    The renal and hepatic distribution of Bence Jones proteins depends on glycosylation: A scintigraphic study in rats

    The aim of the present study was to evaluate renal and liver distribution of two monoclonal immunoglobulin light chains. the chains were purified individually from the urine of patients with multiple myeloma and characterized as lambda light chains with a molecular mass of 28 kDa. They were named BJg (high amount of galactose residues exposed) and BJs (sialic acid residues exposed) on the basis of carbohydrate content. A scintigraphic study was performed on male Wistar rats weighing 250 g for 60 min after iv administration of 1 mg of each protein (7.4 MBq), as the intact proteins and also after carbohydrate oxidation. Images were obtained with a Siemens gamma camera with a high-resolution collimator and processed with a MicroDelta system. Hepatic and renal distribution were established and are reported as percent of injected dose. Liver uptake of BJg was significantly higher than liver uptake of BJs (94.3 vs 81.4%) (P<0.05). This contributed to its greater removal from the intravascular compartment, and consequently lower kidney accumulation of BJg in comparison to BJs (5.7 vs 18.6%) (P<0.05). After carbohydrate oxidation, there was a decrease in hepatic accumulation of both proteins and consequently a higher renal overload. the tissue distribution of periodate-treated BJg was similar to that of native BJs: 82.7 vs 81.4% in the liver and 17.3 vs 18.6% in the kidneys.     Cryptosporidium Spp. And Giardia Spp. In Feces And Water And The Associated Exposure Factors On Dairy Farms

    The aims of this study were to verify the prevalence of Cryptosporidium spp. and Giardia spp. in animal feces and drinking water on dairy farms and to identify a possible relation between the exposure factors and the presence of these parasites. Fecal samples from cattle and humans and water samples were collected on dairy farms in Paraná, Brazil. Analysis of (oo)cysts in the feces was performed by the modified Ziehl-Neelsen staining and centrifugal flotation in zinc sulfate. Test-positive samples were subjected to nested PCR amplification of the 18SSU ribosomal RNA gene for identification of Cryptosporidium and Giardia and of the gp60 gene for subtyping of Cryptosporidium. Microbiological analysis of water was carried out by the multiple-tube method and by means of a chromogenic substrate, and parasitological analysis was performed on 31 samples by direct immunofluorescence and nested PCR of the genes mentioned above. Identification of the species of Cryptosporidium was performed by sequencing and PCR with analysis of restriction fragment length polymorphisms. The prevalence of Giardia and Cryptosporidium was higher in calves than in adults. Among the samples of cattle feces, Cryptosporidium parvum was identified in 41 (64%), C. ryanae in eight (12.5%), C. bovis in four (6.3%), C. andersoni in five (7.8%), and a mixed infection in 20 samples (31.3%). These parasites were not identified in the samples of human feces. Thermotolerant coliform bacteria were identified in 25 samples of water (45.5%). Giardia duodenalis and C. parvum were identified in three water samples. The gp60 gene analysis of C. parvum isolates revealed the presence of two strains (IIaA20G1R1 and IIaA17G2R2) in the fecal samples and one (IIaA17G2R1) in the water samples. The presence of coliforms was associated with the water source, structure and degradation of springs, rain, and turbidity. The prevalence of protozoa was higher in calves up to six months of age.     Adaptability and stability study of soybean lines developed for high yield in Paraná state using four methodologies.

    An adaptability and stability study was carried out using soybean yield data from several locations in Paraná State, obtained from 1990 to 1999. The main objectives were: a) to check the efficiency of the Embrapa Soja breeding program for selecting the highest yielding lines with specific (regional) or broad adaptation; b) to analyze the performance of the control cultivars under favorable and unfavorable conditions; c) to identify the best stability methodology for inbred line evaluation. The evaluated lines were classified into early (L), semi- early (M) and medium (N) maturity groups. A randomized complete block design with four replications was used in each location and all analyses were carried out by maturity group. The number of M maturity lines that scored higher than the controls BR-16 and Embrapa 4 increased along the years, especially in unfavorable environments. On the other hand, the number of L and N maturity groups scoring higher than the controls either remained constant or fluctuated (increasing or decreasing) with time. The controls FT-Guaíra, Embrapa 4, BR-16 and FT-10 and the great majority of the tested lines showed broad adaptation. IAS 5 and FT-Abyara showed adaptation to poor environments and also broad adaptation. The analysis of the means in favorable and unfavorable environments was the most efficient method for soybean line evaluation.     Proposta de classificação dos coeficientes de variação em relação à produtividade e altura da planta de soja.

    O objetivo deste trabalho foi definir classificações de coeficientes de variação para produtividade e altura da planta de soja. Os dados foram obtidos de ensaios intermediários e finais realizados nos estados do Paraná e do Mato Grosso. Foram realizadas classificações distintas para cada localização e ciclo reprodutivo. Considerando-se a média e o desvio-padrão dos coeficientes de variação obtidos das análises de variância dos ensaios, os coeficientes foram classificados como baixo, médio, alto e muito alto. Uma classificação adicional foi feita utilizando a mediana e o pseudo-sigma, em substituição à média e ao desvio-padrão, respectivamente. A classificação dos coeficientes de variação dependeu do caráter e da localização, mas não variou muito em razão do ciclo reprodutivo. Os critérios adotados (média e desvio-padrão ou mediana e pseudo-sigma) foram semelhantes (independentemente da distribuição dos coeficientes de variação) e satisfatórios para determinar a precisão experimental. O limite máximo de coeficiente de variação aceitável para produtividade é de 16% e para altura da planta é de 12%

    Uma avaliacao da eficiencia do programa de melhoramento genetico de soja no Parana no periodo de 1981 a 1986 e realizada utilizando-se dados comumente obtidos nos testes de avaliacao de linhagens da rede oficial de experimentacao. Os dados foram obtidos no Ensaio Intermediario de Linhagens de Soja, constituido de grupos de maturacao precoce e semi-precoce. Os ganhos geneticos obtidos foram de 1,8% e 1,3% para os genotipos de maturacao precoce e semi-precoce, respectivamente. Os dois grupos de maturacao englobam aproximadamente 70% de todo o material cultivado no Estado. Os resultados indicam que e possivel estimar o progresso genetico utilizando informacoes disponiveis na rotina de avaliacao de linhagens. Os ganhos geneticos obtidos mostram que os esforcos empreendidos na busca e cultivares adaptadas e produtivas tem produzido resultados satisfatorios
