    The Cuticular Hydrocarbons of Dasineura Oleae Show Differences Between Sex, Adult Age and Mating Status

    : In insects, cuticular lipids prevent water loss and act as semiochemicals. Because of their ecological function, the profile change across the insects' sex and development offers insight into insect biology and possible tools for pest management. Here, the first work on cecidomyiid cuticular extracts is proposed considering Dasineura oleae (Diptera: Cecidomyiidae) males and females at different adult ages (0-12 h, 12-24 h, 24-36 h) and distinct sexual conditions (virgin and mated). A set of 49 compounds were recorded (12 alkanes, 1 monomethyl alkane, 11 fatty acids, 4 esters, 1 aldehyde, 1 allylbenzene, 1 amine, 1 flavonoid, 1 ketone, 1 phenol, 1 steradiene, 1 sterol, 1 terpene, 1 triterpene and 11 unknown compounds), and 18 of them showed significant differences between groups. Among alkanes, hexacosane (nC26) exhibited a decreasing trend from the youngest to the oldest females, while pentacosane (nC25) and nonacosane (nC29) showed a decreasing trend from 0 to 12 h to 12-24 h virgin females. In addition, nonadecane (nC19) was significantly more abundant in the youngest males compared to older males and females. The alkanes nC25, nC26 and nC29 have been reported to be age-related also in other dipterans, while nC19 has been described as gender-specific chemical cue for platygastrid parasitoids. Further behavioural trials and analyses are required to assign the specific ecological roles to the characterized compounds. Our results may contribute to develop new low-impact control strategies relying on the manipulation of D. oleae's chemical communication (e.g. disruption of mating or species recognition).     High innate attractiveness to black targets in the blue blowfly, Calliphora vomitoria (L.) (Diptera: Calliphoridae)

    Calliphora vomitoria is a myiasis-causing fly in many animal species including humans. The control of blowflies is still anchored on the use of chemicals. However, mass trapping and lure-and-kill techniques represent a promising alternative to pesticides. Visual and olfactory cues are the main stimuli routing the fly's landing behavior. Notably, color attractiveness has been barely explored in flies of medical and veterinary importance, with special reference to blowflies. In this study, we investigated the innate color preferences in C. vomitoria adults, testing binary combinations of painted targets under laboratory conditions. The identity of tested species C. vomitoria was confirmed by DNA sequencing (18S and cox1 genes). C. vomitoria flies showed a significant preference for black colored targets in all tested binary color combinations, after 5, 15, 30 and 60 min of exposure. Black targets were significantly preferred over blue, red, yellow and white ones. Spectral characteristics of all tested color combinations were quantified and the innate attraction of blowflies towards black targets was discussed in relation to their behavioral ecology. To the best of our knowledge, this is the first report on innate color preferences in the Calliphora genus. Our findings can be useful to develop new, cheap and reliable monitoring traps as well as â\u80\u9clure and killâ\u80\u9d tools to control blowfly pests

    The effects of CA IX catalysis products within tumor microenvironment

    Solid tumors are composed of both cancer cells and various types of accessory cells, mainly fibroblasts, that collectively compose the so called tumor-microenvironment. Cancer-associated fibroblasts have been described to actively participate in cancer progression by establishing a cytokine-mediated as well as metabolic crosstalk with cancer cells. In the present paper we show that activated human fibroblasts are able to boost tumor cells proliferation and that this effect is greatly dependent on stromal carbonic anhydrase IX (CA IX) activity. In fact fibroblasts show a strong upregulation of CA IX expression upon activation by cancer cells, while CA IX products, protons and bicarbonate, exert differential effects on cancer cells proliferation. While acidification of extracellular pH, a typical condition of rapidly growing solid tumors, is detrimental for tumor cells proliferation, bicarbonate, through its organication, supplies cancer cells with intermediates useful to sustain their high proliferation rate. Here we propose a new kind of fibroblasts/tumor cells crosstalk within tumor microenvironment, mediated by stromal CA IX products, aimed to favor cancer cells growth, opening new perspectives on CA IX role in tumor microenvironment

