    Cell cycle regulation and novel structural features of thymidine kinase, an essential enzyme in Trypanosoma brucei

    Thymidine kinase (TK) is a key enzyme in the pyrimidine salvage pathway which catalyzes the transfer of the γ-phosphate of ATP to 2′-deoxythymidine (dThd) forming thymidine monophosphate (dTMP). Unlike other type II TKs, the Trypanosoma brucei enzyme (TbTK) is a tandem protein with two TK homolog domains of which only the C-terminal one is active. In this study, we establish that TbTK is essential for parasite viability and cell cycle progression, independently of extracellular pyrimidine concentrations. We show that expression of TbTK is cell cycle regulated and that depletion of TbTK leads to strongly diminished dTTP pools and DNA damage indicating intracellular dThd to be an essential intermediate metabolite for the synthesis of thymine-derived nucleotides. In addition, we report the X-ray structure of the catalytically active domain of TbTK in complex with dThd and dTMP at resolutions up to 2.2 Å. In spite of the high conservation of the active site residues, the structures reveal a widened active site cavity near the nucleobase moiety compared to the human enzyme. Our findings strongly support TbTK as a crucial enzyme in dTTP homeostasis and identify structural differences within the active site that could be exploited in the process of rational drug design

    Power management and control strategies for off-grid hybrid power systems with renewable energies and storage

The DGS consists of Photovoltaic (PV) panels as Renewable Power Source (RPS), a Diesel Generator (DG) for power buck-up and a BESS to accommodate the surplus of energy, which may be employed in times of poor PV generation. While off-grid DGS represent an efficient and cost-effective energy supply solution particularly to rural and remote areas, fluctuations in voltage and frequency due to load variations, weather conditions (temperature, irradiation) and transmission line short-circuits are major challenges. The paper suggests a hierarchical Power Management (PM) and controller structure to improve the reliability and efficiency of the hybrid DGS. The first layer of the overall control scheme includes a Fuzzy Logic Controller (FLC) to adjust the voltage and frequency at the Point of Common Coupling (PCC) and a Clamping Bridge Circuit (CBC) which regulates the DC bus voltage. A maximum power point tracking (MPPT) controller based on FLC is designed to extract the optimum power from the PV. The second control layer coordinates among PV, DG and BESS to ensure reliable and efficient power supply to the load. MATLAB Simulink is used to implement the overall model of the off-grid DGS and to test the performance of the proposed control scheme which is evaluated in a series of simulations scenarios.     IoT Based Smart City Bus Stops

    The advent of smart sensors, single system-on-chip computing devices, Internet of Things (IoT), and cloud computing is facilitating the design and development of smart devices and services. These include smart meters, smart street lightings, smart gas stations, smart parking lots, and smart bus stops. Countries in the Gulf region have hot and humid weather around 6–7 months of the year, which might lead to uncomfortable conditions for public commuters. Transportation authorities have made some major enhancements to existing bus stops by installing air-conditioning units, but without any remote monitoring and control features. This paper proposes a smart IoT-based environmentally - friendly enhanced design for existing bus stop services in the United Arab Emirates. The objective of the proposed design was to optimize energy consumption through estimating bus stop occupancy, remotely monitor air conditioning and lights, automatically report utility breakdowns, and measure the air pollution around the area. In order to accomplish this, bus stops will be equipped with a WiFi-Based standalone microcontroller connected to sensors and actuators. The microcontroller transmits the sensor readings to a real-time database hosted in the cloud and incorporates a mobile app that notifies operators or maintenance personnel in the case of abnormal readings or breakdowns. The mobile app encompasses a map interface enabling operators to remotely monitor the conditions of bus stops such as the temperature, humidity, estimated occupancy, and air pollution levels.     Digital Twin Conceptual Model within the Context of Internet of Things

    As the Internet of Things (IoT) is gaining ground and becoming increasingly popular in smart city applications such as smart energy, smart buildings, smart factories, smart transportation, smart farming, and smart healthcare, the digital twin concept is evolving as complementary to its counter physical part. While an object is on the move, its operational and surrounding environmental parameters are collected by an edge computing device for local decision. A virtual replica of such object (digital twin) is based in the cloud computing platform and hosts the real-time physical object data, 2D and 3D models, historical data, and bill of materials (BOM) for further processing, analytics, and visualization. This paper proposes an end-to-end digital twin conceptual model that represents its complementary physical object from the ground to the cloud. The paper presents the proposed digital twin model’s multi-layers, namely, physical, communication, virtual space, data analytic and visualization, and application as well as the overlapping security layer. The hardware and software technologies that are used in building such a model will be explained in detail. A use case will be presented to show how the layers collect, exchange, and process the physical object data from the ground to the cloud

    Data-Driven Safe Deliveries: The Synergy of IoT and Machine Learning in Shared Mobility

