    Bioavailability of orange juice (poly)phenols: the impact of short-term cessation of training by male endurance athletes

    Background: Physical exercise has been reported to increase the bioavailability of citrus flavanones. Objective: To investigate the bioavailability of orange juice (OJ) (poly)phenols in endurance-trained men before and after cessation of training for 7 days. Design: Ten fit endurance-trained males, with a maximal oxygen consumption of 58.2 ± 5.3 mL/kg/min, followed a low (poly)phenol diet for 2 d before drinking 500 mL of OJ, containing 398 µmol of (poly)phenols of which 330 µmol were flavanones. After the volunteers stopped training for 7 days the feeding study was repeated. Urine samples were collected 12 h pre- and 24 h post-OJ orange consumption. Bioavailability was assessed by the quantitative analysis of urinary flavanone metabolites and (poly)phenol catabolites using HPLC-HR-MS. Results: While training, 0-24 h urinary excretion of flavanone metabolites, mainly hesperetin-3-O-glucuronide, hesperetin-3´-sulfate, naringenin-4´-O-glucuronide, naringenin-7-O-glucuronide, was equivalent to 4.2% of OJ flavanone intake. This increased significantly to 5.2% when OJ was consumed after the volunteers stopped training for 7 days. Overall, this trend, although not significant, was also observed with OJ-derived colonic catabolites which after supplementation in the trained state were excreted in amounts equivalent to 51% of intake compared to 59% after cessation of training. However, urinary excretion of three colonic catabolites of bacterial origin, most notably, 3-(3´-hydroxy-4´-methoxyphenyl)hydracrylic acid, did increase significantly when OJ was consumed post- compared to pre-cessation of training. Data were also obtained on inter-individual variations in flavanone bioavailability. Conclusion: A 7-day cessation of endurance training enhanced, rather than reduced, the bioavailability of OJ flavanones. The biological significance of these differences and, whether or not they extend to the bioavailability of other dietary (poly)phenols, remains to be determined. Hesperetin-3´-O-glucuronide and the colonic microbiota-derived catabolite 3-(3´-hydroxy-4´-methoxyphenyl)hydracrylic acid are key biomarkers of the consumption of hesperetin-O-glycoside-containing OJ and other citrus products

    A computational method for recording and analysis of mandibular movements

    This study proposed the development of a new clinical tool capable of quantifying the movements of opening-closing, protrusion and laterotrusion of the mandible. These movements are important for the clinical evaluation of the temporomandibular function and muscles involved in mastication. Unlike current commercial systems, the proposed system employs a low-cost video camera and a computer program that is used for reconstructing the trajectory of a reflective marker that is fixed on the mandible. In order to illustrate the clinical application of this tool, a clinical experiment consisting on the evaluation of the mandibular movements of 12 subjects was conducted. The results of this study were compatible with those found in the literature with the advantage of using a low cost, simple, non-invasive, and flexible tool customized for the needs of the practical clinic

    Análise comparativa dos ficocolóides produzidos por algas carragenófitas usadas industrialmente e algas carragenófitas portuguesas

