    Parâmetros para avaliação do estado nutricional de ferro

    A avaliação do estado nutricional de ferro em indivíduos e populações tem sido alvo de muitas pesquisas científicas, uma vez que existem algumas questões ainda indefinidas. Através de um levantamento bibliográfico, mediante consulta às bases de dados Medline, Lilacs e Dedalus, foram selecionadas publicações científicas em português e inglês, no período de 1972 a 1998, que se referiam aos parâmetros hematológicos e bioquímicos utilizados na avaliação do estado nutricional de ferro. Os parâmetros disponíveis refletem os três diferentes estágios da carência de ferro, a qual se manifesta de forma gradual e progressiva no organismo, até o desenvolvimento da anemia ferropriva. De forma geral, não possuem boa sensibilidade e especificidade quando considerados isoladamente, apresentando vantagens e limitações que devem ser examinadas no momento da escolha. No sentido de se aumentar a sensibilidade e a especificidade do diagnóstico do estado nutricional de ferro, têm-se utilizado combinações dos diferentes parâmetros disponíveis, considerando-se a contribuição de cada um, de acordo com as características dos indivíduos estudados, as facilidades metodológicas e o custo do processo.The assessment of iron status at individual and populational levels has been the target of many studies because some issues still need to be defined. A Medline, Lilacs and Dedalus literature review was carried out for the period of 1972 to 1998 and scientific publications in both English and Portuguese relating hematological and biochemical parameters were used in the assessment of iron status. The parameters reflect the three different stages of iron storage deficiency, which occur in a gradual and progressive way leading to the development of anaemia. In general, when used alone, these parameters are neither sensitive nor specific for detecting iron deficiency anaemia. Moreover, some advantages and limitations should be taken into consideration for the choice of the appropriate parameter. To improve both sensitivity and specificity, different parameters have been used in association and their specific contribution is determined according to the characteristics of the study population, methodological issues and the costs involved in the process

    Hemodynamic collapse and myocardial dysfunction are among the major causes ofdeath in severe sepsis. The purpose of this study was to assess the role played by TLR4and by the NLRP3 inflammasome in the cardiac dysfunction that occurs after highgradepolymicrobial sepsis. We performed the colon ascendens stent peritonitis (CASP)surgery in Tlr4-/-, Nlrp3-/- and caspase-1-/- mice. We also assessed for the first time theelectrical heart function in the CASP model. The QJ interval was increased in wild-typeC57BL/6J mice after CASP when compared to sham controls, a result paralleled by anincrease in the cardiac action potential duration (APD). The decreases in ejectionfraction (EF), left-ventricle end diastolic volume (LVEDV), stroke volume, and cardiacoutput found after CASP were similar among all groups of mice. Similar heart responsewas found when Nlrp3-/- mice were submitted to high-grade CLP. Despite developingcardiac dysfunction similar to wild-types after CASP, Nlrp3-/- mice had reducedcirculating levels of IL-1β, IL-6 and TNF-α. Our results demonstrate that the geneticablation of Tlr4, Nlrp3, and caspase-1 does not prevent the cardiac dysfunction, despitepreventing the increase in pro-inflammatory cytokines, indicating that these are notfeasible targets to therapy in high-grade sepsis.Fil: López Alarcón, Maria Micaela.     Comparação entre o HemoCue® e um contador automático para determinação de hemoglobina

    Com o objetivo de avaliar a precisão e concordância na determinação de hemoglobina (Hb) em sangue capilar e venoso, utilizando-se o HemoCue® e um contador automático, foram estudadas 29 gestantes. O HemoCue® apresentou baixa repetibilidade para as medidas em duplicata em sangue capilar (CR=0,53 g/dL; CV=13,6%) e venoso (CR=0,53 g/dL; CV=13,6%). As medidas de Hb obtidas em sangue capilar foram mais altas do que as obtidas em sangue venoso (12,4 e 11,7 g/dL, respectivamente; pTo determine the precision and agreement of the hemoglobin (Hb) measurements in capillary and venous blood samples by the HemoCue® and an automated counter. Hb was determined by both equipaments in blood samples of 29 pregnant women. The HemoCue® showed low repeatability of Hb measurements in duplicate in capillary (CR=0.53 g/dL, CV=13.6%) and venous blood (CR=0.53 g/dL, CV=13.6%).     Relação entre o estado nutricional de ferro de parturientes e seus recém-nascidos

