Tuturan Imperatif Mamah Dedeh Pada Acara Mamah Dan Aa Beraksi Di Stasiun Televisi Indosiar
This study discusses the imamative imperative of Mamah Dedeh on Mamah dan Aa Beraksi on television stations Indosiar. The objectives of study are to describe the form and meaning of imperative speech of Mamah Dedeh on Mamah dan Aa Beraksi on television stations Indosiar. The method used is descriptive method with qualitative approach. There are four techniques used for data collection, i.e. observation techniques, listening techniques, recording techniques, and note-taking techniques. The source of data is the whole speech of Mamah Dedeh on Mamah Dedeh and Aa Beraksi on television stations Indosiar. There are 98 data which consist of 49 imperative form, and 49 data imperative meaning. The result of this study shows that there are five form of imamative imperative that are : imperative orders, solicitation, prohibition, permition, and command. Moreover this study found that there are mine imperative meanings that are : prohibition, command, orders, request, appealing, solicition, expectation, congratulation, and recommandation
THE DOSE VARIATIONS EFFECT OF Centella asiatica ETHANOL EXTRACT ON ESCAPE LATENCY’S DISTANCE MORRIS WATER MAZE AFTER CHRONIC ELECTRICAL STRESS
Centella asiatica sp. (pegagan) has neurotropic and neuroprotective properties which inhibit deterioration of memory because of chronic electrical stress. Studies about effective dose of Centtela asiatica still continued. This study was conducted to discover the influence of pegagan extract’s dose variation intake towards memory after chronic stress. This study uses that true experimental design with pretest-posttest control design. Fifteen male Sprague Dawley rats were divided into three groups randomly. Every groups are treated with chronic electrical stress for 4 weeks and aquades (KN), 300 (KN1) or 600 mg/kgBW/day (KP2) of Centella asiatica. The escape latency distance measure using Morris water maze was used to assess rats’ memory. Data were analyzed using the Kolmogorov-Smirnov, ANOVA and paired t-test. The average of escape latency distance before chronic electrical stress were 5,71±0,77m (KN); 4,29±0,36m (KP1); 3,81±0,47m (KP2) and after chronic electrical stress were 1,50±0,05m(KN); 1,55±0,04m(KP1); 1,82±0,11m(KP2). There were significant differences among groups after chronic electrical stress on day-1 (p=0,005) and day-3 (p=0,001). There were significant differences (p<0,05) for 3 days of escape latency test (KN), 5 days of escape latency test (KP1) and 2 days of escape latency test (KP2) from 6 days of memory measurement. In conclusion, dose variation of Centella asiatica did not affect to latency distance using Morris water maze. Ethanol extract of Centella asiatica with doses 300mg/kgBW/day could stimulate increasing memory formation after chronic electrical stress better.Keywords: Centella asiatica - chronic electrical stress– memory– Morris water maze
Expressed breast milk feeding: knowledge and attitude of employed mothers
Appropriate knowledge on expressing, storing and use of breast milk are essential for mothers to continue breastfeeding when they return to work. The objective of this study was to assess the knowledge and attitude of employed mothers towards breast milk expression, storage and usage. This cross-sectional study was conducted among 300 full-time employed mothers who delivered at a tertiary hospital in Kuala Lumpur, Malaysia. A content-validated questionnaire consisting of 28 questions testing knowledge and nine questions assessing attitude was used. The highest possible scores for knowledge and attitude were 28 and 45, respectively. The mean score for knowledge was 20.47 (SD 4.06). Mothers who scored ≥ 21 (≥75% of maximum score) were categorized as having “good knowledge” while those who scored <21 were considered to have “poor knowledge”. One hundred and seventy (57%) mothers had good knowledge. Good knowledge was significantly associated with Malay ethnicity, tertiary education, multiparity status and prior breastfeeding experience. Two hundred and nineteen (73%) mothers had a positive attitude (scored ≥34). Malay ethnicity, tertiary education, multiparity status, prior experience in giving expressed milk and feasibility of expressing breast milk at the workplace were significantly associated with a positive attitude. More than half of the mothers had good knowledge, and a higher proportion had a positive attitude towards breast milk expression, storage and usage. Additional support and education should be given to mothers who are: non-Malay, non-tertiary educated and having their first child as these factors were associated with poor knowledge and negative attitude

Urinary tract infections and antibiotic use in pregnancy - qualitative analysis of online forum content
Background Antibiotics are standard treatment for asymptomatic and symptomatic urinary tract infections (UTIs) in pregnancy. Their overuse, however, can contribute to antimicrobial resistance (AMR) and expose the foetus to drugs that might affect its development. Preventative behaviours are currently the best option to reduce incidences of UTIs and to avoid the use of antibiotics in pregnancy. The aim of this study was to explore women’s experiences of UTIs in pregnancy to develop an understanding of their concerns and to optimise and encourage behaviours that facilitate appropriate use of antibiotics.
