    Potentiation of raloxifene cytotoxicity against MCF-7 breast cancer cell lines via transdermal delivery and loading on self-emulsifying nanoemulsions

    Purpose: To enhance raloxifene (RLX) delivery and cytotoxicity against breast cancer (MCF-7) cell lines. Methods: This was a solubility study of RLX in different oils, surfactants, and co-surfactants. Twelve formulae were tested to reach the smallest  globular size, and hydroxypropyl methylcellulose, (HPMC), and Carbopol 947 polymers were tested for formation of transdermal films. The formula  with the lowest size was compared with raw RLX in diffusion studies using a Franz diffusion cell. Finally, a cytotoxicity study against MCF-7 breast cancer cell lines was conducted. Results: The maximum solubility of RLX was in Tween 80, peppermint oil, and PEG 200; therefore, these were the main components of the 12 formulations. The release of RLX loaded on the selfnanoemulsion drug delivery system (SNEDDS) was increased 3-fold compared with raw RLX.Cytotoxicity results revealed that RLX SNEDDs decreased MCF-7 cell survival by approximately 40 %, compared with raw RLX (control), which augmented the RLX suppression of breast cancer cell lines. Conclusion: Improvement in RLX cytotoxicity is a novel strategy to suppress breast cancer. Keywords: Raloxifene, Osteoporosis, Bioavailability, Nanoemulsion, Nanoparticle

    Study of simulations of double graded InGaN solar cell structures

    The performances of various configurations of InGaN solar cells are compared using nextnano software. Here we compare a flat base graded wall GaN/InGaN structure, with an InxGa1-xN well with sharp GaN contact layers, and an InxGa1-xN structure with InxGa1-xN contact layers, i.e. a homojunction. The doping in the graded structures are the result of polarization doping at each edge (10 nm from each side) due to the graded structure, while the well structures are intentionally doped at each edge (10 nm from each side) equal to the doping concentration in the graded structure. The solar cells are characterized by their open-circuit voltage, V_oc, short circuit current, I_sc, solar efficiency, and energy band diagram. The results indicate that an increase in I_sc and efficiency results from increasing both the fixed and the maximum indium compositions, while the V_oc decreases.     Microwaves effectively examine the extent and type of coking over acid zeolite catalysts

    Coking leads to the deactivation of solid acid catalyst. This phenomenon is a ubiquitous problem in the modern petrochemical and energy transformation industries. Here, we show a method based on microwave cavity perturbation analysis for an effective examination of both the amount and the chemical composition of cokes formed over acid zeolite catalysts. The employed microwave cavity can rapidly and non-intrusively measure the catalytically coked zeolites with sample full body penetration. The overall coke amount is reflected by the obtained dielectric loss (ε″) value, where different coke compositions lead to dramatically different absorption efficiencies (ε″/cokes’ wt%). The deeper-dehydrogenated coke compounds (e.g., polyaromatics) lead to an apparently higher ε″/wt% value thus can be effectively separated from lightly coked compounds. The measurement is based on the nature of coke formation during catalytic reactions, from saturated status (e.g., aliphatic) to graphitized status (e.g., polyaromatics), with more delocalized electrons obtained for enhanced Maxwell–Wagner polarization

    Wax: A benign hydrogen-storage material that rapidly releases H2-rich gases through microwave-assisted catalytic decomposition

    Hydrogen is often described as the fuel of the future, especially for application in hydrogen powered fuel-cell vehicles (HFCV’s). However, its widespread implementation in this role has been thwarted by the lack of a lightweight, safe, on-board hydrogen storage material. Here we show that benign, readily-available hydrocarbon wax is capable of rapidly releasing large amounts of hydrogen through microwave-assisted catalytic decomposition. This discovery offers a new material and system for safe and efficient hydrogen storage and could facilitate its application in a HFCV. Importantly, hydrogen storage materials made of wax can be manufactured through completely sustainable processes utilizing biomass or other renewable feedstocks

    Optimized D-α-tocopherol polyethylene glycol succinate/phospholipid self-assembled mixed micelles: A promising lipid-based nanoplatform for augmenting the antifungal activity of fluconazole

    Fluconazole (FLZ) is the most widely used antifungal agent for treating cutaneous candidiasis. Although oral FLZ has been proved to be effective, the incidence of side effects necessitates the development of an effective formulation that could surpass the pitfalls associated with systemic availability. Accordingly, this research aimed at developing a self-assembled mixed micelles topical delivery system to enhance the topical delivery of the drug. Self-assembled mixed micelles were developed using D-α-tocopheryl polyethylene glycol 1000 succinate and phospholipids and optimized using Box-Behnken design. The optimized formulation with minimized size was then tested in vivo for the antifungal activity against C. albicans in immunocompromised mice. Treatment with the optimized formulation led to decreased peripheral erythema as well as lesions due to fungal infection in comparison to raw FLZ loaded gel. Therefore, the developed formulation was found to be a promising vehicle for the treatment of cutaneous candidiasis

    Anticonvulsant and Neuroprotective Activities of Phragmanthera austroarabica

    Anticonvulsant and neuroprotective activity of Phragmanthera austroarabica extract were tested in pentylenetetrazole-kindled mice. All the chemical constituents of the plant extract were identified. Additionally, the extract was standardized and proved to contain total phenolic contents equal to 379.92±1.32 mg gallic acid equivalents/g dry plant extract. Induction of kindling was achieved by repeated intraperitoneal administration of pentylenetetrazole (35 mg/kg) twice weekly. Male albino mice were given P. austroarabica extract (200, 400, or 800 mg/kg). The two higher doses (400 or 800 mg/kg) of the extract significantly caused notable reduction in seizure activity and hippocampal malondialdehyde level compared to pentylenetetrazole control group. The highest dose enhanced cortical GSH level and showed intact DNA in the laddering assay. Upon studying the neuroprotective effect, mice treated with the higher dose of the extract demonstrated an improvement in the percent of surviving neurons in the cortex and hippocampus. We concluded that P. austroarabica extract ameliorated seizure activity and protected cortical and hippocampal neurons against pentylenetetrazole-induced kindling in mice

