Localization of ASV Integrase-DNA Contacts by Site-Directed Crosslinking and their Structural Analysis
We applied crosslinking techniques as a first step in preparation of stable avian sarcoma virus (ASV) integrase (IN)-DNA complexes for crystallographic investigations. These results were then compared with the crystal structures of the prototype foamy virus (PFV) intasome and with published data for other retroviral IN proteins.Photoaffinity crosslinking and site-directed chemical crosslinking were used to localize the sites of contacts with DNA substrates on the surface of ASV IN. Sulfhydryl groups of cysteines engineered into ASV IN and amino-modified nucleotides in DNA substrates were used for attachment of photocrosslinkers. Analysis of photocrosslinking data revealed several specific DNA-protein contacts. To confirm contact sites, thiol-modified nucleotides were introduced into oligo-DNA substrates at suggested points of contact and chemically crosslinked to the cysteines via formation of disulfide bridges. Cysteines incorporated in positions 124 and 146 in the ASV IN core domain were shown to interact directly with host and viral portions of the Y-mer DNA substrate, respectively. Crosslinking of an R244C ASV IN derivative identified contacts at positions 11 and 12 on both strands of viral DNA. The most efficient disulfide crosslinking was observed for complexes of the ASV IN E157C and D64C derivatives with linear viral DNA substrate carrying a thiol-modified scissile phosphate.Analysis of our crosslinking results as well as published results of retroviral IN protein from other laboratories shows good agreement with the structure of PFV IN and derived ASV, HIV, and MuLV models for the core domain, but only partial agreement for the N- and C-terminal domains. These differences might be explained by structural variations and evolutionary selection for residues at alternate positions to perform analogous functions, and by methodological differences: i.e., a static picture of a particular assembly from crystallography vs. a variety of interactions that might occur during formation of functional IN complexes in solution
External Causes in the Structure of Premature Mortality in the Republic of Sakha (Yakutia)
The purpose of this survey was to study the rate, trends and patterns of mortality from external causes in the Republic of Sakha (Yakutia) [RS(Y)]. A retrospective analysis of statistical data on mortality rate (MR) of the population in Yakutia in the period between 1990 and 2017 was carried out.
The results obtained suggest that the population mortality pattern in Yakutia consists of a high proportion of losses from external causes in young age groups. Suicides dominate the structure of external causes of death in Yakutia (20.7% in 2017), followed by homicides (13.6%), cold temperature exposures (11.4%) and road traffic accidents (8.7%). The annual loss of the working-age population in RS(Y) makes up over 40% in the total number of deaths. In the structure of the causes of mortality of the working-age population during the entire study period, accidents, poisonings and injuries ranked first, followed by circulatory system diseases and neoplasms. Thus, the main trends that have a negative impact on the creation of the workforce system in RS(Y) are as follows: the high MR of the working-age population of both sexes; the overmortality of working-age men; the prevalence of preventable causes of death of an exogenous nature (accidents, poisonings, injuries); and an increased share of retirement-age people, resulting in an increased demographic load on the able-bodied population
Widespread continental mtDNA lineages prevail in the bumblebee fauna of Iceland
Origins of the fauna in Iceland is controversial, although the majority of modern research supports the postglacial colonization of this island by terrestrial invertebrates rather than their long-term survival in glacial refugia. In this study, we use three bumblebee species as a model to test the hypothesis regarding possible cryptic refugia in Iceland and to evaluate a putative origin of recently introduced taxa. Bombus jonellus is thought to be a possible native Icelandic lineage, whereas B. lucorum and B. hortorum were evidently introduced in the second half of the 20th century. These phylogeographic analyses reveal that the Icelandic Bombus jonellus shares two COI lineages, one of which also occurs in populations on the British Isles and in mainland Europe, but a second lineage (BJ-02) has not been recorded anywhere. These results indicate that this species may have colonized Iceland two times and that the lineage BJ-02 may reflect a more ancient Late Pleistocene or Early Holocene founder event (e.g., from the British Isles). The Icelandic populations of both Bombus lucorum and B. hortorum share the COI lineages that were recorded as widespread throughout Eurasia, from the European countries across Russia to China and Japan. The findings presented here highlight that the bumblebee fauna of Iceland comprises mainly widespread ubiquitous lineages that arrived via natural or human-mediated dispersal events from the British Isles or the mainland

A nearly complete database on the records and ecology of the rarest boreal tiger moth from 1840s to 2020