    As Carragenanas (E-407) constituem um dos principais aditivos usados pela indústria alimentar, como agentes gelificantes, emulsionantes, estabilizantes e espessantes. São ingredientes naturais, usados há várias décadas na área alimentar e são considerados como seguros, tendo obtido a classificação GRAS (Generally Recognised As Safe). Neste trabalho foram analisadas a percentagem de peso seco e a composição química (por FTIR e FT-Raman) das carragenanas produzidas pelas carragenófitas (Gigartinales, Rhodophyta) oriundas de diversos países: Kappaphycus alvarezzi (Tanzânia, Indonésia, Filipinas); Kappaphycus striatum (Madagáscar); Eucheuma denticulatum (Tanzânia, Filipinas e Madagáscar); Betaphycus gelatinum (Filipinas); e Sarcothalia crispata (Chile). Para comparação foram também analisadas algas carragenófitas da costa portuguesa (Gigartinales, Rhodophyta): Chondrus crispus, Mastocarpus stellatus; Gigartina pistillata; Chondracanthus teedei var. lusitanicus; Chondracanthus acicularis; Calliblepharis jubata; Gymnogongrus crenulatus; e Ahnfeltiopsis devoniensis. Os principais resultados a reter são: a maior percentagem de peso seco em carragenanas foi obtida a partir das algas carragenófitas Kappaphycus alvarezzi (Filipinas), com 84.4% e Gigartina pistillata (Praia do Norte, Viana do Castelo), com 65.4%. Relativamente à natureza dos ficocolóides produzidos pelas carragenófitas estudadas, as nossas análises espectroscópicas permitiram determinar a presença de um leque diversificado de carragenanas: carragenana iota pura; carragenana kappa quase pura; uma série de carragenanas híbridas kappa-iota, com diferentes rácios iota/kappa; carragenanas kappa-beta, xi-tetha e xi-lambda

    Identification of plasma and urinary metabolites and catabolites derived from orange juice (poly)phenols: analysis by high-performance liquid chromatography–high-resolution mass spectrometry

    Orange juice is a rich source of (poly)phenols, in particular, the flavanones hesperetin-7-O-rutinoside and naringenin-7-O-rutinoside. Following the acute consumption of 500 mL of orange juice containing 398 μmol of (poly)phenols by 12 volunteers, 0–24 h plasma and urine samples were analyzed by targeted high-performance liquid chromatography–high-resolution mass spectrometry in order to identify flavanone metabolites and phenolic acid and aromatic catabolites. A total of 19 flavanone metabolites—comprising di-O-glucuronide, O-glucuronide, O-glucuronyl-sulfate, and sulfate derivatives of hesperetin, naringenin, and eriodictyol—and 65 microbial-derived phenolic catabolites, such as phenylpropanoid, phenylpropionic, phenylacetic, benzoic, and hydroxycarboxylic acids and benzenetriol and benzoylglycine derivatives, including free phenolics and phase II sulfate, glucuronide, and methyl metabolites, were identified or partially identified in plasma and/or urine samples. The data obtained provide a detailed evaluation of the fate of orange juice (poly)phenols as they pass through the gastrointestinal tract and are absorbed into the circulatory system prior to renal excretion.     Biological and anthropogenic predictors of metal concentration in the Eurasian otter, a sentinel of freshwater ecosystems

    Toxic metals have been linked to a range of adverse health effects in freshwater organisms. However, for higher vertebrates, there is little understanding of the large-scale drivers of exposure. We quantified toxic metal/semi-metal concentrations in a sentinel freshwater top predator, the Eurasian otter (Lutra lutra), across England and Wales, and determined how this varied with key natural and anthropogenic factors. We related liver concentrations in 278 otters that died between 2006 and 2017 to habitat biogeochemistry, proximity to point source contamination and to biological characteristics (length, sex, condition). Evidence for any positive association with putative anthropogenic sources (mining, human population, known discharges) was weak or lacking in nearly all cases, with the exception of a positive association between lead and human population density. Despite concerns that burgeoning use of nanosilver in consumer products might increase silver concentrations in waste waters, there was no increase over time. Spatial variation in soil/sediment pH, precipitation, and soil calcium oxide are indicated as significant predictors of metal concentrations in otters (higher cadmium and silver in areas with lower pH and higher rainfall, and higher chromium and lead in areas of lower calcium oxide). Liver chromium and nickel concentrations declined significantly over time (Cr 0.030 ± 1.2 to 0.015 ± 1.3 μg/g dry weight, Ni 0.0038 ± 1.2 to 0.00068 ± 1.5 μg/g, between 2006–2009 and 2014–2017), but other metals showed no temporal change. Biotic associations were important, with age related accumulation indicated for mercury and cadmium (as well as interactions with body condition). Our results suggest that larger-scale geochemical and hydrological processes are important in determining metal exposure in otters, and we provide an indication of risk factors that may be of relevance for freshwater vertebrates in other countries with well-developed water pollution management

