    Motoneurone synchronization for intercostal and abdominal muscles: interneurone influences in two different species

    The contribution of branched-axon monosynaptic inputs in the generation of short-term synchronization of motoneurones remains uncertain. Here, synchronization was measured for intercostal and abdominal motoneurones supplying the lower thorax and upper abdomen, mostly showing expiratory discharges. Synchronization in the anaesthetized cat, where the motoneurones receive a strong direct descending drive, is compared with that in anaesthetized or decerebrate rats, where the direct descending drive is much weaker. In the cat, some examples could be explained by branched-axon monosynaptic inputs, but many others could not, by virtue of peaks in cross-correlation histograms whose widths (relatively wide) and timing indicated common inputs with more complex linkages, e.g., disynaptic excitatory. In contrast, in the rat, correlations for pairs of internal intercostal nerves were dominated by very narrow peaks, indicative of branched-axon monosynaptic inputs. However, the presence of activity in both inspiration and expiration in many of the nerves allowed additional synchronization measurements between internal and external intercostal nerves. Time courses of synchronization for these often consisted of combinations of peaks and troughs, which have never been previously described for motoneurone synchronization and which we interpret as indicating combinations of inputs, excitation of one group of motoneurones being common with either excitation or inhibition of the other. Significant species differences in the circuits controlling the motoneurones are indicated, but in both cases, the roles of spinal interneurones are emphasised. The results demonstrate the potential of motoneurone synchronization for investigating inhibition and have important general implications for the interpretation of neural connectivity measurements by cross-correlation

    Chemical Composition and Antifungal Properties of Essential Oil of Origanum vulgare Linnaeus (Lamiaceae) against Sporothrix schenckii and Sporothrix brasiliensis

    Purpose: To evaluate the effect of the essential oil of Origanum vulgare Linnaeus (Lamiaceae) on the growth of Sporothrix schenckii and Sporothrix brasiliensis.Methods: The chemical composition of the essential oil was investigated by gas chromatography/flame ionization detector (GC-FID). The minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) were determined by broth micro-dilution method. Scanning electron microscopy (SEM) was also performed to reveal morphological alterations in Sporothrix spp. cells.Results: The major components of the essential oil were Îł-terpinene (30.5%), carvacrol (15.7 %) and 4-terpineol (13.0 %). Îł-Terpinene showed potential antifungal activity with MIC ranging from 62.5 to 500.0 ÎŒg mL-1 for S. schenckii, and 125.0 to 250.0 ÎŒg mL-1 for S. brasiliensis. SEM micrographs revealed morphological alterations in hyphae and reduction of the adhered conidia numbers.Conclusion: Origanum vulgare Linnaeus essential oil possesses potential antifungal activity, and can, therefore, can be developed as an alternative agent for the treatment of sporotrichosis.Keywords: Antifungal Activity, Essential Oil, Gas Chromatography, Origanum vulgare, Sporotrichosi

    Reduction of Nondeterministic Tree Automata

    We present an efficient algorithm to reduce the size of nondeterministic tree automata, while retaining their language. It is based on new transition pruning techniques, and quotienting of the state space w.r.t. suitable equivalences. It uses criteria based on combinations of downward and upward simulation preorder on trees, and the more general downward and upward language inclusions. Since tree-language inclusion is EXPTIME-complete, we describe methods to compute good approximations in polynomial time. We implemented our algorithm as a module of the well-known libvata tree automata library, and tested its performance on a given collection of tree automata from various applications of libvata in regular model checking and shape analysis, as well as on various classes of randomly generated tree automata. Our algorithm yields substantially smaller and sparser automata than all previously known reduction techniques, and it is still fast enough to handle large instances.Comment: Extended version (including proofs) of material presented at TACAS 201

    Dual Space of a Lattice as the Completion of a Pervin Space

A Pervin space is a set X equipped with a set of subsets,called the blocks of the Pervin space. Blocks are closed under finite intersections and finite unions and hence form a lattice of subsets of X. Pervin spaces are thus easier to define than topological spaces or (quasi)-uniform spaces. As a consequence, most of the standard topological notions, like convergence and cluster points, specialisation order, filtersand Cauchy filters, complete spaces and completion are much easier to define for Pervin spaces. In particular, the completion of a Pervin space turns out to be the dual space (in the sense of Stone) of the original lattice.We show that any lattice of subsets can be described by a set of inequations of the form u ≀ v, where u and v are elements of its dual space. Applications to formal languages and complexity classes are given.Cet article de synthĂšse prĂ©sente des rĂ©sultats bien connus sous un nouvel angle. Un espace de Pervin est unensemble X Ă©quipĂ© d'un ensemble de parties, appelĂ© les blocs de l'espace de Pervin.     Pulmonary cryptococcosis induces chitinase in the rat

