    Development of a critical structure state alarm system based on the instrumentation of the Botafoc breakwater nº 8 caisson

    Balearic Port Authority has developed an instrumentation system for the #8 caisson of the Botafoc breakwater that integrates 12 pressure sensors located at three surfaces, two in contact with the sea water and another with the bottom. This design was completed with an inertial system that measures the angular velocities and the accelerations over the three Cartesian axes. Consequently, the system measures actions (pressures) and reactions (movements and accelerations) experimented by the caisson, due to sea waves and/or other service loads. R+D department of the Port Authority and Polytechnic University of Madrid are working on two directions, the development of new theories on vertical breakwater design that go beyond Goda and Sainflou, and on the creation of a real-time critical structure alarm system, based on the instrumentation installed. This alarm system has two main parts: the instrumentation itself that collects data and processes it on real-time (the data processing compares the pressure law suffered by the caisson in every step process with the design critical state of the caisson, in this case the Goda pressure law for a 6.5 m wave), giving a security coefficient that points out the risk level on real-time; and the alarm system consisting of a monitoring panel located in the Port Control Center that shows the risk level and advises in case of an incidental evacuation of this critical portuary installation

    Dominant negative phenotype of Bacillus thuringiensis Cry1Ab, Cry11Aa and Cry4Ba mutants suggest hetero-oligomer formation among different Cry toxins.

    Background - Bacillus thuringiensis Cry toxins are used worldwide in the control of different insect pests important in agriculture or in human health. The Cry proteins are pore-forming toxins that affect the midgut cell of target insects. It was shown that non-toxic Cry1Ab helix a-4 mutants had a dominant negative (DN) phenotype inhibiting the toxicity of wildtype Cry1Ab when used in equimolar or sub-stoichiometric ratios (1:1, 0.5:1, mutant:wt) indicating that oligomer formation is a key step in toxicity of Cry toxins. Methodology/Principal Findings - The DN Cry1Ab-D136N/T143D mutant that is able to block toxicity of Cry1Ab toxin, was used to analyze its capacity to block the activity against Manduca sexta larvae of other Cry1 toxins, such as Cry1Aa, Cry1Ac, Cry1Ca, Cry1Da, Cry1Ea and Cry1Fa. Cry1Ab-DN mutant inhibited toxicity of Cry1Aa, Cry1Ac and Cry1Fa. In addition, we isolated mutants in helix a-4 of Cry4Ba and Cry11Aa, and demonstrate that Cry4Ba-E159K and Cry11Aa-V142D are inactive and completely block the toxicity against Aedes aegypti of both wildtype toxins, when used at sub-stoichiometric ratios, confirming a DN phenotype. As controls we analyzed Cry1Ab-R99A or Cry11Aa-E97A mutants that are located in helix a-3 and are affected in toxin oligomerization. These mutants do not show a DN phenotype but were able to block toxicity when used in 10:1 or 100:1 ratios (mutant:wt) probably by competition of binding with toxin receptors. Conclusions/Significance - We show that DN phenotype can be observed among different Cry toxins suggesting that may interact in vivo forming hetero-oligomers. The DN phenotype cannot be observed in mutants affected in oligomerization, suggesting that this step is important to inhibit toxicity of other toxin

    Sea Level in the Strait of Gibraltar: Tides

    A network of tide gauges with eight observation recording points was in operation in the Strait of Gibraltar during the years 1984 and 1985, which made it possible to draw up detailed charts of the tides showing their refined structure. For the first order, the Strait of Gibraltar represents the nodal line of the stationary wave of the western Mediterranean, which, hypothetically, would end at the Cadiz meridian (6°17.0W). The tide is basically semi-diurnal; on average, 90% of the energy is associated with the second order and, for this, the Strait of Gibraltar tends to represent an antinode of the stationary wave, although the influence of the bottom topography and the rotation is interpreted in phase delays in the sill area (slightly progressive wave) and in increased non-linear constituents of higher orders. The radiational tide S is evaluated in the area studied and it is ascertained that it shows the same characteristics as the tides having strictly gravitational constituents, which implies that it is fundamentally co-oscillating. Order 4 displays characteristics of resonant amplification due to the existence of the free oscillation mode of the western Mediterranean basin the period of which is close to 6 hours. Of order 3 it should be stressed that M3, though small, is perfectly detectable in the area

    Percolation-Modeling Comparison Between the Conductivities of Zinc-Graphene Quantum Dot Nanocomposite and Graphite during Extracellular Electron Transfer in Microbial Fuel Cell Electrodes

    While promising as an energy production alternative through its sustainability and wastewater treatment utility, a microbial fuel cell is not widely used due to its low power output and high cost. The development of advanced electrode materials is currently being pursued to solve this problem. A zinc-graphene quantum dot nanocomposite was modeled using percolation theory as a prospective advanced electrode material. During extracellular electron transfer, the electrical conductivity properties of the material were studied through cellular percolation models, percolation probability functions, and electrical conductivity curves. These models were compared against those of the conventional graphite electrodes and the leading graphene electrodes. The nanocomposite was found to conduct at low probabilities of open sites and exhibit the highest electrical conductivity of the three materials for the longest duration across the interval. Based on the models, Zn-GQD was demonstrated to be an ideal MFC electrode material for its balance between the early onset of conduction and decently high electrical conductivity

    3D Printable Conducting and Biocompatible PEDOT-graft-PLA Copolymers by Direct Ink Writing

    Tailor-made polymers are needed to fully exploit the possibilities of additive manufacturing, constructing complex, and functional devices in areas such as bioelectronics. In this paper, the synthesis of a conducting and biocompatible graft copolymer which can be 3D printed using direct melting extrusion methods is shown. For this purpose, graft copolymers composed by conducting polymer poly(3,4-ethylenedioxythiophene) (PEDOT) and a biocompatible polymer polylactide (PLA) are designed. The PEDOT-g-PLA copolymers are synthesized by chemical oxidative polymerization between 3,4-ethylenedioxythiophene and PLA macromonomers. PEDOT-g-PLA copolymers with different compositions are obtained and fully characterized. The rheological characterization indicates that copolymers containing below 20 wt% of PEDOT show the right complex viscosity values suitable for direct ink writing (DIW). The 3D printing tests using the DIW methodology allows printing different parts with different shapes with high resolution (200\ua0\ub5m). The conductive and biocompatible printed patterns of PEDOT-g-PLA show excellent cell growth and maturation of neonatal cardiac myocytes cocultured with fibroblasts