    PLC Virtualization and Software Defined Architectures in Industrial Control Systems

    Today’s automation systems are going through a transition called Industry 4.0, referring to the Fourth Industrial Revolution. New concepts, such as cyber-physical systems, mi-croservices and Smart Factory are introduced. This brings up the question of how some of these new technologies can be utilized in Industrial Control Systems. Machines and production lines are nowadays controlled by hardware PLCs and this is considered as a state-of-the-art solution. However, the market demands are continuously increasing and pushing the industry e.g. to lower the operational costs and to develop more agile solutions. Industry 4.0 provides promising approaches to take a step forward and consider PLC virtualization. The purpose of this thesis was to evaluate PLC virtualization possibilities using different Software Defined Architectures. Requirements and benefits of different solutions were evaluated. The major objective of the case study was to compare container- and hypervisor-based virtualization solutions using Docker and KVM. The case study provides a modular and scalable IIoT solution in which a virtual PLC takes over the control instead of a hardware PLC. Node-RED was used as a runtime environment and an I/O-module was needed to set up a control loop test. Response time of the control loop was measured by capturing Modbus traffic with tcpdump. Multiple iterations were performed to show minimum, maximum, average, median and 90th pctl. latencies. The results indicate that the container-based solution has a smaller overhead than the hypervisor-based solution and it has a very little overhead in general. Peak latencies are a concern and even the average latencies show that this solution would not be suitable for any hard real-time or safety-related applications. Further investigation on the topic would be needed to estimate the actual potential of PLC virtualization on hard real-time applications. First of all, a more powerful hardware PC would be needed to perform such tests. Secondly, a faster industrial protocol than Modbus TCP/IP would be required. Perhaps another kind of approach would be needed to overcome the issues that were experienced in this case study. It would be interesting to test a direct communication between virtual PLC and I/O and use Node-RED nodes for example to trigger inputs. Anyhow, it seems that container-based solution is holding much promise as a virtualization approach

    Improvement of solubility, dissolution, and bioavailability of phenytoin intercalated in Mg-Al layered double hydroxide

    Layered double hydroxides (LDHs) are highly effective drug delivery systems, owing to their capacity to intercalate or adsorb biomaterials, flexible structure, swelling property, high stability, good biocompatibility, and ease of synthesis. Phenytoin (PHT) is an antiseizure BCS (Biopharmaceutics Classification System) class II drug, presenting low aqueous solubility. Therefore, the current study aimed at increasing its solubility, dissolution, and bioavailability. PHT was intercalated to the MgAl-LDH formed in situ and successful intercalation to form MgAl-PHT-LDH was confirmed by FTIR, PXRD, DSC, and TGA. Examination of particle size and morphology (by photon correlation spectroscopy and electron microscopy, respectively) confirmed the formation and intercalation of nanostructured LDH. Intercalation enhanced the saturation solubility of PHT at 25°C in 0.1N HCl and phosphate buffer (pH 6.8) by 6.57 and 10.5 times respectively. The selected drug excipient powder blend for the formulation of MgAl-PHT-LDH tablets exhibited satisfactory properties in both pre-compression parameters (angle of repose, bulk density, tapped density, Carr’s index, and Hausner ratio) and tablet characteristics (weight variation, thickness, hardness, friability, content uniformity, and disintegration time). MgAl-PHT-LDH tablets showed better dissolution of PHT compared to unprocessed PHT tablets at all time points. Oral bioavailability of MgAl-PHT-LDH tablets and unprocessed PHT tablets was tested in two groups of Sprague Dawley rats based on analysis of serum levels of both forms of PHT by UPLC-ESI-MS/MS serum. MgAl-PHT-LDH tablets demonstrated a relative bioavailability of 130.15% compared to unprocessed PHT tablets, confirming a significantly higher oral bioavailability of MgAl-PHT-LDH. In conclusion, MgAl-PHT-LDH could provide a strategy for enhancing solubility, dissolution, and thereby bioavailability of PHT, enabling the evaluation of theclinical efficacy of MgAl-PHT-LDH tablets for the treatment of seizures at lower PHT doses

    Knowledge of community pharmacists in Saudi Arabia regarding human monkeypox, its management, prevention and vaccination : findings and implications

    Many cases of monkeypox have recently been reported in countries where this disease is not endemic raising a global health concern. Consequently, healthcare professionals (HCPs) including pharmacists need to be aware of the disease, its prevention, including the role of vaccines, and its management to reduce transmission. A cross-sectional, questionnaire-based study was con-ducted among conveniently sampled community pharmacists in Qassim region, Saudi Arabia. 189 community pharmacists participated in the study giving a response rate of 72.97%. From these, 86.77% were male, 51.32% were ≤ 30 years old, 36.51% were aged between 31-40 years and 43.39% had 1-5 years of experience as community pharmacists. Their overall knowledge was 17.72 ± 5.56 out of a maximum of 28. The overall rate of correct answers for the knowledge statements was 63.29%, with 52.4% answering the knowledge questions correctly ≥ 50
