    Visualized effect of oxidation on magnetic recording fidelity in pseudo-single-domain magnetite particles

    Magnetite (​Fe3O4) is an important magnetic mineral to Earth scientists, as it carries the dominant magnetic signature in rocks, and the understanding of its magnetic recording fidelity provides a critical tool in the field of palaeomagnetism. However, reliable interpretation of the recording fidelity of ​Fe3O4 particles is greatly diminished over time by progressive oxidation to less magnetic iron oxides, such as maghemite (γ-Fe2O3), with consequent alteration of remanent magnetization potentially having important geological significance. Here we use the complementary techniques of environmental transmission electron microscopy and off-axis electron holography to induce and visualize the effects of oxidation on the magnetization of individual nanoscale ​Fe3O4 particles as they transform towards γ-Fe2O3. Magnetic induction maps demonstrate a change in both strength and direction of remanent magnetization within ​Fe3O4 particles in the size range dominant in rocks, confirming that oxidation can modify the original stored magnetic information

    The role of VGI towards 3D property cadastral systems (2): a purpose driven web application

    VGI has not proved to be readily suitable to replace well-established accurate methods and technologies such as those of full standard cadastral surveys. Even so, VGI potentialities as relevant source of geospatial data have been widely acknowledged. As such, some authors have defended that VGI may in fact play an important role such as at a local cadastral jurisdiction level towards local spatial data infrastructures. As far as property cadastre is concerned, the full extent 3D complexity inside a property is in many instances only known to their occupants, thus making crowd sourcing perhaps the only economically feasible approach for its capture. While the crowd cannot be expected to conduct a full cadastral survey, it may be possible to ask them to indicate at least the location of complex 3D situations and thus to facilitate local authorities’ understanding of the extent of some cadastral issues. As such, it was argued in our previous work that geoinformation from the crowd might in fact be taken into account as an interim step before a full surveyed 3D cadastre is eventually achieved. As such, possible room for VGI in the context of 3D cadastre was discussed, and a hierarchical framework of levels of data acquisition to be used at local cadastral jurisdiction level was proposed. Such framework is revisited in this paper.Given context above, this paper focuses primarily on two aspects. Firstly, to review technical requirements of the official cadastral process in Portugal in order to identify which sorts of cadastral data are likely to be acquirable/not acquirable through VGI. Secondly, to design and to implement the prototype of a web-based application (IGV3Dcad) envisaged for general public usage to flag different land and property ownership situations. Having information about the extent of the 2D/3D issue is also fundamental to making a decision as to whether a 3D cadastral approach is actually needed and hence to further invest resources in even more expensive 3D survey

    Highly Dynamic Host Actin Reorganization around Developing Plasmodium Inside Hepatocytes

    Plasmodium sporozoites are transmitted by Anopheles mosquitoes and infect hepatocytes, where a single sporozoite replicates into thousands of merozoites inside a parasitophorous vacuole. The nature of the Plasmodium-host cell interface, as well as the interactions occurring between these two organisms, remains largely unknown. Here we show that highly dynamic hepatocyte actin reorganization events occur around developing Plasmodium berghei parasites inside human hepatoma cells. Actin reorganization is most prominent between 10 to 16 hours post infection and depends on the actin severing and capping protein, gelsolin. Live cell imaging studies also suggest that the hepatocyte cytoskeleton may contribute to parasite elimination during Plasmodium development in the liver

    Comparative analysis of the lambda-interferons IL-28A and IL-29 regarding their transcriptome and their antiviral properties against hepatitis C virus.

    Specific differences in signaling and antiviral properties between the different Lambda-interferons, a novel group of interferons composed of IL-28A, IL-28B and IL-29, are currently unknown. This is the first study comparatively investigating the transcriptome and the antiviral properties of the Lambda-interferons IL-28A and IL-29. Expression studies were performed by microarray analysis, quantitative PCR (qPCR), reporter gene assays and immunoluminometric assays. Signaling was analyzed by Western blot. HCV replication was measured in Huh-7 cells expressing subgenomic HCV replicon. All hepatic cell lines investigated as well as primary hepatocytes expressed both IFN-λ receptor subunits IL-10R2 and IFN-λR1. Both, IL-28A and IL-29 activated STAT1 signaling. As revealed by microarray analysis, similar genes were induced by both cytokines in Huh-7 cells (IL-28A: 117 genes; IL-29: 111 genes), many of them playing a role in antiviral immunity. However, only IL-28A was able to significantly down-regulate gene expression (n = 272 down-regulated genes). Both cytokines significantly decreased HCV replication in Huh-7 cells. In comparison to liver biopsies of patients with non-viral liver disease, liver biopsies of patients with HCV showed significantly increased mRNA expression of IL-28A and IL-29. Moreover, IL-28A serum protein levels were elevated in HCV patients. In a murine model of viral hepatitis, IL-28 expression was significantly increased. IL-28A and IL-29 are up-regulated in HCV patients and are similarly effective in inducing antiviral genes and inhibiting HCV replication. In contrast to IL-29, IL-28A is a potent gene repressor. Both IFN-λs may have therapeutic potential in the treatment of chronic HCV