    OBJECTIVE: To determine the relationship between iron nutritional status of pregnant women and their newborns using a combination of hematological and biochemical parameters for the diagnosis of iron deficiency. METHODS: A cross-sectional study was conducted in Jundiaí, Southeastern Brazil, in 2000. Venous blood samples collected from 95 pregnant women and from their umbilical cord and used for the determination of complete blood count, serum iron, total iron-binding capacity, serum ferritin, zinc protoporphyrin, and transferrin saturation. Women were classified into three groups: anemic, iron deficient and non-iron deficient. Statistical analysis included the Tukey-HSD test, Pearson's correlation coefficient and multiple linear regression analysis. RESULTS: Among pregnant women, 19% were anemic (97.9% mildly anemic and 2.1% moderately anemic) and 30.5% were iron deficient. No significant difference was seen in mean values of any parameter studied between newborns in the three groups (p>;0.05). Multiple linear regression analysis showed weak association between neonatal and maternal parameters. CONCLUSIONS: The iron nutritional status of pregnant women with iron deficiency or mild anemia does not seem to have a significant impact on the iron levels of their children.OBJETIVO: Determinar a relação entre os níveis de ferro de gestantes e seus filhos recém-nascidos, utilizando uma combinação de parâmetros hematológicos e bioquímicos para o diagnóstico da deficiência de ferro. MÉTODOS: Estudo transversal realizado em Jundiaí, SP, em 2000. Amostras de sangue venoso foram coletadas de 95 gestantes e do cordão umbilical de cada uma, e utilizadas na determinação de hemograma completo, ferro sérico, capacidade total de ligação do ferro, ferritina sérica, zinco-protoporfirina, e saturação de transferrina. As mulheres foram classificadas em três grupos: anêmicas, deficientes de ferro e não-deficientes de ferro. As análises estatísticas utilizadas foram o teste de Tukey-HSD, o coeficiente de correlação de Pearson e regressão linear múltipla. RESULTADOS: Entre as gestantes, 19% estavam anêmicas (97,9% levemente anêmicas e 2,1% moderadamente anêmicas) e 30,5% apresentavam deficiência de ferro. Não foi observada diferença significativa nas médias dos valores dos parâmetros estudados nos recém-nascidos dos três grupos (p>;0,05). A análise de regressão linear múltipla mostrou fraca associação entre os parâmetros maternos e neonatais. CONCLUSÕES: Os níveis de ferro de gestantes com deficiência de ferro ou com anemia leve/moderada parecem não influenciar de forma significativa os níveis de ferro de seus filhos

    Isolation of a seed coagulant Moringa oleifera lectin

    In this work hemagglutinating activity (HA) was investigated in distinct Moringa oleifera tissue extracts. A new lectin from seeds (cMoL) was purified and characterized; hemagglutinating and coagulating activities were evaluated. HA was detected in 0.15 M NaCl extracts from flowers and rachis inflorescence (5%, w/v), seeds, leaves, fundamental tissue of stem and steam bark (10%, w/v). cMoL isolated after saline extraction and guar gel column chromatography was active at pH range 4.0–9.0 agglutinating erythrocytes from rabbit and human blood types. Extracts of tissues and cMoL activities were carbohydrate inhibited; azocasein and asialofetuin abolished cMoL HA. The lectin was thermostable at 100 °C during 7 h. Polyacrylamide gel electrophoresis under reduced conditions revealed a main polypeptide band of 26.5 kDa; native basic cMoL was detected as a unique band.     Electroanalysis of Imidacloprid Insecticide in River Waters Using Functionalized Multi-Walled Carbon Nanotubes Modified Glassy Carbon Electrode

    In this work, a functionalized multi-walled carbon nanotubes modified glassy carbon electrode (GCE/MWCNT-f) was optimized for the direct determination of imidacloprid (IMC) insecticide in river water. The functionalized material was characterized by infrared spectroscopy with Fourier transform (FTIR) and the modified electrode by scanning electron microscopy (SEM) and cyclic voltammetry (CV). Results revealed that the GCE/MWCNT-f effectively increased the response toward IMC reduction by enhancing the reduction peak current and decreasing the peak potential in comparison with the bare electrode. After optimizing the electroanalytical conditions, the GCE/MWCNT-f showed a linear voltammetric response at concentration ranging from 2.40 × 10−7 to 3.50 × 10−6 mol L−1, with detection and quantification limits of 4.15 × 10−7 mol L−1 and 1.38 × 10−6 mol L−1, respectively. The recovery rate of IMC in spiked river water samples varied from 90–95%.     Nicotinic acid induces antinociceptive and anti-inflammatory effects in different experimental models

    AbstractAlthough in vitro studies have shown that nicotinic acid inhibits some aspects of the inflammatory response, a reduced number of in vivo studies have investigated this activity. To the best of our knowledge, the effects induced by nicotinic acid in models of nociceptive and inflammatory pain are not known. Per os (p.o.) administration of nicotinic acid (250, 500 or 1000mg/kg, −1h) inhibited the first and the second phases of the nociceptive response induced by formalin in mice. Nicotinic acid (250 or 500mg/kg, −1 and 3h) also inhibited the mechanical allodynia induced by carrageenan in rats, a model of inflammatory pain. However, in a model of nociceptive pain, exposure of mice to a hot-plate, nicotinic acid was devoid of activity. In addition to inhibiting the nociceptive response in models of inflammatory pain, nicotinic acid (250 or 500mg/kg, p.o., −1 and 3h) inhibited paw edema induced by carrageenan in mice and rats. Picolinic acid (62.5 or 125mg/kg, p.o., −1h), a nicotinic acid isomer, inhibited both phases of the nociceptive response induced by formalin, but not paw edema induced by carrageenan in mice. The other nicotinic acid isomer, isonicotinic acid, was devoid of activity in these two models. In conclusion, our results represent the first demonstration of the activity of nicotinic acid in experimental models of nociceptive and inflammatory pain and also provide further support to its anti-inflammatory activity. It is unlikely that conversion to nicotinamide represents an important mechanism to explain the antinociceptive and anti-inflammatory activities of nicotinic acid. The demonstration of new activities of nicotinic acid, a drug that has already been approved for clinical use and presents a positive safety record, may contribute to raise the interest in conducting clinical trials to investigate its usefulness in the treatment of painful and inflammatory diseases