    Prevalência de brucelose bovina no Estado da Paraíba

    A serosurvey was conducted to estimate the prevalence of bovine brucellosis in the State of Paraíba, Brazil. A random sample of six municipalities was selected from each of three ecoregions of the State: the Agreste/Litoral region (1), the Cariri/Curimatau region (2) and the Sertão region (3). These 18 municipalities were divided in quadrants and one herd was selected at random from each quadrant. The sampling scheme included herds with more than 50 bovines, which excludes small-holder subsistence livestock production. At least 32 animals were sampled within each herd, stratified in four age categories, as follows: (1) at least 8 animals under 6 months of age, (2) at least 8 animals of 7 to 18 months of age, (3) at least 8 animals of 19 to 30 months of age and (4) at least 8 animals over 30 months of age. Antibodies to Brucella abortus were observed in 8 out of 18 municipalities (44.44%),in 8 out of 72 herds included in this study (11.11%) and in 8 animals out of 2343 samples collected (0.34%). The prevalence of test positive cattle by ecoregion/cattle production regions in the state showed no significant differences of risk among them. Likewise, there was no significant difference of risk among the different age categories studied. It was observed that brucellosis is widely spread across the state albeit with low animal prevalence. These results may be used in the design of a strategy for controlling bovine brucellosis in the State of Paraíba.Foi realizado um estudo para a determinação da prevalência sorológica de brucelose bovina no Estado da Paraíba. Seis municípios foram selecionados aleatoriamente em cada uma das três ecorregiões do Estado: a região do Agreste/Litoral (1), a região do Cariri/Curimatau (2) e a região do Sertão (3). Estes 18 municípios foram divididos em quadrantes e uma propriedade foi selecionada em cada quadrante. Foram amostradas propriedades com mais de 50 animais, o que exclui as pequenas propriedades de subsistência. Pelo menos 32 animais foram amostrados de cada rebanho, estratificados nas quatro faixas etárias seguintes: (1) no mínimo 8 animais até 6 meses de idade, (2) no mínimo 8 animais com 7 a 18 meses de idade, (3) no mínimo 8 animais com 19 a 30 meses de idade e (4) no mínimo 8 animais com mais de 30 meses de idade. Anticorpos anti-B. abortus foram observados em 8 de 18 municípios (44,44%), em 8 de 72 rebanhos (11,11%) e em 8 animais dos 2343 amostrados (0,34%). A análise dos resultados de prevalência por ecorregião do Estado não revelou diferença significativa de risco entre elas. A análise dos resultados por categorias de faixa etária também não evidenciaram diferença significativa de risco entre as mesmas. Foi observado que a brucelose está amplamente disseminada no estado embora com uma baixa prevalência animal. Estes resultados podem ser úteis no delineamento de estratégias de controle da brucelose bovina no estado da Paraíba

    New insights into the bioavailability of red raspberry anthocyanins and ellagitannins

    Red raspberries, containing ellagitannins and cyanidin-based anthocyanins, were fed to volunteers and metabolites appearing in plasma and urine were analysed by UHPLC-MS. Anthocyanins were not absorbed to any extent with sub nmol/L concentrations of cyanidin-3-O-glucoside and a cyanidin-O-glucuronide appearing transiently in plasma. Anthocyanins excreted in urine corresponded to 0.007% of intake. More substantial amounts of phase II metabolites of ferulic acid and isoferulic acid, along with 4′-hydroxyhippuric acid, potentially originating from pH-mediated degradation of cyanidin in the proximal gastrointestinal tract, appeared in urine and also plasma where peak concentrations were attained 1–1.5 h after raspberry intake. Excretion of 18 anthocyanin-derived metabolites corresponded to 15.0% of intake, a figure substantially higher than obtained in other anthocyanin feeding studies. Ellagitannins pass from the small to the large intestine where the colonic microbiota mediate their conversion to urolithins A and B which appeared in plasma and were excreted almost exclusively as sulfate and glucuronide metabolites. The urolithin metabolites persisted in the circulatory system and were excreted in urine for much longer periods of time than the anthocyanin metabolites although their overall urinary recovery was lower at 7.0% of intake. It is events originating in the proximal and distal gastrointestinal tract, and subsequent phase II metabolism, that play an important role in the bioavailability of both anthocyanins and ellagitannins and it is their metabolites which appear in the circulatory system, that are key to elucidating the mode of action(s) underlying the protective effects of these compounds on human health

