    Acute but not chronic metabolic acidosis potentiates the acetylcholine-induced reduction in blood pressure: an endothelium-dependent effect

    Metabolic acidosis has profound effects on vascular tone. This study investigated the in vivo effects of acute metabolic acidosis (AMA) and chronic metabolic acidosis (CMA) on hemodynamic parameters and endothelial function. CMA was induced by ad libitum intake of 1% NH4Cl for 7 days, and AMA was induced by a 3-h infusion of 6 M NH4Cl (1 mL/kg, diluted 1:10). Phenylephrine (Phe) and acetylcholine (Ach) dose-response curves were performed by venous infusion with simultaneous venous and arterial blood pressure monitoring. Plasma nitrite/nitrate (NOx) was measured by chemiluminescence. The CMA group had a blood pH of 7.15±0.03, which was associated with reduced bicarbonate (13.8±0.98 mmol/L) and no change in the partial pressure of arterial carbon dioxide (PaCO2). The AMA group had a pH of 7.20±0.01, which was associated with decreases in bicarbonate (10.8±0.54 mmol/L) and PaCO2 (47.8±2.54 to 23.2±0.74 mmHg) and accompanied by hyperventilation. Phe or ACh infusion did not affect arterial or venous blood pressure in the CMA group. However, the ACh infusion decreased the arterial blood pressure (ΔBP: -28.0±2.35 mm Hg [AMA] to -4.5±2.89 mmHg [control]) in the AMA group. Plasma NOx was normal after CMA but increased after AMA (25.3±0.88 to 31.3±0.54 μM). These results indicate that AMA, but not CMA, potentiated the Ach-induced decrease in blood pressure and led to an increase in plasma NOx, reinforcing the effect of pH imbalance on vascular tone and blood pressure control

    Anti-inflammatory and physicochemical characterization of the croton rhamnifolioides essential oil inclusion complex in β-cyclodextrin

    Croton rhamnifolioides is used in popular medicine for the treatment of inflammatory diseases. The objective of this study was to characterize and evaluate the anti-inflammatory effect of C. rhamnifolioides essential oil complexed in \u3b2-cyclodextrin (COEFC). The physicochemical characterization of the complexes was performed using different physical methods. The anti-inflammatory activity was evaluated in vivo by ear edema, paw edema, cotton pellet-induced granuloma, and vascular permeability by Evans blue extravasation. The mechanism of action was validated by molecular docking of the major constituent into the cyclooxygenase-2 (COX-2 enzyme). All doses of the COEFC reduced acute paw edema induced by carrageenan and dextran, as well as vascular permeability. Our results suggest the lowest effective dose of all samples inhibited the response induced by histamine or arachidonic acid as well as the granuloma formation. The complexation process showed that the pharmacological effects were maintained, however, showing similar results using much lower doses. The results demonstrated an involvement of the inhibition of pathways dependent on eicosanoids and histamine. Complexation of \u3b2-cyclodextrin/Essential oil (\u3b2-CD/EO) may present an important tool in the study of new compounds for the development of anti-inflammatory drugs

    Com o objetivo de avaliar os efeitos da substituição do farelo de soja pela mistura raspa de mandioca+uréia sobre o desempenho, o consumo e a digestibilidade dos nutrientes em dietas à base de palma forrageira (Opuntia ficus indica, Mill) e silagem de sorgo, oito vacas mestiças Holandês:Gir (60&plusmn;30 dias em lactação e 491 kg de PV inicial) foram distribuídas em dois quadrados latinos (4 &times; 4). A mistura raspa de mandioca+uréia substituiu 0, 33, 67 e 100% do farelo de soja nas dietas. Os consumos de matéria seca (expressos em kg/dia, % do PV, g/kg0,75 do PV), MO, PB, EE, FDN e CHOT diminuíram linearmente com a inclusão da raspa de mandioca. As ingestões de FDN (%PV), FDA, CNF e NDT não foram afetadas pelos níveis de raspa de mandioca+uréia nas dietas. A inclusão de raspa de mandioca+uréia influenciou a digestibilidade aparente de todos os nutrientes, sendo descrita por função quadrática. A produção de leite, a produção de leite corrigida para 4,0% de gordura e a produção de gordura diminuíram linearmente em 30, 20 e 0,8 g/dia, respectivamente, enquanto o teor de gordura e a eficiência alimentar, em kg de leite corrigido para 4,0% de gordura/kg de MS, não foram influenciadas pelos níveis de substituição do farelo de soja na dieta.<br>This trial was conducted to investigate the replacement of soybean meal with a mixture of urea plus cassava scrapings on nutrient intake, digestibility, and production of crossbred lactating cows fed diets containing forage cactus (Opuntia ficus indica, Mill), sorghum silage, and concentrate. Eight crossbred holstein:zebu cows averaging 60&plusmn;30 days in milk and initial body weight (BW) of 491 kg were randomly assigned to two replicated 4 &times; 4 Latin squares and were fed diets with increasing levels of urea plus cassava scrapings: 0, 33, 67 or 100%. Replacing soybean meal with urea plus cassava scrapings linearly decreased the intakes of DM (kg, % BW and g/BW kg0.75), OM, CP, EE, NDF, and TC. However, the increasing dietary levels of urea plus cassava sacrapings did not affetc the intakes of NDF (% BW), ADF, NFC, and TDN. Apparent digestibilities of all nutrients showed significant quadratic effects with increasing urea plus cassava scrapings. Milk yield, milk yield corrected for 4.0% fat and milk fat yield all decreased linearly by 30, 20 and, 0.8 g/day, respectively, while fat content and feed efficiency, expressed as kg of 4.0% fat corrected milk/kg of DM, were not changed by decreasing soybean meal in the diet

