    Evaluation of Trace Element Levels and Antioxidant Metabolism in Cattle with Cutaneous Papillomatosis

    Background: Cutaneous papillomas are benign proliferative neoplasms. The aetiology and pathogenesis of the disease are very complex. Papillomatosis lesions can be detected anywhere on the body, frequently appearing on the surface of the head, neck, teats, udder, external genital mucosa and upper part of the gastrointestinal tract. The disease is not life threatening but due to loss of productivity, owners often cull animals with papillomatosis. The relationships between trace elements, heavy metals and cancer are still researching. The aim of this study was the evaluation of the relationships between trace elements and antioxidant metabolism, and cutaneous papillomatosis, in cattle.Materials, Methods & Results: Eleven cattle with cutaneous papillomatosis and 10 healthy cattle constituted the study group and control group, respectively. Clinical examinations were performed on all animals. Blood samples were collected and Cr, Cu, Fe, Mg, Mn, Se, Zn, As, B, Ni, Si and Co levels were determined. The analyses were conducted with an inductively coupled plasma-optical emission spectrophotometer. In addition, catalase (CAT), glutathione (GSH), malondialdehyde (MDA) and ceruloplasmin (Cp) concentrations were measured. The Ni, Si and MDA levels of the study group were significantly higher than those of the control group (P < 0.05) but the Se levels of the study group were significantly lower than in the control group (P < 0.05). The normality of the data was determined with the Shapiro-Wilk Test and the Independent Samples t-Test or Mann-Whitney U test was used for statistical comparison of the groups.Discussion: Trace elements are essential components of biological systems. The levels of trace elements, such as Cu, Co, I, Se, Zn and Mn, which are affected by grazing activity, influence ruminants’ reproductive performance and other production parameters. The elements Al, As, Cr, Ni and Sn are also presumed to be essential for ruminant metabolism, although sufficient research data are not available. In the present study, Cr, Cu, Fe, Mg, Mn, Zn, As, B and Co levels were not significantly different between the papilloma and control groups. Se is an important trace element for skin health. This study revealed a mean serum Se level in animals with papillomatosis that was significantly lower than in the healthy group. Although the mode of anticancer activity of Se is not clear, some factors, such as antioxidant protection, improved immune system surveillance, carcinogen detoxification, modulation of cell proliferation and inhibition of tumour cell invasion and angiogenesis, are important. Ni is classified as a toxic heavy metal. In addition, it could be carcinogenic to humans. Ni level was two times higher in the papillomatosis group than in the healthy animals, in the present study. This result may indicate that Ni has a role in the genesis of papillomatosis in cattle. Si injection can cause inflammation, granulomas and cancer. There are few reports about Si levels in animals and the significant difference in the Si levels in cattle with cutaneous papillomatosis reported in this study may be a new indicator for the disease. The skin contains antioxidant molecules that include GSH, alpha-tocopherol or vitamin E, ascorbic acid or vitamin C, glutathione peroxidases, glutathione reductase, glutathione S-transferases (GSTs), superoxide dismutases (SODs), CAT and quinine reductase. There is strong interaction between ROS and/or their oxidant products and antioxidant molecules in the skin. In this study, although MDA levels were higher in the study group than in the control group, there was no significant difference between the groups in their CAT levels, and also in the GSH and CP levels. In conclusion, these results may evidence the systemic effects of papillomatosis on trace element levels and on oxidant metabolism in cattle

    Coins are foreign objects that are commonly ingested by children and pets due to their shiny appearance and bright colors. The current study investigated whether Turkish coins can lead to zinc (Zn) and copper (Cu) toxicity as a result of exposure to simulated gastric juice (i.e. hydrochloric acid solution simulating the gastric environment). Five groups of coins were exposed to simulated gastric juice (0.15 N, pH:1-2) for a period of 4 (Group 1), 12 (Group 2), 24 (Group 3), 48 (Group 4), 72 (Group 5) and 120 h (Group 5) at body temp. (37 degrees C). Zinc and copper levels were determined in the gastric acid solution by using an inductively coupled plasma optical emission spectrophotometer (ICP-OES). The coins were also evaluated for corrosive damage and weight loss. Group 5 had statistically higher Cu and Zn levels versus the other groups. However, at body temp. (37 degrees C), copper and zinc levels increased steadily in parallel to the duration for which the coin remained in the stomach after ingestion. After 120-hour exposure at 37 degrees C, all coins had various types of damage compared to the baseline, such as color alteration, erosion, and visible surface cavities. The mechanisms relating to local and systemic copper and zinc toxicity caused by coin ingestion is yet to be clarified for both Turkish and international coins. Therefore, it can be foreseen that intervention is required in the first 24 h after ingesting 5 kurus coins and, unless removed spontaneously in 48 h, such intervention is needed for the other coins

