    VEGF-A/Notch-induced podosomes proteolyse basement membrane collagen-IV during retinal sprouting angiogenesis

    During angiogenic sprouting, endothelial tip cells emerge from existing vessels in a process that requires vascular basement membrane degradation. Here, we show that F-actin/cortactin/P-Src-based matrix-degrading microdomains called podosomes contribute to this step. In vitro, VEGF-A/Notch signaling regulates the formation of functional podosomes in endothelial cells. Using a retinal neovascularization model, we demonstrate that tip cells assemble podosomes during physiological angiogenesis in vivo. In the retina, podosomes are also part of an interconnected network that surrounds large microvessels and impinges on the underlying basement membrane. Consistently, collagen-IV is scarce in podosome areas. Moreover, Notch inhibition exacerbates podosome formation and collagen-IV loss. We propose that the localized proteolytic action of podosomes on basement membrane collagen-IV facilitates endothelial cell sprouting and anastomosis within the developing vasculature. The identification of podosomes as key components of the sprouting machinery provides another opportunity to target angiogenesis therapeutically

Therefore, in this chapter, we refer to the descriptive and topographical anatomy written by French doctors L. Testut and A. Latarjet. In earlier times, for several reasons, the medical surgeon operated all body organs and was involved in resolving the pathology of different areas. Currently and in the future, medical knowledge in thoracic surgery will cover subspecialties specifically divided, for example: assisted video surgery, interventional bronchoscopy, and mediastinoscopies to take mediastinal biopsies using robotic surgery of pulmonary exeresis to shorten the hospitalization period and even the days spent in the intensive care unit

    Using Optimistic Atomic Broadcast in Transaction Processing Systems

    Atomic broadcast primitives are often proposed as a mechanism to allow fault-tolerant cooperation between sites in a distributed system. Unfortunately, the delay incurred before a message can be delivered makes it difficult to implement high performance, scalable applications on top of atomic broadcast primitives. Recently, a new approach has been proposed for atomic broadcast which, based on optimistic assumptions about the communication system, reduces the average delay for message delivery to the application. In this paper, we develop this idea further and show how applications can take even more advantage of the optimistic assumption by overlapping the coordination phase of the atomic broadcast algorithm with the processing of delivered messages. In particular, we present a replicated database architecture that employs the new atomic broadcast primitive in such a way that communication and transaction processing are fully overlapped, providing high performance without relaxing transaction correctness

    Relevance of Biofilm Models in Periodontal Research: From Static to Dynamic Systems

    Microbial biofilm modeling has improved in sophistication and scope, although only a limited number of standardized protocols are available. This review presents an example of a biofilm model, along with its evolution and application in studying periodontal and peri-implant diseases. In 2011, the ETEP (Etiology and Therapy of Periodontal and Peri-Implant Diseases) research group at the University Complutense of Madrid developed an in vitro biofilm static model using representative bacteria from the subgingival microbiota, demonstrating a pattern of bacterial colonization and maturation similar to in vivo subgingival biofilms. When the model and its methodology were standardized, the ETEP research group employed the validated in vitro biofilm model for testing in different applications. The evolution of this model is described in this manuscript, from the mere observation of biofilm growth and maturation on static models on hydroxyapatite or titanium discs, to the evaluation of the impact of dental implant surface composition and micro-structure using the dynamic biofilm model. This evolution was based on reproducing the ideal microenvironmental conditions for bacterial growth within a bioreactor and reaching the target surfaces using the fluid dynamics mimicking the salivary flow. The development of this relevant biofilm model has become a powerful tool to study the essential processes that regulate the formation and maturation of these important microbial communities, as well as their behavior when exposed to different antimicrobial compounds

    The Antimicrobial Activity of Curcumin and Xanthohumol on Bacterial Biofilms Developed over Dental Implant Surfaces

    In search for natural products with antimicrobial properties for use in the prevention and treatment of peri-implantitis, the purpose of this investigation was to evaluate the antimicrobial activity of curcumin and xanthohumol, using an in vitro multi-species dynamic biofilm model including Streptococcus oralis, Actinomyces naeslundii, Veillonella parvula, Fusobacterium nucleatum, Porphyromonas gingivalis and Aggregatibacter actinomycetemcomitans. The antimicrobial activities of curcumin (5 mM) and xanthohumol (100 μM) extracts, and the respective controls, were evaluated with 72-h biofilms formed over dental implants by their submersion for 60 seconds. The evaluation was assessed by quantitative polymerase chain reaction (qPCR), confocal laser scanning microscopy (CLSM) and scanning electron microscopy (SEM). For the data analysis, comparisons were tested applying ANOVA tests with post-hoc Bonferroni corrections to evaluate the antimicrobial activity of both extracts. With qPCR, statistically significant reductions in bacterial counts were observed for curcumin and xanthohumol, when compared to the negative control. The results with CLSM and SEM were consistent with those reported with qPCR.     Multiplicación in vitro de Morinda royoc L. en Sistemas de Inmersión Temporal