    Erectile dysfunction is frequent in systemic sclerosis and associated with severe disease: a study of the EULAR Scleroderma Trial and Research group

    Introduction: Erectile dysfunction (ED) is common in men with systemic sclerosis (SSc) but the demographics, risk factors and treatment coverage for ED are not well known. Method: This study was carried out prospectively in the multinational EULAR Scleroderma Trial and Research database by amending the electronic data-entry system with the International Index of Erectile Function-5 and items related to ED risk factors and treatment. Centres participating in this EULAR Scleroderma Trial and Research substudy were asked to recruit patients consecutively. Results: Of the 130 men studied, only 23 (17.7%) had a normal International Index of Erectile Function-5 score. Thirty-eight per cent of all participants had severe ED (International Index of Erectile Function-5 score ≤ 7). Men with ED were significantly older than subjects without ED (54.8 years vs. 43.3 years, P < 0.001) and more frequently had simultaneous non-SSc-related risk factors such as alcohol consumption. In 82% of SSc patients, the onset of ED was after the manifestation of the first non-Raynaud's symptom (median delay 4.1 years). ED was associated with severe cutaneous, muscular or renal involvement of SSc, elevated pulmonary pressures and restrictive lung disease. ED was treated in only 27.8% of men. The most common treatment was sildenafil, whose efficacy is not established in ED of SSc patients. Conclusions: Severe ED is a common and early problem in men with SSc. Physicians should address modifiable risk factors actively. More research into the pathophysiology, longitudinal development, treatment and psychosocial impact of ED is needed

    Expression profiling and cross-species RNA interference (RNAi) of desiccation-induced transcripts in the anhydrobiotic nematode Aphelenchus avenae.

    BACKGROUND: Some organisms can survive extreme desiccation by entering a state of suspended animation known as anhydrobiosis. The free-living mycophagous nematode Aphelenchus avenae can be induced to enter anhydrobiosis by pre-exposure to moderate reductions in relative humidity (RH) prior to extreme desiccation. This preconditioning phase is thought to allow modification of the transcriptome by activation of genes required for desiccation tolerance. RESULTS: To identify such genes, a panel of expressed sequence tags (ESTs) enriched for sequences upregulated in A. avenae during preconditioning was created. A subset of 30 genes with significant matches in databases, together with a number of apparently novel sequences, were chosen for further study. Several of the recognisable genes are associated with water stress, encoding, for example, two new hydrophilic proteins related to the late embryogenesis abundant (LEA) protein family. Expression studies confirmed EST panel members to be upregulated by evaporative water loss, and the majority of genes was also induced by osmotic stress and cold, but rather fewer by heat. We attempted to use RNA interference (RNAi) to demonstrate the importance of this gene set for anhydrobiosis, but found A. avenae to be recalcitrant with the techniques used. Instead, therefore, we developed a cross-species RNAi procedure using A. avenae sequences in another anhydrobiotic nematode, Panagrolaimus superbus, which is amenable to gene silencing. Of 20 A. avenae ESTs screened, a significant reduction in survival of desiccation in treated P. superbus populations was observed with two sequences, one of which was novel, while the other encoded a glutathione peroxidase. To confirm a role for glutathione peroxidases in anhydrobiosis, RNAi with cognate sequences from P. superbus was performed and was also shown to reduce desiccation tolerance in this species. CONCLUSIONS: This study has identified and characterised the expression profiles of members of the anhydrobiotic gene set in A. avenae.