    <p>Abstract</p> <p>Background</p> <p>We previously demonstrated that chronic pulmonary infection with <it>Cryptococcus neoformans </it>results in enhanced allergic inflammation and airway hyperreactivity in a rat model. Because the cell wall of <it>C. neoformans </it>consists of chitin, and since acidic mammalian chitinase (AMCase) has recently been implicated as a novel mediator of asthma, we sought to determine whether such infection induces chitinase activity and expression of AMCase in the rat.</p> <p>Methods</p> <p>We utilized a previously-established model of chronic <it>C. neoformans </it>pulmonary infection in the rat to analyze the activity, expression and localization of AMCase.</p> <p>Results</p> <p>Our studies indicate that intratracheal inoculation of <it>C. neoformans </it>induces chitinase activity within the lung and bronchoalveolar lavage fluid of infected rats. Chitinase activity is also elicited by pulmonary infection with other fungi (e.g. <it>C. albicans</it>), but not by the inoculation of dead organisms. Enhanced chitinase activity reflects increased AMCase expression by airway epithelial cells and alveolar macrophages. Systemic cryptococcosis is not associated with increased pulmonary chitinase activity or AMCase expression.</p> <p>Conclusion</p> <p>Our findings indicate a possible link between respiratory fungal infections, including <it>C. neoformans</it>, and asthma through the induction of AMCase.</p

    Influence of dietary supplementation with an amino acid mixture on inflammatory markers, immune status and serum proteome in lps-challenged weaned piglets

    In order to investigate the effect of a dietary amino acid mixture supplementation in lipopolysaccharide (LPS)-challenged weaned piglets, twenty-seven 28-day-old (8.2 ± 1.0 kg) newly weaned piglets were randomly allocated to one of three experimental treatments for five weeks. Diet 1: a CTRL treatment. Diet 2: an LPS treatment, where piglets were intraperitoneally administered LPS (25 ”g/kg) on day 7. Diet 3: an LPS+MIX treatment, where piglets were intraperitoneally administered LPS on day 7 and fed a diet supplemented with a mixture of 0.3% of arginine, branched-chain amino acids (leucine, valine, and isoleucine), and cystine (MIX). Blood samples were drawn on day 10 and day 35, and serum was analysed for selected chemical parameters and proteomics. The LPS and LPS+MIX groups exhibited an increase in haptoglobin concentrations on day 10. The LPS group showed an increased cortisol concentration, while this concentration was reduced in the LPS+MIX group compared to the control group. Similarly, the LPS+MIX group showed a decreased haptoglobin concentration on day 35 compared to the two other groups. Immunoglobulin concentrations were affected by treatments. Indeed, on day 10, the concentrations of IgG and IgM were decreased by the LPS challenge, as illustrated by the lower concentrations of these two immunoglobulins in the LPS group compared to the control group. In addition, the supplementation with the amino acid mixture in the LPS+MIX further decreased IgG and increased IgM concentrations compared to the LPS group. Although a proteomics approach did not reveal important alterations in the protein profile in response to treatments, LPS-challenged piglets had an increase in proteins linked to the immune response, when compared to piglets supplemented with the amino acid mixture.     Classification of patients with knee osteoarthritis in clinical phenotypes: data from the osteoarthritis initiative

    <div><p>Objectives</p><p>The existence of phenotypes has been hypothesized to explain the large heterogeneity characterizing the knee osteoarthritis. In a previous systematic review of the literature, six main phenotypes were identified: Minimal Joint Disease (MJD), Malaligned Biomechanical (MB), Chronic Pain (CP), Inflammatory (I), Metabolic Syndrome (MS) and Bone and Cartilage Metabolism (BCM). The purpose of this study was to classify a sample of individuals with knee osteoarthritis (KOA) into pre-defined groups characterized by specific variables that can be linked to different disease mechanisms, and compare these phenotypes for demographic and health outcomes.</p><p>Methods</p><p>599 patients were selected from the OAI database FNIH at 24 months’ time to conduct the study. For each phenotype, cut offs of key variables were identified matching the results from previous studies in the field and the data available for the sample. The selection process consisted of 3 steps. At the end of each step, the subjects classified were excluded from the further classification stages. Patients meeting the criteria for more than one phenotype were classified separately into a ‘complex KOA’ group.</p><p>Results</p><p>Phenotype allocation (including complex KOA) was successful for 84% of cases with an overlap of 20%. Disease duration was shorter in the MJD while the CP phenotype included a larger number of Women (81%). A significant effect of phenotypes on WOMAC pain (F = 16.736 p <0.001) and WOMAC physical function (F = 14.676, p < 0.001) was identified after controlling for disease duration.</p><p>Conclusion</p><p>This study signifies the feasibility of a classification of KOA subjects in distinct phenotypes based on subgroup-specific characteristics.</p></div

    Errors in chromosome segregation during oogenesis and early embryogenesis

    Errors in chromosome segregation occurring during human oogenesis and early embryogenesis are very common. Meiotic chromosome development during oogenesis is subdivided into three distinct phases. The crucial events, including meiotic chromosome pairing and recombination, take place from around 11 weeks until birth. Oogenesis is then arrested until ovulation, when the first meiotic division takes place, with the second meiotic division not completed until after fertilization. It is generally accepted that most aneuploid fetal conditions, such as trisomy 21 Down syndrome, are due to maternal chromosome segregation errors. The underlying reasons are not yet fully understood. It is also clear that superimposed on the maternal meiotic chromosome segregation errors, there are a large number of mitotic errors taking place post-zygotically during the first few cell divisions in the embryo. In this chapter, we summarise current knowledge of errors in chromosome segregation during oogenesis and early embryogenesis, with special reference to the clinical implications for successful assisted reproduction
