    A model for the assessment of bluetongue virus serotype 1 persistence in Spain

    Bluetongue virus (BTV) is an arbovirus of ruminants that has been circulating in Europe continuously for more than two decades and has become endemic in some countries such as Spain. Spain is ideal for BTV epidemiological studies since BTV outbreaks from different sources and serotypes have occurred continuously there since 2000; BTV-1 has been reported there from 2007 to 2017. Here we develop a model for BTV-1 endemic scenario to estimate the risk of an area becoming endemic, as well as to identify the most influential factors for BTV-1 persistence. We created abundance maps at 1-km2 spatial resolution for the main vectors in Spain, Culicoides imicola and Obsoletus and Pulicaris complexes, by combining environmental satellite data with occurrence models and a random forest machine learning algorithm. The endemic model included vector abundance and host-related variables (farm density). The three most relevant variables in the endemic model were the abundance of C. imicola and Obsoletus complex and density of goat farms (AUC 0.86); this model suggests that BTV-1 is more likely to become endemic in central and southwestern regions of Spain. It only requires host- and vector-related variables to identify areas at greater risk of becoming endemic for bluetongue. Our results highlight the importance of suitable Culicoides spp. prediction maps for bluetongue epidemiological studies and decision-making about control and eradication measures

    A study of the composition of the Obsoletus complex and genetic diversity of Culicoides obsoletus populations in Spain

    Background: The Culicoides obsoletus species complex (henceforth ‘Obsoletus complex’) is implicated in the transmission of several arboviruses that can cause severe disease in livestock, such as bluetongue, African horse sickness, epizootic hemorrhagic disease and Schmallenberg disease. Thus, this study aimed to increase our knowledge of the composition and genetic diversity of the Obsoletus complex by partial sequencing of the cytochrome c oxidase I (cox1) gene in poorly studied areas of Spain. Methods: A study of C. obsoletus populations was carried out using a single-tube multiplex polymerase chain reaction (PCR) assay that was designed to differentiate the Obsoletus complex sibling species Culicoides obsoletus and Culicoides scoticus, based on the partial amplification of the cox1 gene, as well as cox1 georeferenced sequences from Spain available at GenBank. We sampled 117 insects of the Obsoletus complex from six locations and used a total of 238 sequences of C. obsoletus (ss) individuals (sampled here, and from GenBank) from 14 sites in mainland Spain, the Balearic Islands and the Canary Islands for genetic diversity and phylogenetic analyses. Results: We identified 90 C. obsoletus (ss), 19 Culicoides scoticus and five Culicoides montanus midges from the six collection sites sampled, and found that the genetic diversity of C. obsoletus (ss) were higher in mainland Spain than in the Canary Islands. The multiplex PCR had limitations in terms of specificity, and no cryptic species within the Obsoletus complex were identified. Conclusions: Within the Obsoletus complex, C. obsoletus (ss) was the predominant species in the analyzed sites of mainland Spain. Information about the species composition of the Obsoletus complex could be of relevance for future epidemiological studies when specific aspects of the vector competence and capacity of each species have been identified. Our results indicate that the intraspecific divergence is higher in C. obsoletus (ss) northern populations, and demonstrate the isolation of C. obsoletus (ss) populations of the Canary Islands.     Identifying spanish areas at more risk of monthly BTV transmission with a basic reproduction number approach

    Bluetongue virus (BTV) causes a disease that is endemic in Spain and its two major biological vector species, C. imicola and the Obsoletus complex species, differ greatly in their ecology and distribution. Understanding the seasonality of BTV transmission in risk areas is key to improving surveillance and control programs, as well as to better understand the pathogen transmission networks between wildlife and livestock. Here, monthly risk transmission maps were generated using risk categories based on well-known BTV R0 equations and predicted abundances of the two most relevant vectors in Spain. Previously, Culicoides spp. predicted abundances in mainland Spain and the Balearic Islands were obtained using remote sensing data and random forest machine learning algorithm. Risk transmission maps were externally assessed with the estimated date of infection of BTV-1 and BTV-4 historical outbreaks. Our results highlight the differences in risk transmission during April-October, June-August being the period with higher R0 values. Likewise, a natural barrier has been identified between northern and central-southern areas at risk that may hamper BTV spread between them. Our results can be relevant to implement risk-based interventions for the prevention, control and surveillance of BTV and other diseases shared between livestock and wildlife host populations