    Shared mobility is one of the smart city applications in which traditional individually owned vehicles are transformed into shared and distributed ownership. Ensuring the safety of both drivers and riders is a fundamental requirement in shared mobility. This work aims to design and implement an adequate framework for shared mobility within the context of a smart city. The characteristics of shared mobility are identified, leading to the proposal of an effective solution for real-time data collection, tracking, and automated decisions focusing on safety. Driver and rider safety is considered by identifying dangerous driving behaviors and the prompt response to accidents. Furthermore, a trip log is recorded to identify the reasons behind the accident. A prototype implementation is presented to validate the proposed framework for a delivery service using motorbikes. The results demonstrate the scalability of the proposed design and the integration of the overall system to enhance the rider’s safety using machine learning techniques. The machine learning approach identifies dangerous driving behaviors with an accuracy of 91.59% using the decision tree approach when compared against the support vector machine and K-nearest neighbor approaches

    Fog Computing Approach for Shared Mobility in Smart Cities

    Smart transportation a smart city application where traditional individual models are transforming to shared and distributed ownership. These models are used to serve commuters for inter- and intra-city travel. However, short-range urban transportation services within campuses, residential compounds, and public parks are not explored to their full capacity compared to the distributed vehicle model. This paper aims to explore and design an adequate framework for battery-operated shared mobility within a large community for short-range travel. This work identifies the characteristics of the shared mobility for battery-operated vehicles and accordingly proposes an adequate solution that deals with real-time data collection, tracking, and automated decisions. Furthermore, given the requirement for real-time decisions with low latency for critical requests, the paper deploys the proposed framework within the 3-tier computing model, namely edge, fog, and cloud tiers. The solution design considers the power consumption requirement at the edge by offloading the computational requests to the fog tier and utilizing the LoRaWAN communication technology. A prototype implementation is presented to validate the proposed framework for a university campus using e-bikes. The results show the scalability of the proposed design and the achievement of low latency for requests that require real-time decisions

    Dietary Supplements Use among Athletes in Lebanon: Knowledge, Attitudes, Practices, and Correlates

    Athletes are under the utmost pressure to reach excellence in their performance and achieve the desired outcomes in competitions, prompting them to use dietary supplements. Given the threats to both health and eligibility, it is crucial to observe the prevalence, sources of information, knowledge, attitudes, and practices (KAPs) among Lebanese athletes practicing their sports for at least two years. In the present paper, a cross-sectional study is performed using the snowball sampling method, in which a self-administered KAP questionnaire is used to collect data from 455 athletes (mean age: 27.4 ± 7.9 years; men: 73.1%) participating in four sports categories (ball games, combat sports, endurance sports, and weightlifting). Among the Lebanese athletes, the prevalence of dietary supplement (DS) use was 74%, where half of them had predominately used sports supplements. Athletes in Lebanon heavily rely on coaches (74%) and online sources, including webpages and social media (64%), as key information sources for DSs. The findings suggest that significant proportions of athletes show knowledge deficits and unsatisfactory attitudes towards multiple aspects related to supplementation. Moreover, education and sports type modulate the use of DSs among athletes. Furthermore, 34% reported using supplements without a recommendation from specialists, and 69% admitted to not reading the supplement’s nutrition facts. This study urges the need for the regulation of concerned authorities and education programs to help overcome the existing challenges

    Fintech, base of the pyramid entrepreneurs and social value creation

    Abstract Purpose: Fintech is having a profound impact in Sub-Saharan Africa (SSA) because it offers more financial inclusion. In this paper, the authors examine the interrelationship of Fintech, base of the pyramid (BOP) entrepreneurs and social value creation, particularly in the SSA context. Design/methodology/approach: The current paper uses a qualitative research design with open-ended, in-depth interviews as the main data sources. The authors interviewed respondents from the Sierra Leone Fintech Association and four BOP entrepreneurs operating in different sectors. Findings: The authors find that Fintech services, specifically mobile money, play a significant role in reducing uncertainty surrounding business operations. FinTech also offers growth possibilities for BOP entrepreneurs and creates social value by providing transactional security, convenience and reducing physical cash robberies. At the same time, Fintech contributes to social value by enhancing BOP entrepreneurs as well as consumers’ skills development. Research limitations/implications: This study highlights the importance of context-specific theorization when analyzing the interlinkage between BOP entrepreneurship, social value creation and Fintech. For example, the possibility of safety from a street robbery may not appear to be part of social value creation by a technological development like Fintech. However, in a country like Sierra Leone, which has experienced both a civil war and Ebola outbreak, insecurity has been one of the biggest concerns expressed by BOP inhabitants. Hence, scholars need to incorporate contextual elements of risk, uncertainty and volatility while theorizing on Fintech’s application in BOP contexts. Practical implications: A key managerial implication relates to micro-firm entrepreneurs and information specific benefits. Fintech offers entrepreneurs the possibility to be in regular contact with customers and evaluate their purchasing patterns as well as emergent needs. Fintech offers BOP entrepreneurs a possibility to further develop their technological skills as learning to use such apps can be used as a basis for further skills development. From a policy perspective, our study highlights the importance of regulating Fintech charges so that the affordability is increased, which is expected to result in significantly more BOP entrepreneurs using these services. Social implications: The authors find that at the same time, Fintech contributes to social value by enhancing skills development of BOP consumers who interact with case firms. Originality/value: This paper is one of the first studies that specifically focuses on BOP entrepreneurship and social value creation by Fintech services in an SSA context. It is also one of the few studies that incorporates views from both entrepreneurs and the country’s Fintech association, rather than focusing solely on either entrepreneurs or Fintech firms.