Global environmental changes may cause dramatic insect declines but over century-long time series of certain species’ records are rarely available for scientific research. The Menetries’ Tiger Moth (Arctia menetriesii) appears to be the most enigmatic example among boreal insects. Although it occurs throughout the entire Eurasian taiga biome, it is so rare that less than 100 specimens were recorded since its original description in 1846. Here, we present the database, which contains nearly all available information on the species’ records collected from 1840s to 2020. The data on A. menetriesii records (N = 78) through geographic regions, environments, and different timeframes are compiled and unified. The database may serve as the basis for a wide array of future research such as the distribution modeling and predictions of range shifts under climate changes. It represents a unique example of a more than century-long dataset of distributional, ecological, and phenological data designed for an exceptionally rare but widespread boreal insect, which primarily occurs in hard-to-reach, uninhabited areas of Eurasia.Peer reviewe
Crowdsourcing Fungal Biodiversity : Revision of Inaturalist Observations in Northwestern Siberia
The paper presents the first analysis of crowdsourcing data of all observations of fungi (including lichens) and myxomycetes in Northwestern Siberia uploaded to iNaturalist.org to date (24.02.2022). The Introduction presents an analysis of fungal diversity crowdsourcing globally, in Russia, and in the region of interest. Materials and methods describe the protocol of uploading data to iNaturalist.org, the structure of the crowdsourcing community. initiative to revise the accumulated data. procedures of data analysis, and compilation of a dataset of revised crowdsourced data. The Results present the analysis of accumulated data by several parameters: temporal, geographical and taxonomical scope, observation and identification efforts, identifiability of various taxa, species novelty and Red Data Book categories and the protection status of registered observations. The Discussion provides data on usability of crowdsourcing data for biodiversity research and conservation of fungi, including pros and contras. The Electronic Supplements to the paper include an annotated checklist of observations of protected species with information on Red Data Book categories and the protection status, and an annotated checklist of regional records of new taxa. The paper is supplemented with a dataset of about 15 000 revised and annotated records available through Global Biodiversity Information Facility (GBIF). The tradition of crowdsourcing is rooted in mycological societies around the world, including Russia. In Northwestern Siberia, a regional mycological club was established in 2018, encouraging its members to contribute observations of fungi on iNaturalist.org. A total of about 15 000 observations of fungi and myxomycetes were uploaded so far, by about 200 observers, from three administrative regions (Yamalo-Nenetsky Autonomous Okrug, Khanty-Mansi Autonomous Okrug, and Tyumen Region). The geographical coverage of crowdsourcing observations remains low. However. the observation activity has increased in the last four years. The goal of this study consisted of a collaborative effort of professional mycologists invited to help with the identification of these observations and analysis of the accumulated data. As a result, all observations were reviewed by at least one expert. About half of all the observations have been identified reliably to the species level and received Research Grade status. Of those, 90 species (195 records) represented records of taxa new to their respective regions: 876 records of 53 species of protected species provide important data for conservation programmes. The other half of the observations consists of records still under-identified for various reasons: poor quality photographs, complex taxa (impossible to identify without microscopic or molecular study). or lack of experts in a particular taxonomic group. The Discussion section summarises the pros and cons of the use of crowdsourcing for the study and conservation of regional fungal diversity, and summarises the dispute on this subject among mycologists. Further research initiatives involving crowdsourcing data must focus on an increase in the quality of observations and strive to introduce the habit of collecting voucher specimens among the community of amateurs. The timely feedback from experts is also important to provide quality and the increase of personal involvement.Peer reviewe
Bioerosion of siliceous rocks driven by rock-boring freshwater insects
Macrobioerosion of mineral substrates in fresh water is a little-known geological process. Two examples of rock-boring bivalve molluscs were recently described from freshwater environments. To the best of our knowledge, rock-boring freshwater insects were previously unknown. Here, we report on the discovery of insect larvae boring into submerged siltstone (aleurolite) rocks in tropical Asia. These larvae belong to a new mayfly species and perform their borings using enlarged mandibles. Their traces represent a horizontally oriented, tunnel-like macroboring with two apertures. To date, only three rock-boring animals are known to occur in fresh water globally: a mayfly, a piddock, and a shipworm. All the three species originated within primarily wood-boring clades, indicating a simplified evolutionary shift from wood to hardground substrate based on a set of morphological and anatomical preadaptations evolved in wood borers (e.g., massive larval mandibular tusks in mayflies and specific body, shell, and muscle structure in bivalves)