    Abstract: Paca (Cuniculus paca), one of the largest rodents of the Brazilian fauna, has inherent characteristics of its species which can conribute as a new option for animal experimantation. As there is a growing demand for suitable experimental models in audiologic and otologic surgical research, the gross anatomy and ultrastructural ear of this rodent have been analyzed and described in detail. Fifteen adult pacas from the Wild Animals Sector herd of Faculdade de Ciências Agrárias e Veterinárias, Unesp-Jaboticabal, were used in this study. After anesthesia and euthanasia, we evaluated the entire composition of the external ear, registering and ddescribing the details; the temporal region was often dissected for a better view and detailing of the tympanic bulla which was removed and opened to expose the ear structures analyzed mascroscopically and ultrastructurally. The ear pinna has a triangular and concave shape with irregular ridges and sharp apex. The external auditory canal is winding in its path to the tympanic mebrane. The tympanic bulla is is on the back-bottom of the skull. The middle ear is formed by a cavity region filled with bone and membranous structures bounded by the tympanic membrane and the oval and round windows. The tympanic membrane is flat and seals the ear canal. The anatomy of the paca ear is similar to the guinea pig and from the viewpoint of experimental model has major advantages compared with the mouse ear

    Objetivou-se, com este experimento, avaliar o efeito da substituição do milho por palma forrageira sobre o desempenho de vacas mestiças em lactação e a digestibilidade dos nutrientes. Foram utilizadas oito vacas mestiças (5/8 HZ), distribuídas em dois quadrados latinos 4 x 4 em esquema fatorial 2 x 2 (duas cultivares de palma forrageira, com ou sem milho). Não houve interação entre palma e milho e o efeito dos dois alimentos foi analisado de forma isolada. Os consumos de matéria seca, de fibra em detergente ácido, dematéria orgânica, de proteína bruta, de extrato etéreo e de carboidratos totais não foram influenciados pelas cultivares de palma utilizadas. Animais que receberam palma gigante nas dietas apresentaram maior consumo de fibra em detergente neutro. As dietas com milho proporcionaram maior consumo de matéria seca (kg/dia e % de PV). O consumo de NDT, os coeficientes de digestibilidade de todos os nutrientes, a produção de leite corrigida, o teor de gordura e a eficiência alimentar não foram influenciados pelas cultivares de palma forrageira ou pela inclusão milho.<br>The experiment was conduced to evaluate the effects of corn replacement for forage cactus on the performance of crossbreed lactating cows and nutrient digestibility. Eight cows were assigned a two latin square design in a factorial 2 x 2 arrangement (two forage cactus cultivars, with or without corn).There was not interaction between forage cactus and corn and the effect of both were analyzed in an isolated way. The intake of dry matter, organic matter, acid detergent fiber, crude protein, ether extract and total carbohydrates were not affected by the forage cactus. Animals that received giant cultivar presented larger neutral detergent fiber intake. The diets with corn presented larger dry matter intake (kg/day and % of LW). The intake of TDN, the coefficients of digestibility of all nutrients, the fat corrected milk production, fat content and kg of milk: dry matter intake ratio were not affected not by the forage cactus cultivars neither by the inclusion of corn