    Environment, vector, or host? Using machine learning to untangle the mechanisms driving arbovirus outbreaks

    Climatic, landscape, and host features are critical components in shaping out-breaks of vector-borne diseases. However, the relationship between the outbreaks of vector-borne pathogens and their environmental drivers is typically complicated, nonlinear, and mayvary by taxonomic units below the species level (e.g., strain or serotype). Here, we aim tountangle how these complex forces shape the risk of outbreaks of Bluetongue virus (BTV); avector-borne pathogen that is continuously emerging and re-emerging across Europe, with sev-ere economic implications. We tested if the ecological predictors of BTV outbreak risk wereserotype-specific by examining the most prevalent serotypes recorded in Europe (1, 4, and 8).We used a robust machine learning (ML) pipeline and 23 relevant environmental features to fitpredictive models to 24,245 outbreaks reported in 25 European countries between 2000 and2019. Our ML models demonstrated high predictive performance for all BTV serotypes (accu-racies>0.87) and revealed strong nonlinear relationships between BTV outbreak risk andenvironmental and host features. Serotype-specific analysis suggests, however, that each of themajor serotypes (1, 4, and 8) had a unique outbreak risk profile. For example, temperature andmidge abundance were as the most important characteristics shaping serotype 1, whereas forserotype 4 goat density and temperature were more important. We were also able to identifystrong interactive effects between environmental and host characteristics that were also sero-type specific. Our ML pipeline was able to reveal more in-depth insights into the complex epi-demiology of BTVs and can guide policymakers in intervention strategies to help reduce theeconomic implications and social cost of this important pathogen

    Integrated molecular signaling involving mitochondrial dysfunction and alteration of cell metabolism induced by tyrosine kinase inhibitors in cancer

    Cancer cells have unlimited replicative potential, insensitivity to growth-inhibitory signals, evasion of apoptosis, cellular stress, and sustained angiogenesis, invasiveness and metastatic potential. Cancer cells adequately adapt cell metabolism and integrate several intracellular and redox signaling to promote cell survival in an inflammatory and hypoxic microenvironment in order to maintain/expand tumor phenotype. The administration of tyrosine kinase inhibitor (TKI) constitutes the recommended therapeutic strategy in different malignancies at advanced stages. There are important interrelationships between cell stress, redox status, mitochondrial function, metabolism and cellular signaling pathways leading to cell survival/death. The induction of apoptosis and cell cycle arrest widely related to the antitumoral properties of TKIs result from tightly controlled events involving different cellular compartments and signaling pathways. The aim of the present review is to update the most relevant studies dealing with the impact of TKI treatment on cell function. The induction of endoplasmic reticulum (ER) stress and Ca2+ disturbances, leading to alteration of mitochondrial function, redox status and phosphatidylinositol 3-kinase (PI3K)-protein kinase B (Akt)-mammalian target of rapamycin (mTOR) and AMP-activated protein kinase (AMPK) signaling pathways that involve cell metabolism reprogramming in cancer cells will be covered.     Caracterización fenotípica del ganado criollo en el Parque nacional Huascarán – Ancash

