This diploma paper arose as a part of project, which considers by realization of simulated workplace of intensive care unit for biomedical technicians on Faculty of biomedical ingeneering. That was solved by several partial points: list of proposed instruments, space solution of laboratory, orientation calculation of recovery of investment. At first I got acquainted with Standard of Ministry of Health About intensive care unit and I analyzed the present state of this area. The first point was solved by analyse offer of instruments and patient simulator. Than individual components of laboratory were taken up in selection procedures. The second partial point was the concept of space solution of laboratory several instruments and a bed in laboratory 9. I used a recommended computer program Room Arranger for creation of solution. The third partial point was orientation calculation of time of recovery of investment, which proved, that time of recovery of investment is 9 years

MOESM1 of Hydroxyurea down-regulates BCL11A, KLF-1 and MYB through miRNA-mediated actions to induce γ-globin expression: implications for new therapeutic approaches of sickle cell disease

Additional file 1: Figure S1. Time dependent expression of markers of late erythroid differentiation (CD235a and CD71) in erythroid cells treated with HU. Expression of CD235a (A) and CD71 (B) during late erythroid differentiation show no significant differences between HU treated (+) and untreated (−) cells. The lack of significant changes in the expression of these markers is indicates that HU had minimal effect on the processes of erythroid differentiation, thus suggesting that the results, particularly the induction of γ-globin and repression of BCL11A, KLF-1 and MYB, were not artefacts of stress erythropoiesis

Additional file 4: Figure S4. Time dependent luciferase activity in response to HU treatment of anti-miR-151-3p; anti-miR-451 and anti-miR-494 transfected K562 cells. Luciferase activity was highest after 6 and/or 12 h of HU treatment in for all anti-miRNAs, demonstrating a sigmoidal pattern of luminescence. Although less apparent as compared to anti-miR-26b, the anti-miR-151-3p concentration gradient also showed a similar gradual increase in luciferase activity associated with the decrease in anti-miR-151-3p concentration (A and B). There was minimal data to suggest interaction between the MYB-3′-UTR and miR-451 and miR-494, in a 24-hour time course experiment (C and D)

Panel A displays a much higher prevalence of 3.7α-globin gene deletions among patients compared to unaffected controls [HbAA and HbSS combined (p = 0.003)]. This difference was mostly driven by a much lower proportion of 3.7 kb α-globin gene deletions among HbAA controls. Panel B displays the allele frequencies of the 3.7 kb α-globin gene deletions among patient and control. The frequencies were 22% among patients and 11.8% among controls (HbAS and HbAA combined) (p = 0.006). HbAS controls had more 3.7 kb α-globin gene deletions than HbAA controls (p = 0.02).</p

*Hb electrophoresis was also obtained from 344 patients (55.5%) using alkali denaturation test (ADT), with a mean of 11.4±9.4 for HbF and 4.1±2.1 for HbA2 levels. For the analysis, to correct for the skewness of the HbF distribution, we log10-transformed and normalized the data to obtain (after correcting for age, gender, and electrophoresis technique and history of transfusion) the quantitative trait used in the association analysis.