    Synthesis, structural and magnetic characterization of lead-metaniobate/cobalt-ferrite nanocomposite films deposited by pulsed laser ablation

    Detailed structural, microstructural and magnetic measurements were performed on (PbNb2O6)(1-x) -(CoFe2O4) (x) nanocomposite thin films deposited by laser ablation on Si(001)\Pt substrates, with different cobalt ferrite concentrations. The tuning of the lead concentration, due to the lead volatility, was found to be particularly important in order to obtain the orthorhombic (ferroelectric) lead niobate phase. The lattice parameter of CoFe2O4 was below the bulk value, indicating the presence of compressive strains on this phase. A magnetic anisotropy was observed, which favored the orientation of the magnetization in the direction perpendicular to the plane of the films, for cobalt ferrite concentrations 40-50 %. The shape, stress and magnetocrystalline anisotropy fields on the composites were calculated and compared.     Mood is a key determinant of cognitive performance in community-dwelling older adults: a cross-sectional analysis

    First Online: 06 October 2012Identification of predictors of cognitive trajectories through the establishment of composite or single-parameter dimensional categories of cognition and mood may facilitate development of strategies to improve quality of life in the elderly. Participants (n = 487, aged 50+ years) were representative of the Portuguese population in terms of age, gender, and educational status. Cognitive and mood profiles were established using a battery of neurocognitive and psychological tests. Data were subjected to principal component analysis to identify core dimensions of cognition and mood, encompassing multiple test variables. Dimensions were correlated with age and with respect to gender, education, and occupational status. Cluster analysis was applied to isolate distinct patterns of cognitive performance and binary logistic regression models to explore interrelationships between aging, cognition, mood, and socio-demographic characteristics. Four main dimensions were identified: memory, executive function, global cognitive status, and mood. Based on these, strong and weak cognitive performers were distinguishable. Cluster analysis revealed further distinction within these two main categories into very good, good, poor, and very poor performers. Mood was the principal factor contributing to the separation between very good and good, as well as poor and very poor, performers. Clustering was also influenced by gender and education, albeit to a lesser extent; notably, however, female gender × lower educational background predicted significantly poorer cognitive performance with increasing age. Mood has a significant impact on the rate of cognitive decline in the elderly.     Multiplex PCR technique could be an alternative approach for early detection of leprosy among close contacts - a pilot study from India

    <p>Abstract</p> <p>Background</p> <p>Implementation of Multi drug Therapy (MDT) regimen has resulted in the decline of the total number of leprosy cases in the world. Though the prevalence rate has been declining, the incidence rate remains more or less constant and high in South East Asian countries particularly in India, Nepal, Bangladesh, Pakistan and Srilanka. Leprosy, particularly that of multibacillary type spreads silently before it is clinically detected. An early detection and treatment would help to prevent transmission in the community. Multiplex PCR (M-PCR) technique appears to be promising towards early detection among contacts of leprosy cases.</p> <p>Methods</p> <p>A total of 234 paucibacillary (PB) and 205 multibacillary (MB) leprosy cases were studied in a community of an endemic area of Bankura district of West Bengal (Eastern India). They were assessed by smear examination for acid-fast bacilli (AFB) and M-PCR technique. These patients were treated with Multidrug Therapy (MDT) as prescribed by WHO following detection. A total of 110 MB and 72 PB contacts were studied by performing M-PCR in their nasal swab samples.</p> <p>Results</p> <p>83.4% of MB patients were observed to be positive by smear examination for AFB and 89.2% by M-PCR. While 22.2% of PB patients were found to be positive by smear examination for AFB, 80.3% of these patients were positive by M-PCR. Among leprosy contacts (using M-PCR), 10.9% were found to be positive among MB contacts and 1.3% among PB contacts. Interestingly, two contacts of M-PCR positive MB cases developed leprosy during the period of two years follow up.</p> <p>Conclusion</p> <p>The M-PCR technique appears to be an efficient tool for early detection of leprosy cases in community based contact tracing amongst close associates of PB and MB cases. Early contact tracing using a molecular biology tool can be of great help in curbing the incidence of leprosy further.</p
