    Adverse Effects of Antimicrobials via Predictable or Idiosyncratic Inhibition of Host Mitochondrial Components

    This minireview explores mitochondria as a site for antibiotic-host interactions that lead to pathophysiologic responses manifested as nonantibacterial side effects. Mitochondrion-based side effects are possibly related to the notion that these organelles are archaic bacterial ancestors or commandeered remnants that have co-evolved in eukaryotic cells; thus, this minireview focuses on mitochondrial damage that may be analogous to the antibacterial effects of the drugs. Special attention is devoted to aminoglycosides, chloramphenicol, and fluoroquinolones and their respective single side effects related to mitochondrial disturbances. Linezolid/oxazolidinone multisystemic toxicity is also discussed. Aminoglycosides and oxazolidinones are inhibitors of bacterial ribosomes, and some of their side effects appear to be based on direct inhibition of mitochondrial ribosomes. Chloramphenicol and fluoroquinolones target bacterial ribosomes and gyrases/topoisomerases, respectively, both of which are present in mitochondria. However, the side effects of chloramphenicol and the fluoroquinolones appear to be based on idiosyncratic damage to host mitochondria. Nonetheless, it appears that mitochondrion-associated side effects are a potential aspect of antibiotics whose targets are shared by prokaryotes and mitochondria—an important consideration for future drug design

    Sperm-mediated Effects of Predation Risk on Reproduction in Male Threespined Sticklebacks

    Threespined sticklebacks, Gasterosteus aculeatus, exhibit male-only parental care, but will decrease paternal care if exposed to predators while given care or if their mate has had prior experiences with predators. The offspring will experience lasting effects based on the experience of their parents (behavior-mediated transgenerational plasticity). While other studies have concluded that male exposure to predators prior to fertilization (sperm-mediated transgenerational plasticity) impacts offspring, whether sperm-mediated effects exist in sticklebacks, and for how many generations the effects persist is unknown. Courtship trials with both predator exposed and predator unexposed females with six male treatment groups shows significant impact of maternal grandfather predation exposure, suggesting that daughters of predator-exposed fathers are passing down cues to their sons

    Receiver Mount Design, Transmitter Depth, and Wind Speed Affect Detection Probability of Acoustic Telemetry Transmitters in a Missouri River Tributary

    Background One of the most important considerations for acoustic telemetry study designs is det:ection probability between the transmitter and the receiver. Variation in environmental (i.e., wind and flow) and abiotic (i.e., bathymetry) conditions among aquatic systems can lead to differences in detection probability temporally or between systems. In this study we evaluate the effect of distance, receiver mount design, transmitter depth, and wind speed on detection probabilities of two models of acoustic transmitters in a mid-sized river. InnovaSea V16-6H (hereafter V16) and V13-1L (hereafter V13) tags were deployed in the James River, SD at 0.36 m (deep) and 2.29 m (V16 tag) or 1.98 m (V13 tag; shallow) above the benthic surface downstream of InnovaSea VR2W stationary receivers at distances of 100, 200, or 300 m. We used two receiver mount designs that included a fixed position within a PVC pipe on the downstream side of a bridge piling or a metal frame deployed in the middle of the river channel. Tags were deployed for 72 h at each location, and hourly detections were summarized. We evaluated downstream distance, receiver mount design, tag depth, and wind effects on tag detection using Bayesian logistic regression. Results: Detection probability decreased as distance increased for all combinations of tag types and mount designs and varied from nearly 100% at 100 m to less than 10% at 300 m. The V16 transmitter had greater detection probability by the receiver mounted in the pipe than in the midriver frame. For both mounts, the deep V16 transmitter had greater detection probability than the V16 shallow transmitter. Detection probability of the V13 transmitter was similar between receiver mounts or transmitter depths. Wind speed had a negative impact on detection probabilities of both transmitter types and depths, except the deep V16 transmitter. Conclusions: Deploying acoustic receivers in PVC pipes rather than midriver frames provided greater downstream detection probabilities for V16 transmitters under conditions evaluated in this study. In addition, V16 transmitters had greater detection probabilities when positioned deep within the water column rather than near the surface.     A comparison of welfare outcomes for weaner and mature Bos indicus bulls surgically or tension ban castrated with or without analgesia: 2. responses related to stress, health and productivity

    Tension banding castration of cattle is gaining favour because it is relatively simple to perform and is promoted by retailers of the banders as a humane castration method. Two experiments were conducted, under tropical conditions using Bos indicus bulls comparing tension banding (Band) and surgical (Surgical) castration of weaner (7-10 months old) and mature (22-25 months old) bulls with and without pain management (NSAID (ketoprofen) or saline injected intramuscularly immediately prior to castration). Welfare outcomes were assessed using a range of measures; this paper reports on some physiological, morbidity and productivity-related responses to augment the behavioural responses reported in an accompanying paper. Blood samples were taken on the day of castration (day 0) at the time of restraint (0. min) and 30. min (weaners) or 40. min (mature bulls), 2. h, and 7. h; and days 1, 2, 3, 7, 14, 21 and 28 post-castration. Plasmas from day 0 were assayed for cortisol, creatine kinase, total protein and packed cell volume. Plasmas from the other samples were assayed for cortisol and haptoglobin (plus the 0. min sample). Liveweights were recorded approximately weekly to 6 weeks and at 2 and 3 months post-castration. Castration sites were checked at these same times to 2 months post-castration to score the extent of healing and presence of sepsis.     Citizen science and the United Nations Sustainable Development Goals

    Traditional data sources are not sufficient for measuring the United Nations Sustainable Development Goals. New and non-traditional sources of data are required. Citizen science is an emerging example of a non-traditional data source that is already making a contribution. In this Perspective, we present a roadmap that outlines how citizen science can be integrated into the formal Sustainable Development Goals reporting mechanisms. Success will require leadership from the United Nations, innovation from National Statistical Offices and focus from the citizen-science community to identify the indicators for which citizen science can make a real contribution
