    Análise hematológica por hemograma, creatinina, triglicérides e colesterol em ratos diabéticos portadores de infecção pulpar e/ou periodontal

    The aim of this study was to evaluate the influence of pulpal and/or periodontal infections associated with diabetes in hematological analysis for blood count, creatinine, triglycerides and cholesterol. We used 80 male Wistar rats (Rattus norvegicus Albinus, Wistar) that were divided into 8 groups of 10 animals: G1 - control rats (normoglycemic); G2 - rats with pulpal disease, G3 - rats with periodontal disease; G4 - rats with pulpal and periodontal disease; G5 - diabetic rats; G6 - diabetic rats with pulpal disease; G7 - diabetic rats with periodontal disease; G8 - diabetic rats with pulpal and periodontal disease. Diabetes was induced by application of streptozotocin in penile vein. The pulpal infection was induced by dental pulp exposure of the upper right first molar to the oral environment. The periodontal infection was induced by a ligature around the neck of the upper left second molar. After 30 days, the animals were sedated and 5ml of blood was collected by cardiac puncture for blood count, creatinine, triglycerides and cholesterol analyzes. The results showed an increase in mean corpuscular volume of red blood cells in diabetic rats, increased leukocyte counts, especially neutrophils and lymphocytes, serum creatinine and triglyceride levels in diabetic rats with induced oral infections. It is demonstrated that the association of the pulpal and periodontal infections with diabetes can alter blood parameters of rats, as mean corpuscular volume, leukocytes, neutrophils, lymphocytes, creatinine and triglyceridesO objetivo deste trabalho foi avaliar a influência das infecções pulpar e/ou periodontal associadas com a diabetes na análise hematológica por hemograma, creatinina, triglicérides e colesterol. Foram utilizados 80 ratos machos da linhagem Wistar (Rattus norvegicus albinus, Wistar) divididos em 8 grupos de 10 animais: G1 - ratos controle (normoglicêmicos); G2 - ratos com infecção pulpar; G3 - ratos com infecção periodontal; G4 - ratos com infecção pulpar e periodontal; G5 - ratos diabéticos; G6 - ratos diabéticos com infecção pulpar; G7 - ratos diabéticos com infecção periodontal; G8 - ratos diabéticos com infecção pulpar e periodontal. A diabetes foi induzida por meio de uma aplicação de estreptozotocina via veia peniana. A infecção pulpar foi induzida por meio da exposição pulpar do primeiro molar superior direito ao meio oral. A infecção periodontal foi induzida por meio da confecção de uma amarria junto ao colo dentário do segundo molar superior esquerdo. Após 30 dias, os animais foram sedados e por meio de uma punção cardíaca foi coletado 5ml de sangue para as análises de hemograma, creatinina, triglicérides e colesterol. Os resultados mostraram aumento no volume corpuscular médio das hemácias para ratos diabéticos, aumento na contagem de leucócitos, principalmente neutrófilos e linfócitos, aumento nos níveis séricos de creatinina e aumento nos níveis de triglicérides para os ratos diabéticos com as infecções orais induzidas. Fica demonstrado que a infecção pulpar e periodontal associadas são capazes de alterar os parâmetros sanguíneos de ratos diabéticos, como volume corpuscular médio, leucócitos, neutrófilos, linfócitos, creatinina e triglicéride

    The Number of Bleaching Sessions Influences Pulp Tissue Damage in Rat Teeth

    Introduction: Hydrogen peroxide tooth bleaching is claimed to cause alterations in dental tissue structures. This study investigated the influence of the number of bleaching sessions on pulp tissue in rats. Methods: Male Wistar rats were studied in 5 groups (groups 1S-5S) of 10 each, which differed by the number (1-5) of bleaching sessions. In each session, the animals were anesthetized, and 35% hydrogen peroxide gel was applied to 3 upper right molars. Two days after the experimental period, the animals were killed, and their jaws were processed for light microscope evaluation. Pulp tissue reactions were scored as follows: 1, no or few inflammatory cells and no reaction; 2, <25 cells and a mild reaction; 3, between 25 and 125 cells and a moderate reaction; and 4, 125 or more cells and a severe reaction. Results from each experimental group were compared between groups and within groups to the corresponding unbleached upper left molars and analyzed for significant differences using the Kruskal-Wallis test (P < .05). Results: All tissue sections showed significant bleaching-induced changes in the dental pulp. After 1 bleaching session, necrotic tissue in the pulp horns and underlying inflammatory changes were observed. The extent and intensity of these changes increased with the number of bleaching sessions. After 5 sessions, the changes included necrotic areas in the pulp tissue involving the second third of the radicular pulp and intense inflammation in the apical third. Conclusions: The number of bleaching sessions directly influenced the extent of pulp damage. © 2013 American Association of Endodontists

    Restauração transcirúrgica como alternativa na resolução das perfurações endodônticas