    The aim of this study was to evaluate the phenotypic characteristics of the Creole cattle of the Huascarán National Park (PNH), Ancash, Peru through zoometric measurements and indexes of productive interest. The study was conducted in a population of Creole cattle in the Llanganuco zone of the PNH. Data were collected from 32 cows bovines. The zoometric measurements were head width (AC), head length (LC), chest girth (PT), body length (LCU), height withers (ALC) and height at the rump (ALG), anterior rump width (AAG), rear rump with (APG) and rump length (LG). Indexes related to meat ability were calculated: transversal pelvic index (IPT) and longitudinal pelvic index (IPL), and ethnological indexes: anamorphosis index (AI), corporal index (IC), pelvic index (PI) and proportionality index (IPR). The data were processed with the statistical program R. The colour and pattern of the coat were registered applying the nomenclature for Spanish bullfighting cattle. The zoomometric measurements showed a low CV (4.29 – 9.25), except for APG (14.75), reflecting morphological uniformity in the sample. The zoometric indices showed CV less than 10% (5.89 – 7.11), except for the AI ​​(16.28) and IC (11.5). As for the coat, a tendency towards simple colours (46.9%) with a predominance of black was observed. In general, the Creole bovine of the Quebrada de Llanganuco presents similar indexes with the Creole bovine from Ayacucho, Peru, classifying it as a short-bodied animal.El objetivo del presente estudio evaluar las características fenotípicas del ganado criollo del Parque Nacional Huascarán (PNH), Ancash, Perú, mediante la determinación de mediciones zoométricas e índices de interés productivo. El estudio se realizó en una población de bovinos criollos en la quebrada de Llanganuco del PNH. Se colectaron datos de 32 bovinos hembra adultos. Las medidas zoométricas tomadas fueron ancho (AC) y largo (LC) de cabeza, perímetro torácico (PT), largo de cuerpo (LCU), alzada a la cruz (ALC) y a la grupa (ALG), ancho anterior (AAG) y posterior de grupa (APG) y largo de grupa (LG). Se calcularon índices asociados a capacidad cárnica: índice pelviano transversal (IPT) e índice pelviano longitudinal (IPL), e índices etnológicos: índice de anamorfosis (IA), índice corporal (IC), índice pelviano (IP) e índice de proporcionalidad (IPR). Los datos fueron procesados con el programa estadístico R. Se registró el color y patrón del pelaje aplicando la nomenclatura para ganado de lidia. Las medidas zoométricas mostraron un bajo CV (4.29 – 9.25), excepto para APG (14.75), reflejando uniformidad morfológica en la muestra. Los índices zoométricos mostraron CV menores al 10% (5.89 – 7.11), excepto para el IA (16.28) e IC (11.5). En el pelaje se observó una tendencia hacia los colores simples (46.9%) con predominio del negro. En general, el bovino criollo de la Quebrada de Llanganuco presenta índices similares con el bovino criollo de Ayacucho, Perú, clasificándolo como un animal brevilíneo

    The shadow of the past: Convergence of young and old South American desert lizards as measured by head shape traits

    Convergence is a pervasive phenomenon in the Tree of Life, and evolution of similar phenotypes sharing the same environmental conditions is expected in phylogenetically closely related species. In contrast, contingent factors are probably more influential in shaping phenotypic diversity for distantly related taxa. Here, we test putative convergent evolution of lizard head morphologies among relatively closely related desert dwelling Liolaemus species, and the very distantly related Ctenoblepharys adspersa. We estimated a multilocus time-calibrated phylogeny of 57 species of South American liolaemus lizards, based on seven molecular markers. We collected head shape data for 468 specimens, and used three phylogenetic comparative methods (SURFACE, CONVEVOL, and WHEATSHEAF index) to test for and estimate the strength of convergence. We found strong evidence for convergence among Pacific desert lizard C. adspersa, Liolaemus audivetulatus, Liolaemus insolitus, Liolaemus poconchilensis, Liolaemus stolzmanni, and a candidate species (Liolaemus “Moquegua”). Our results suggest that, despite the long divergence and phylogenetic distance of C. adspersa with respect to convergent Liolaemus species, natural selection was probably more important than historical contingency in shaping phenotypic evolution in these desert lizards.Fil: Aguilar Puntriano, César.     Experiencia inicial en prostatectomía radical laparoscópica con el dispositivo de acceso laparoscópico umbilical Single-Incision Laparoscopic Surgery Port

    Since 2007, various urological procedures have been performed with laparoendoscopic single-site surgery (LESS surgery), including nephrectomy, pyeloplasty, simple prostatectomy and, with the refinement of laparoscopic instrumentation, radical prostatectomy. This paper reports our initial experience in radical prostatectomy using the SILS Port from Covidiem and two lateral 5-mm trocars for triangulation. The SILS Port allows for accurate, simple insertion through a Hadson incision. The flexible port accommodates three 5-mm cannulas or two 5-mm cannulas and a 12-mm port for easier instrument exchange through a single incision. This approach decreases morbidity from bleeding, hernia and/or internal organ damage and improves cosmetic. One-port single-incision laparoscopy is part of the natural development of minimally invasive surgery. Future research is required to assess the intraoperative and postoperative benefits of LESS surgery as compared to standard laparoscopy