    The impact of adult diet on parasitoid reproductive performance

    Diet is one of the most common influences on parasitoid reproductive traits. The life span, mating ability, fecundity, fertility and sex ratio of parasitoids can be affected by the quality of the adult diet. In the field, parasitoids can rely on different hosts and non-host nutrient sources, such as floral and extrafloral nectar, hemipteran honeydew and pollen, and various artificial diets have been used in mass rearing. In addition, some parasitoid species obtain nutrients by feeding on their host while adult (host feeding). In this review, we summarize current knowledge on the impact of the adult diet on the reproductive behavior of hymenopteran and dipteran parasitoids, with a particular focus on longevity, offspring production and host searching traits. First, we focus on food preferences and learning abilities of parasitoids to discriminate high-quality diets. Second, we analyze the impact of the adult diet on longevity, examining different natural and artificial food sources as well as the effect of their concentration and frequency. Third, we highlight the impact of the adult diet on host foraging. Fourth, we review the impact of adult diet on parasitoid offspring with special reference to (1) egg load, maturation and resorption, (2) parasitism and (3) progeny production and sex ratio. Finally, a number of implications for biological control and integrated pest management are discussed

    Thermal regime and spatial variability of the active layer in Deception Island, Antarctica

    El programa de monitoreo de capa activa (CALM) fue desarrollado en las últimas décadas con la finalidad de comprender el impacto del cambio climático sobre los ambientes con permafrost. Este trabajo analiza los resultados obtenidos, en los últimos seis años, de los sitios CALM-S Irizar, Cráter Lake y Refugio Chileno, ubicados en Isla Decepción. En ellos se ha medido el espesor, el estado térmico y la distribución espacial de la capa activa. En los sitios Irizar y Refugio Chileno la evolución del espesor de capa activa varió interanualmente sin una clara tendencia dentro del corto período analizado; por el contrario, el sitio Cráter Lake evidenció una tendencia a la disminución de espesor. La distribución espacial como el espesor de dicha capa en los tres sitios de monitoreo, mostraron estar condicionados principalmente por la potencia de la cubierta de nieve, la litología y la exposición a los vientos. El relieve, la topografía de detalle, la orientación de las laderas frente a la radiación solar incidente, ejercieron un control menor sobre esta distribución. El estado térmico de la capa activa evidenció el control ejercido sobre ella de la temperatura del aire y de la cubierta nival. Así bajo reducidos espesores de nieve, el modelo de penetración en profundidad de la isoterma de 0 ºC, ha permitido aproximar satisfactoriamente el espesor de capa activa.The Circumpolar Active Layer Monitoring (CALM) program developed over the last two decades has a leading edge in comprehensive efforts to study the impacts of climate change in permafrost environments. This paper describes results obtained during the last six years, on the active layer thickness, thermal regime and spatial patterns of thaw, at the CALM-S Irizar, Crater Lake and Refugio Chileno sites in Deception Island. In Irizar and Refugio Chileno CALM-S sites the active layer depth varied interannually without any clear trend over the short record available. By contrast, Crater Lake CALM-S showed a slight thinning trend. In all sites, the spatial patterns of thaw and active layer thickness were mainly controlled by snow cover conditions, lithology and the effect of aspect on wind. The relief, the detailed topography and the effect of aspect on solar radiation also exert a local control over its patterns. Air temperature and snow cover controlled the thermal state of active layer.     SHMT2-mediated mitochondrial serine metabolism drives 5-FU resistance by fueling nucleotide biosynthesis

    5-Fluorouracil (5-FU) is a key component of chemotherapy for colorectal cancer (CRC). 5-FU efficacy is established by intracellular levels of folate cofactors and DNA damage repair strategies. However, drug resistance still represents a major challenge. Here, we report that alterations in serine metabolism affect 5-FU sensitivity in in vitro and in vivo CRC models. In particular, 5-FU-resistant CRC cells display a strong serine dependency achieved either by upregulating endogenous serine synthesis or increasing exogenous serine uptake. Importantly, regardless of the serine feeder strategy, serine hydroxymethyltransferase-2 (SHMT2)-driven compartmentalization of one-carbon metabolism inside the mitochondria represents a specific adaptation of resistant cells to support purine biosynthesis and potentiate DNA damage response. Interfering with serine availability or affecting its mitochondrial metabolism revert 5-FU resistance. These data disclose a relevant mechanism of mitochondrial serine use supporting 5-FU resistance in CRC and provide perspectives for therapeutic approaches