    Este artigo é resultado da pesquisa de Iniciação Científica (PIBIC-PVF6325) realizada na Universidade Federal de Sergipe entre agosto de 2018 e julho de 2019 que analisou as características cromáticas presentes nas camadas de superfícies arquitetônicas de edificações históricas, identificando e mapeando as cores do Forro da Sacristia do Convento do Carmo Pequeno na cidade de São Cristóvão no Estado de Sergipe, com o intuito de conhecer a produção das cores antigas e o saber fazer local, assim como as patologias que atuam sobre essas superfícies pintadas, apreendendo e compreendendo a memória pictórica no Nordeste no Período Colonial Brasileiro. As pinturas do Forro do Carmo Pequeno, representativas da vida da Nossa Senhora Protetora (Virgem do Carmo), provavelmente pintadas no final do XVII até meados do século XVIII, compreendem doze painéis decorados e emoldurados por caixotões que ocupam todo o teto da Sacristia da Igreja da Ordem Terceira dos Calçados. A metodologia empregou desenhos digitalizados dos painéis a partir de extenso levantamento fotográfico baseado nas observações visuais locais, que serviram de base para produção de Fichas de Identificação, Mapeamento e Patologias. Este processo possibilitou, além da identificação e mapeamento das cores com a utilização de um colorímetro digital NCS 200, desvendar minúcias, particularidades, hibridismos figurativos, simbologias e técnicas antigas nas pinturas, resultando em um outro olhar científico sobre o patrimônio e no reconhecimento e valoração da identidade e memória cultural de uma determinada região e sociedade no seu tempo

    Effects induced by Apis mellifera venom and its components in experimental models of nociceptive and inflammatory pain

    AbstractThe effects induced by Apis mellifera venom (AMV), melittin-free AMV, fraction with molecular mass < 10 kDa (F<10) or melittin in nociceptive and inflammatory pain models in mice were investigated. Subcutaneous administration of AMV (2, 4 or 6 mg/kg) or melittin-free AMV (1, 2 or 4 mg/kg) into the dorsum of mice inhibited both phases of formaldehyde-induced nociception. However, F<10 (2, 4 or 6 mg/kg) or melittin (2 or 3 mg/kg) inhibited only the second phase. AMV (4 or 6 mg/kg), but not F<10, melittin-free AMV or melittin, induced antinociception in the hot-plate model. Paw injection of AMV (0.05 or 0.10 mg), F<10 (0.05 or 0.1 mg) or melittin (0.025 or 0.050 mg) induced a nociceptive response. In spite of inducing nociception after paw injection, scorpion (Tityus serrulatus) or snake (Bothrops jararaca) venom injected into the dorsum of mice did not inhibit formaldehyde-induced nociception. In addition, AMV (6 mg/kg), but not F<10 (6 mg/kg) or melittin (3 mg/kg), inhibited formaldehyde paw oedema. Concluding, AMV, F<10 and melittin induce two contrasting effects: nociception and antinociception. AMV antinociception involves the action of different components and does not result from non-specific activation of endogenous antinociceptive mechanisms activated by exposure to noxious stimuli

    Macrophage-dependent IL-1β production induces cardiac arrhythmias in diabetic mice

    Diabetes mellitus (DM) encompasses a multitude of secondary disorders, including heart disease. One of the most frequent and potentially life threatening disorders of DM-induced heart disease is ventricular tachycardia (VT). Here we show that toll-like receptor 2 (TLR2) and NLRP3 inflammasome activation in cardiac macrophages mediate the production of IL-1β in DM mice. IL-1β causes prolongation of the action potential duration, induces a decrease in potassium current and an increase in calcium sparks in cardiomyocytes, which are changes that underlie arrhythmia propensity. IL-1β-induced spontaneous contractile events are associated with CaMKII oxidation and phosphorylation. We further show that DM-induced arrhythmias can be successfully treated by inhibiting the IL-1β axis with either IL-1 receptor antagonist or by inhibiting the NLRP3 inflammasome. Our results establish IL-1β as an inflammatory connection between metabolic dysfunction and arrhythmias in DM.Facultad de Ciencias MédicasCentro de Investigaciones Cardiovasculare
