    Insulin-like and IGF-like peptides in the silkmoth Bombyx mori: discovery, structure, secretion, and function

    A quarter of a century has passed since bombyxin, the first insulin-like peptide identified in insects, was discovered in the silkmoth Bombyx mori. During these years, bombyxin has been studied for its structure, genes, distribution, hemolymph titers, secretion control, as well as physiological functions, thereby stimulating a wide range of studies on insulin-like peptides in other insects. Moreover, recent studies have identified a new class of insulin family peptides, IGF-like peptides, in B. mori and Drosophila melanogaster, broadening the base of the research area of the insulin-related peptides in insects. In this review, we describe the achievements of the studies on insulin-like and IGF-like peptides mainly in B. mori with short histories of their discovery. Our emphasis is that bombyxins, secreted by the brain neurosecretory cells, regulate nutrient-dependent growth and metabolism, whereas the IGF-like peptides, secreted by the fat body and other peripheral tissues, regulate stage-dependent growth of tissues

    Discovery of Multiple High-Velocity Narrow Circumstellar Na I D Lines in Nova V1280 Sco

    We discovered multiple high-velocity (ranging from -900 to -650 km/s) and narrow (FWHM = 15 km/s) absorption components corresponding to both the D2 and the D1 lines of Na I on a high dispersion spectrum of V1280 Sco observed on 2009 May 9 (UT), 814 d after the V-band maximum. Subsequent observations carried out on 2009 June and July confirmed at least 11 distinct absorption components in both systems. Some components had deepened during the two months period while their HWHMs and wavelengths remained nearly constant. We suggest these high velocity components originate in cool clumpy gas clouds moving on the line of sight, produced in interactions between pre-existing cool circumstellar gas and high velocity gas ejected in the nova explosion. The optical region spectrum of V1280 Sco in 2009 is dominated by the continuum radiation and exhibits no forbidden line characterizing the nebular phase of typical novae. Permitted Fe II lines show doubly peaked emission profiles and some strong Fe II lines are accompanied by a blue shifted (about -255 km/s) absorption component. However, no high-velocity and narrow components corresponding to those of Na I could be detected in Fe II lines nor in the Balmer lines. The 255 km/s low velocity absorption component is most probably originating in the wind from the nova.Comment: Accepted for PASJ letter, 5 pages, 8 figure

    Organoammonium salts of dipeptide-derived chiral tri-amines or diamines with TfOH catalyzed the enantioselective 1,3-dipolar cycloaddition reactions of alpha-acyloxyacroleins with nitrones to give the corresponding adducts in good yields (up to 96%) and with high diastereo- and enantioselectivities (up to 89% ee). Although alpha-(p-methoxybenzoyloxy)acrolein is rather unstable under the reaction conditions, alpha-(3-pyrroline-1-carbonyloxy)acrolein is stable enough to be smoothly converted into the corresponding adducts with the aid of the chiral organoammonium salt catalysts

    Thioureas bearing electron-deficient aryl groups show high catalytic activity in the biomimetic bromocyclization of geranyl derivatives. The reaction of geranyl derivatives with N-bromosuccinimide (NBS) proceeds rapidly in CH2Cl2 to give the corresponding bromocyclization products in high yields as a ca. 1:1 mixture of endo- and exo-isomers. The reactivity of geranyl derivatives highly depends on the terminal substituent: electron-donating substituents increase the reactivity, while electron-withdrawing substituents decrease the reactivity

    Annulative coupling of vinylboronic esters: aryne-triggered 1,2-metallate rearrangement

    A stereoselective annulative coupling of a vinylboronic ester ate-complex with arynes producing cyclic borinic esters has been developed. An annulation reaction that proceeded through the formation of two C-C bonds and a C-B bond was realized by exploiting a 1,2-metallate rearrangement of boronate triggered by the addition of a vinyl group to the strained triple bond of an aryne. The generated aryl anion would then cyclize to a boron atom to complete the annulation cascade. The annulated borinic ester could be converted to boronic acids and their derivatives by oxidation, halogenation, and cross-coupling. Particularly, halogenation and Suzuki-Miyaura coupling proceeded in a site-selective fashion and produced highly substituted alkylboronic acid derivatives

    Bombyxin (Bombyx Insulin-Like Peptide) Increases the Respiration Rate Through Facilitation of Carbohydrate Catabolism in Bombyx mori

    Bombyxin-II, an insulin-like peptide of the silkmoth Bombyx mori, has been shown to reduce both the trehalose concentration in the hemolymph and the glycogen content in some tissues of B. mori larvae. However, little is known about how these storage carbohydrates are utilized. To address this question, the effects of bombyxin-II injection into Bombyx larvae on the tissue lipid level, respiration rate, and glycolytic activity of tissues were investigated. Bombyxin-II did not affect lipid accumulation in the hemolymph and fat body, while it increased the rate of oxygen consumption and increased the content of fructose 2, 6-bisphosphate, a potent activator of glycolysis, in the gonads, imaginal discs, and midgut. These results suggest that bombyxin facilitates cellular energy production thereby supporting the tissue growth of insects

    A Fat Body-Derived IGF-like Peptide Regulates Postfeeding Growth in Drosophila

    SummaryMembers of the insulin family of peptides have conserved roles in the regulation of growth and metabolism in a wide variety of metazoans. Here we show that Drosophila insulin-like peptide 6 (DILP6), which is structurally similar to vertebrate insulin-like growth factor (IGF), is predominantly expressed in the fat body, a functional equivalent of the vertebrate liver and adipocytes. This expression occurs during the postfeeding stage under the direct regulation of ecdysteroid. We further reveal that dilp6 mutants show growth defects during the postfeeding stage, which results in reduced adult body size through a decrease in cell number. This phenotype is rescued by fat body-specific expression of dilp6. These data indicate that DILP6 is a functional, as well as a structural, counterpart of vertebrate IGFs. Our data provide in vivo evidence for a role of ILPs in determining adult body size through the regulation of postfeeding growth

    D=5 Simple Supergravity on AdS_2 \times S^3

    The Kaluza-Klein spectrum of D=5 simple supergravity compactified on S^3 is studied. A classical background solution which preserves maximal supersymmetry is fulfilled by the geometry of AdS_2\times S^3.