    Morinda royoc L. is a medicinal plant which has identified numerous secondary metabolites important for medical and pharmaceutical industry. The use of in vitro culture techniques could contribute to the production of these. The aim of this work was to multiply M. royoc using Temporary Immersion System (TIS). It was used TIS of 1000 ml capacity. Each contained 250 ml MS medium with 4.4 μM benzyladenine (BA) and 2.9 μM indole acetic acid (IAA). The SIT were inoculated with 30 individual explants (shoot tips and nodal segments). It was determined the effect of the immersion frequency (two, four and six immersions of two minutes per day) and the type of explants (shoot tips, nodal segments) on the multiplication of shoots and biomass production. It was found that with four and six immersions per day the highest values of multiplication coefficient and shoot length were obtained. The maximum biomass production was achieved with six immersions per day. No hyperhidricity symptoms were observed in shoots. It was noted that the nodal segments produced more shoots per explant, increased multiplication coefficient and biomass than the apex, while the latter resulted in longer shoots.Keywords: micropropagation, shoots multiplicationMorinda royoc L. es una planta medicinal de la cual se han identificado numerosos metabolitos secundarios de importancia para la industria médica y farmacéutica. El empleo de técnicas de cultivo in vitro podría contribuir a la producción de estos. El objetivo de este trabajo fue multiplicar M. royoc con el uso de SIT. Se emplearon Sistemas de Inmersión Temporal (TIS) de 1000 ml de capacidad. Cada SIT contenía 250 ml de medio de cultivo de multiplicación MS con 4.4 μM de benciladenina (BA) y 2.9 μM de ácido indol acético (AIA). Los SIT fueron inoculados con 30 explantes individuales (ápices y segmentos nodales). Se determinó el efecto de la frecuencia de inmersión (dos, cuatro y seis inmersiones de dos minutos por día) y el tipo de explante (ápices, segmentos nodales) sobre la multiplicación de los brotes y la producción de biomasa. Se comprobó que con cuatro y seis inmersiones por día se obtuvieron los mayores valores de coeficiente de multiplicación y de longitud de los brotes. El máximo valor en la producción de biomasa se alcanzó con seis inmersiones por día. No se observaron síntomas de hiperhidricidad en los brotes. Se observó que los segmentos nodales produjeron más brotes por explante, mayor coeficiente de multiplicación y biomasa que los ápices, mientras que estos últimos dieron lugar a brotes de mayor longitud.Palabras clave: micropropagación, multiplicación de brote

    Socioeconomic, Clinical, and Molecular Features of Breast Cancer Influence Overall Survival of Latin American Women

    Molecular profile of breast cancer in Latin-American women was studied in five countries: Argentina, Brazil, Chile, Mexico, and Uruguay. Data about socioeconomic characteristics, risk factors, prognostic factors, and molecular subtypes were described, and the 60- month overall cumulative survival probabilities (OS) were estimated. From 2011 to 2013, 1,300 eligible Latin-American women 18 years or older, with a diagnosis of breast cancer in clinical stage II or III, and performance status ≦̸ 1 were invited to participate in a prospective cohort study. Face-to-face interviews were conducted, and clinical and outcome data, including death, were extracted from medical records. Unadjusted associations were evaluated by Chi-squared and Fisher’s exact tests and the OS by Kaplan–Meier method. Log-rank test was used to determine differences between cumulative probability curves. Multivariable adjustment was carried out by entering potential confounders in the Cox regression model. The OS at 60 months was 83.9%. Multivariable-adjusted death hazard differences were found for women living in Argentina (2.27), Chile (1.95), and Uruguay (2.42) compared with Mexican women, for older (≥60 years) (1.84) compared with younger (≤40 years) women, for basal-like subtype (5.8), luminal B (2.43), and HER2-enriched (2.52) compared with luminal A subtype, and for tumor clinical stages IIB (1.91), IIIA (3.54), and IIIB (3.94) compared with stage IIA women. OS was associated with country of residence, PAM50 intrinsic subtype, age, and tumor stage at diagnosis. While the latter is known to be influenced by access to care, including cancer screening, timely diagnosis and treatment, including access to more effective treatment protocols, it may also influence epigenetic changes that, potentially, impact molecular subtypes.     Aprender educando a personas con discapacidad: experiencia de Aprendizaje Servicio en Odontología y Magisterio

    Este texto es fruto de un Proyecto de Innovación Docente de la convocatoria 2018-2019, llevado a cabo en la Facultad de odontología de la UCM. El documento desarrolla la metodología ApS desde un punto de vista metodológico, y está estructurado en dos partes. En la primera parte se plantean las necesidades que cubre este tipo de aprendizaje, sobre todo en el área de las Ciencias de la Salud, y las competencias clave que, de forma general, puede ayudar a desarrollar. Además, se ha intentado realizar una guía para marcar las directrices necesarias a la hora de elaborar las distintas fases de este tipo de proyecto. En la segunda parte se describe la experiencia práctica llevada a cabo por un equipo interprofesional formado por estudiantes de Odontología de la UCM y de Educación de la Salle