    Analysis of the Relationship Between Hemorheologic Parameters, Aluminum, Manganese, and Selenium in Smokers

    Smoking is a significant risk factor in fatal pathologies including cardio-cerebrovascular and respiratory diseases. Aluminum (Al) is a toxic element without known biological function, but with recognized toxic effects. Manganese (Mn) and selenium (Se) are essential trace elements involved in cellular antioxidant defense mechanisms. Al, Mn, and Se carry out their metabolic activities via blood flow and tissue oxygenation. The structure and number of red blood cells (RBC) play important role in tissue oxygenation throughout blood flow. Increased hematocrit (Hct) as a result of probable hypoxia induces disturbed blood flow, RBC aggregation (RBC Agg), RBC deformability index (Tk), and oxygen delivery index (ODI). Therefore, we aimed to investigate the effects of altered Al, Mn, and Se levels on number, structure, and function of RBCs (Hct, blood and plasma viscosity (BV and PV, respectively), RBC Agg, Tk, ODI) in smokers without diagnosis of chronic obstructive pulmonary disease (COPD) in a study group (n=128) categorized as ex-smokers (ES), smokers (S), and healthy controls (HC). Elements were analyzed in serum using ICP-OES. BV and PV were measured via Brookfield and Harkness viscometers at 37 degrees C, respectively. Smokers had statistically higher serum Al and Mn levels, BV, RBC, Hgb, Hct, PV, fibrinogen, RBC Agg, Tk(45), and pulmonary blood flow rate, but lower serum Se levels and ODI45 values versus HC. In conclusion, increased Al, Mn, and hemorheological parameters and decreased Se and ODI45 might result from inflammatory response in defense mechanism in smokers without diagnosis of COPD. Our results point out that serum Al, Mn, and Se with hemorheological parameters may be beneficial markers of tissue oxygenation and defense mechanism before the clinic onset of COPD in smokers

    Evaluation of trace elements in essential thrombocytosis and reactive thrombocytosis

    Background: Trace elements (TE) are vital for cellular mechanisms at biological, chemical and molecular levels. The effects of TE in diagnosis, progression and treatment of essential thrombocytosis (ET), which is one of the chronic myeloproliferative neoplasms is a rare clonal stem cell disease characterized by increased thrombocyte numbers with impaired function, have not been elucidated in detail yet. The aim of the present study was to investigate the effects of TE alterations in an ET model and the efficacy of TE in ET treatment protocol by means of a vast number of TE.Methods: Study groups were categorized as patients with ET diagnosis (ET group, n:30), patients with reactive thrombocytosis secondary to iron deficiency anemia (IDA-RT) (IDA-RT group, n:30) and healthy controls (HC group, n:30). Serum levels of copper (Cu), iron (Fe), cobalt (Co), chromium (Cr), aluminum (Al), silicon (Si), nickel (Ni), zinc (Zn), selenium (Se), manganese (Mn), boron (B) and magnesium (Mg) were analyzed utilizing inductively coupled plasma-optical emission spectrophotometer instrument (ICP-OES). Statistical analysis was evaluated using SPSS 23.0.Results: ET group had statistically higher serum levels of Co and Mg (p < 0.05), Ni and Mn (p < 0.001), and lower Si (p < 0.05) than IDA-RT group. ET group had statistically higher serum levels of Co and Mn (p < 0.05), and Ni (p < 0.001), and lower Al, Si and Se (p < 0.001) than HC group. Serum levels of Fe, Al and Se (p < 0.001), and Mg (p < 0.01), and Zn (p < 0.05) in IDA-RT group were significantly lower than HC group.Conclusion: This novel study pointed out that alterations of many serum TE by means of both increment or decrement might have close relationship with mechanisms and complications of ET onset and follow-up. We consider that further research of TE would elucidate ethiopathogenesis and prognosis of ET. Thus, analysis of serum trace elements in essential thrombocytosis patients may be an important protocol by means of diagnosis, treatment and follow-up intervals

