-consensus) single nucleotide variant (mSNV) identification, the biological samples, the sequence data from 3 sequencing platforms and the *.bam quality-trimmed alignment files of raw data of 3 influenza A/H5N8 viruses were shared. This analysis demonstrated that variation in the final result could be attributed to all stages in the process, but the most critical were the well-known homopolymer errors introduced by 454 sequencing, and the alignment processes in the different data processing pipelines which affected the consistency of mSNV detection. However, homopolymer errors aside, there was generally a good agreement between consensus sequences that were obtained for all combinations of sequencing platforms and data processing pipelines. Nevertheless, minority variant analysis will need a different level of careful standardization and awareness about the possible limitations, as shown in this study

    Ten-year audit of Lichtenstein hernioplasty under local anaesthesia performed by surgical residents

    <p>Abstract</p> <p>Background</p> <p>To analyse in a prospective trial the long-term results of Lichtenstein hernioplasty performed by surgical trainees.</p> <p>Methods</p> <p>Training of tension-free Lichtenstein hernia operation was started in our ambulatory unit as an outpatient procedure under local anaesthesia in 1996. After performing 36 teaching operations together with residents and their supervising specialist, 281 patients were operated during 1996-2000 either by one senior consultant (n = 141) or by 12 surgical trainees (n = 140). After 10 years, 247 (88%) patients were available for the long-term assessment.</p> <p>Results</p> <p>After one month postoperatively, the rate of wound infections (consultant 1.1%, residents 0.7%) and hematomas (consultant 1.1%, residents 3.0%) were low and not related to surgeon's training level (ns). Only 6 (2.1%) clinically evident recurrences were found after 10 years: two after specialist repair and four after trainee repair (ns). Although one third of the patients reported some discomfort after 3 and 10 years, 93-95% of the patients were very satisfied with the operation, with no statistical difference between the surgeons.</p> <p>Conclusion</p> <p>Ambulatory open mesh repair under local anaesthesia was a safe operation and the long-term results were acceptable among the patients operated by surgical trainees.</p

    A novel and well-defined benchmarking method for second generation read mapping

    Background Second generation sequencing technologies yield DNA sequence data at ultra high-throughput. Common to most biological applications is a mapping of the reads to an almost identical or highly similar reference genome. The assessment of the quality of read mapping results is not straightforward and has not been formalized so far. Hence, it has not been easy to compare different read mapping approaches in a unified way and to determine which program is the best for what task. Results We present a new benchmark method, called Rabema (Read Alignment BEnchMArk), for read mappers. It consists of a strict definition of the read mapping problem and of tools to evaluate the result of arbitrary read mappers supporting the SAM output format. Conclusions We show the usefulness of the benchmark program by performing a comparison of popular read mappers.