Methods
An online pregnancy forum in the United Kingdom (UK) was used to collect data on women’s discussions of UTIs. A total of 202 individual threads generated by 675 different usernames were selected for analysis. The data was organised using NVivo 11® software and then analysed qualitatively using inductive thematic analysis.
Results
Women’s perceptions of UTIs and antibiotic use in pregnancy were driven by their pre-natal attachment to the foetus. UTIs were thought to be common and high risk in pregnancy, which meant that antibiotics were viewed as essential in the presence of suspected symptoms. The dominant view about antibiotics was that their use was safe and of little concern in pregnancy. Women reported an emotional reaction to developing a UTI. They coped by seeking information about behaviour change strategies to assist with recovery and through emotional support from the online forum.
Conclusions
Women face dual risks when they experience UTIs; the risk from the infection and the risk from antibiotic treatment. Pre-natal attachment to the foetus is highlighted in the decision making process. The focus is on the shorter term risk from UTIs while undermining the longer term risks from antibiotic use, especially the risk of AMR. A balanced view needs to be presented, and evidence-based infection prevention strategies should be promoted, to women to ensure appropriate antibiotic use in pregnancy, to address the global challenge of AMR

Changes in socioeconomic inequality in Indonesian children's cognitive function from 2000 to 2007: a decomposition analysis
Background: Measuring social inequalities in health is common; however, research examining inequalities in child cognitive function is more limited. We investigated household expenditure-related inequality in children’s cognitive function in Indonesia in 2000 and 2007, the contributors to inequality in both time periods, and changes in the contributors to cognitive function inequalities between the periods. Methods: Data from the 2000 and 2007 round of the Indonesian Family Life Survey (IFLS) were used. Study participants were children aged 7–14 years (n = 6179 and n = 6680 in 2000 and 2007, respectively). The relative concentration index (RCI) was used to measure the magnitude of inequality. Contribution of various contributors to inequality was estimated by decomposing the concentration index in 2000 and 2007. Oaxaca-type decomposition was used to estimate changes in contributors to inequality between 2000 and 2007. Results: Expenditure inequality decreased by 45% from an RCI = 0.29 (95% CI 0.22 to 0.36) in 2000 to 0.16 (95% CI 0.13 to 0.20) in 2007 but the burden of poorer cognitive function was higher among the disadvantaged in both years. The largest contributors to inequality in child cognitive function were inequalities in per capita expenditure, use of improved sanitation and maternal high school attendance. Changes in maternal high school participation (27%), use of improved sanitation (25%) and per capita expenditures (18%) were largely responsible for the decreasing inequality in children’s cognitive function between 2000 and 2007. Conclusions: Government policy to increase basic education coverage for women along with economic growth may have influenced gains in children’s cognitive function and reductions in inequalities in Indonesia.Amelia Maika, Murthy N. Nuclear DNA content variation in life history phases of the Bonnemaisoniaceae (Rhodophyta)
Nuclear DNA content in gametophytes and sporophytes or the prostrate phases of the following species of Bonnemaisoniaceae (Asparagopsis armata, Asparagopsis taxiformis, Bonnemaisonia asparagoides, Bonnemaisonia clavata and Bonnemaisonia hamifera) were estimated by image analysis and static microspectrophotometry using the DNA-localizing fluorochrome DAPI (4′, 6-diamidino-2-phenylindole, dilactate) and the chicken erythrocytes standard. These estimates expand on the Kew database of DNA nuclear content. DNA content values for 1C nuclei in the gametophytes (spermatia and vegetative cells) range from 0.5 pg to 0.8 pg, and for 2C nuclei in the sporophytes or the prostrate phases range from 1.15-1.7 pg. Although only the 2C and 4C values were observed in the sporophyte or the prostrate phase, in the vegetative cells of the gametophyte the values oscillated from 1C to 4C, showing the possible start of endopolyploidy. The results confirm the alternation of nuclear phases in these Bonnemaisoniaceae species, in those that have tetrasporogenesis, as well as those that have somatic meiosis. The availability of a consensus phylogenetic tree for Bonnemaisoniaceae has opened the way to determine evolutionary trends in DNA contents. Both the estimated genome sizes and the published chromosome numbers for Bonnemaisoniaceae suggest a narrow range of values consistent with the conservation of an ancestral genome
β-Amyloid 1-42 Oligomers Impair Function of Human Embryonic Stem Cell-Derived Forebrain Cholinergic Neurons