    The endodontic perforations are iatrogenic communications that result in a communication between the endodonto and periodontium. This occurrence represents about 10 % of cas- es of failure in endodontic treatment. The management may vary and is related to the drilling site endodontic. This paper aims to report a case of iatrogenic perforation and one of the possible alternatives for the treatment of endodontic perforations -level cervical tooth. During the opening coronary perforation occurred due to improper slope cervical penetration drill. Considering the drilling site, we opted for sealing com- posite resin after surgical access and absolute isolation. After sealing, the channel was filled with medication and instru- mented based on calcium hydroxide. Later the canal was obturated by lateral condensation technique with Sealapex. The radiographic follow-up showed regression of the lesion and the clinical silence confirmed the success of the treatment.As perfurações endodônticas são comunicações iatrogênicas que resultam em uma comunicação entre o endodonto e o periodonto. Esta ocorrência representa cerca de 10% dos casos de insucessos nos tratamentos endodônticos. A con- duta terapêutica pode variar e está relacionada com o local da perfuração endodôntica. O presente trabalho se propõe a relatar um caso de perfuração iatrogênica e uma das alternativas possíveis para o tratamento das perfurações endodônticas em nível de colo dentário. Durante a abertura coronária ocorreu perfuração cervical devido à incorreta inclinação de penetração da broca. Considerando o local da perfuração, optou-se pelo selamento com resina composta, após acesso cirúrgico e isolamento absoluto. Após o sela- mento, o canal foi instrumentado e preenchido com medicação à base de hidróxido de cálcio. Posteriormente o canal foi obturado pela técnica da condensação lateral ativa com cimento Sealapex. O acompanhamento radiográfico mostrou regressão da lesão e o silêncio clínico confirmou o su- cesso do tratamento

    Pulpal and periodontal diseases increase triglyceride levels in diabetic rats

    Objectives: The aim of this study was to evaluate triglyceride and cholesterol levels in diabetic rats and their relationship with pulpal and periodontal diseases. Methods: Eighty male rats (Rattus norvegicus albinus, Wistar) were divided into the following eight groups comprising ten animals each: normal rats (G1), rats with pulpal diseases (G2), rats with periodontal diseases (G3), rats with both pulpal and periodontal diseases (G4), diabetic rats (G5), diabetic rats with pulpal diseases (G6), diabetic rats with periodontal diseases (G7), and diabetic rats with both periodontal and pulpal diseases (G8). Diabetes was induced by injecting streptozotocin, periapical lesions were induced by exposing pulpal tissue to the oral environment, and periodontal diseases were induced by periodontal ligature. The animals were killed after 30 days, and lipid profile was enzymatically measured using Trinder's method. The total assessed values were statistically analyzed by analysis of variance and Tukey test (p < 0.05). Results: The triglyceride levels of diabetic rats with periodontal disease and of diabetic rats with both periodontal and pulpal diseases were significantly higher than those of normal rats and nondiabetic group rats, respectively. The differences in the cholesterol levels among the groups were not significant. Conclusions: We found that the association of pulpal and periodontal diseases with diabetes increased triglyceride levels in rats. Clinical significance: Changes in lipid profile may be related to the presence of oral infections and diabetes. © 2012 Springer-Verlag Berlin Heidelberg

    Effect of oral infections on serum creatinine levels in diabetic rats

Methods: Eighty male rats (Rattus norvegicus albinus, Wistar) were divided into the following 8 groups compris-ing 10 animals each: normal (G1), with pulpal disease (G2), with periodontal disease (G3), with both pulpal and periodontal disease (G4), diabetic (G5), diabetic with pulpal disease (G6), diabetic with periodontal disease (G7), and diabetic with both pulpal and periodontal disease (G8). Diabetes was induced by injecting streptozotocin, pul-pal disease were induced by exposing pulpal tissue to the oral environment, and periodontal disease was induced by periodontal ligature. After 30 days, blood was collected by cardiac puncture and the animals were killed. The maxillae were processed for histopathology. Serum creatinine levels were measured by the enzymatic method. The total assessed values were statistically analyzed by analysis of variance and Tukey’s test (p 0.05). Conclusions: We found that the serum creatinine level was higher in diabetic rats and may be related to the pres-ence of oral infections. Clinical significance: Changes in serum creatinine level may be related to the presence of oral infections and diabetes

    Biocompatibility and biomineralization assessment of a new root canal sealer and root-end filling material

    Objective: This study investigated the short-term subcutaneous tissue reaction and biomineralization ability of two epoxy-based root canal sealers containing calcium hydroxide (MBP and MBPc) and ProRoot MTA. Materials and methods: Polyethylene tubes containing the materials were implanted into the dorsal connective tissue of Wistar rats (n = 52) for 7 or 30 days; empty implanted tubes served as controls. Specimens were stained with hematoxylin-eosin and von Kossa stain or left unstained for observation under polarized light. Qualitative and quantitative evaluations of all tissue reactions were performed. One-way anova and the Kruskal-Wallis test were used for statistical analysis (P < 0.05). Results: No significant differences were observed among the groups. All three materials induced mild-to-moderate tissue reactions at 7 days, which decreased over time. Dystrophic mineralization and birefringent structures were observed only in the ProRoot MTA ® group. Conclusion: Both MBP and MBPc appear to be biocompatible but do not stimulate biomineralization. © 2012 John Wiley & Sons A/S