    Evaluation of trace elements in equine piroplasmosis

    Equine piroplasmosis (EP) is a tick-borne disease caused by Babesia caballi and Theileria equi, which causes a significant economic loss. The aim of this study was to determine the disease-related changes of serum trace elements that play an important role in the immune system. Our study was conducted on 20 horses with piroplasmosis and 20 healthy horses. It was performed to evaluate some of the serum trace element levels (chromium (Cr), copper (Cu), iron (Fe), magnesium (Mg) manganese (Mn), selenium (Se), zinc (Zn), arsenic (As), boron (B), and nickel (Ni)) in equine piroplasmosis. The elements were analyzed in serum using 1CP-OES. Infected horses had significantly higher serum Mg and Mn levels. In conclusion, changes of serum trace elements in infected horses can provide valuable information about pathophysiological aspects and they are also good indicators for diagnosis, prognosis and treatment evaluation. Much more scientific research is required to evaluate the trace elements related to equine piroplasmosis

    Examination of metal mobilization from a gunshot by scanning acoustic microscopy, scanning electron microscopy, energy-dispersive X-ray spectroscopy, and inductively coupled plasma optical emission spectroscopy: a case report

    Abstract Background Projectile foreign bodies are known to cause chronic heavy metal toxicity due to the release of metal into the bloodstream. However, the local effect around the metallic object has not been investigated and the main goal of our study is to examine the influence of the object in close proximity of the object. Case presentation A 36-year-old Caucasian woman with one metallic pellet close to her sciatic nerve due to a previous shotgun injury at the gluteal area presented with a diagnosis of recurrent lumbar disk herniation at L4–5 level. A physical examination confirmed chronic neuropathy and she underwent a two-stage surgery. The surgery included removal of the foreign body, followed by discectomy and fusion at the involved level. During the removal of the metallic foreign body, a tissue sample around the pellet and another tissue sample from a remote area were obtained. The samples were analyzed by scanning acoustic microscopy, scanning electron microscopy, and energy-dispersive X-ray spectroscopy. Lead, chromium, copper, cadmium, iron, manganese, selenium, and zinc elements in tissue, blood, and serum specimens were detected by inductively coupled plasma optical emission spectroscopy. Conclusions An acoustic impedance map of the tissue closer to the metallic body showed higher values indicating further accumulation of elements. Energy-dispersive X-ray spectroscopy results confirmed scanning acoustic microscopy results by measuring a higher concentration of elements closer to the metallic body. Scanning electron microscopy images showed that original structure was not disturbed far away; however, deformation of the structure existed in the tissue closer to the foreign body. Element analysis showed that element levels within blood and serum were more or less within acceptable ranges; on the other hand, element levels within the tissues showed pronounced differences indicating primarily lead intoxication in the proximity of the metallic body. We can state that residues of metallic foreign bodies of gunshot injuries cause chronic metal infiltration to the surrounding tissue and induce significant damage to nearby neural elements; this is supported by the results of scanning acoustic microscopy, scanning electron microscopy, energy-dispersive X-ray spectroscopy, and inductively coupled plasma optical emission spectroscopy

    Equine herpesvirus 1 (EHV-1) is responsible for several syndromes, such as respiratory disease, abortion, fatal viral pneumonia in neonatal foals, and a neurological syndrome, currently referred to as EHV-1 myeloencephalopathy. The analysis of trace element levels in the sera of EHV-1 infected horses could be useful in clarifying the pathogenesis or pathophysiology of these EHVinduced clinical syndromes. Previous studies have shown significant alterations for some serum trace elements (zinc, iron, copper) in EHV-1 infected horses, and this could be justified by the putative role of these elements in many immunological pathways or by their antiviral activity. The aim of the present study was to perform a comparison by retrospective serological study of 52 EHV-1 infected and non-infected horses, both healthy and ill, to establish whether there were possible alterations in serum levels of arsenic, copper, boron, zinc, iron, chromium, magnesium, manganese, selenium, and silicium. Horses were categorized based on the type of syndrome (respiratory disease, abortion, or neurological disease) and the presence of seroconversion (by virus neutralization) and the result of polymerase chain reaction (PCR) for EHV-1. Levels of serum chromium, copper, selenium, boron, and silicium were significantly different among different groups of EHV-1 infected or non-infected horses. Serum chromium levels were higher in infected horses compared to non-infected individuals (p=0.0001). Levels of serum copper (p=0.001), magnesium (p=0.05), selenium (p=0.004), and silicium (p=0.004) were significantly lower in the horses with neurological disease. While levels of serum chromium (p=0.005) were higher, those of boron (p=0.002) were significantly lower in cases of EHV1 abortion. Overall, the present study revealed alterations in the serum levels of some trace elements between EHV-1 non-infected and infected horses, such as those that aborted or developed neurological signs. However, the relationship between the trace elements and the outcomes of the infection could not be established. Further research is needed to enlighten the effects of trace element alterations on the equine herpesvirus-1 infection pathogenesis in horses