Cognitive impairment in Alzheimer's disease (AD) patients is associated with a decline in the levels of growth factors, impairment of axonal transport and marked degeneration of basal forebrain cholinergic neurons (BFCNs). Neurogenesis persists in the adult human brain, and the stimulation of regenerative processes in the CNS is an attractive prospect for neuroreplacement therapy in neurodegenerative diseases such as AD. Currently, it is still not clear how the pathophysiological environment in the AD brain affects stem cell biology. Previous studies investigating the effects of the β-amyloid (Aβ) peptide on neurogenesis have been inconclusive, since both neurogenic and neurotoxic effects on progenitor cell populations have been reported. In this study, we treated pluripotent human embryonic stem (hES) cells with nerve growth factor (NGF) as well as with fibrillar and oligomeric Aβ1-40 and Aβ1-42 (nM-µM concentrations) and thereafter studied the differentiation in vitro during 28-35 days. The process applied real time quantitative PCR, immunocytochemistry as well as functional studies of intracellular calcium signaling. Treatment with NGF promoted the differentiation into functionally mature BFCNs. In comparison to untreated cells, oligomeric Aβ1–40 increased the number of functional neurons, whereas oligomeric Aβ1–42 suppressed the number of functional neurons. Interestingly, oligomeric Aβ exposure did not influence the number of hES cell-derived neurons compared with untreated cells, while in contrast fibrillar Aβ1–40 and Aβ1–42 induced gliogenesis. These findings indicate that Aβ1–42 oligomers may impair the function of stem cell-derived neurons. We propose that it may be possible for future AD therapies to promote the maturation of functional stem cell-derived neurons by altering the brain microenvironment with trophic support and by targeting different aggregation forms of Aβ
Complex Reorganization and Predominant Non-Homologous Repair Following Chromosomal Breakage in Karyotypically Balanced Germline Rearrangements and Transgenic Integration
We defined the genetic landscape of balanced chromosomal rearrangements at nucleotide resolution by sequencing 141 breakpoints from cytogenetically-interpreted translocations and inversions. We confirm that the recently described phenomenon of “chromothripsis” (massive chromosomal shattering and reorganization) is not unique to cancer cells but also occurs in the germline where it can resolve to a karyotypically balanced state with frequent inversions. We detected a high incidence of complex rearrangements (19.2%) and substantially less reliance on microhomology (31%) than previously observed in benign CNVs. We compared these results to experimentally-generated DNA breakage-repair by sequencing seven transgenic animals, and revealed extensive rearrangement of the transgene and host genome with similar complexity to human germline alterations. Inversion is the most common rearrangement, suggesting that a combined mechanism involving template switching and non-homologous repair mediates the formation of balanced complex rearrangements that are viable, stably replicated and transmitted unaltered to subsequent generations
CD4 T Cell Immunity Is Critical for the Control of Simian Varicella Virus Infection in a Nonhuman Primate Model of VZV Infection
Primary infection with varicella zoster virus (VZV) results in varicella (more commonly known as chickenpox) after which VZV establishes latency in sensory ganglia. VZV can reactivate to cause herpes zoster (shingles), a debilitating disease that affects one million individuals in the US alone annually. Current vaccines against varicella (Varivax) and herpes zoster (Zostavax) are not 100% efficacious. Specifically, studies have shown that 1 dose of varivax can lead to breakthrough varicella, albeit rarely, in children and a 2-dose regimen is now recommended. Similarly, although Zostavax results in a 50% reduction in HZ cases, a significant number of recipients remain at risk. To design more efficacious vaccines, we need a better understanding of the immune response to VZV. Clinical observations suggest that T cell immunity plays a more critical role in the protection against VZV primary infection and reactivation. However, no studies to date have directly tested this hypothesis due to the scarcity of animal models that recapitulate the immune response to VZV. We have recently shown that SVV infection of rhesus macaques models the hallmarks of primary VZV infection in children. In this study, we used this model to experimentally determine the role of CD4, CD8 and B cell responses in the resolution of primary SVV infection in unvaccinated animals. Data presented in this manuscript show that while CD20 depletion leads to a significant delay and decrease in the antibody response to SVV, loss of B cells does not alter the severity of varicella or the kinetics/magnitude of the T cell response. Loss of CD8 T cells resulted in slightly higher viral loads and prolonged viremia. In contrast, CD4 depletion led to higher viral loads, prolonged viremia and disseminated varicella. CD4 depleted animals also had delayed and reduced antibody and CD8 T cell responses. These results are similar to clinical observations that children with agammaglobulinemia have uncomplicated varicella whereas children with T cell deficiencies are at increased risk of progressive varicella with significant complications. Moreover, our studies indicate that CD4 T cell responses to SVV play a more critical role than antibody or CD8 T cell responses in the control of primary SVV infection and suggest that one potential mechanism for enhancing the efficacy of VZV vaccines is by eliciting robust CD4 T cell responses
