    A note on the magnetic spatial forcing of a ferrofluid layer

    We report on the response of a thin layer of ferrofluid to a spatially modulated magnetic field. This field is generated by means of a constant current in a special arrangement of aluminum wires. The full surface profile of the liquid layer is recorded by means of the absorption of X-rays. The outcome is analyzed particularly with regard to the magnetic self focusing effect under a deformable fluid layer

    Growth of surface undulations at the Rosensweig instability

    We investigate the growth of a pattern of liquid crests emerging in a layer of magnetic liquid when subjected to a magnetic field oriented normally to the fluid surface. After a steplike increase of the magnetic field, the temporal evolution of the pattern amplitude is measured by means of a Hall-sensor array. The extracted growth rate is compared with predictions from linear stability analysis by taking into account the proper nonlinear magnetization curve M(H). The remaining discrepancy can be resolved by numerical calculations via the finite-element method. By starting with a finite surface perturbation, it can reproduce the temporal evolution of the pattern amplitude and the growth rate. The investigations are performed for two magnetic liquids, one with low and one with high viscosity.Comment: 12 pages, 12 figure

    Novel insight into the reaction of nitro, nitroso and hydroxylamino benzothiazinones and of benzoxacinones with Mycobacterium tuberculosis DprE1

    Documento escrito por un elevado nĂşmero de autores/as, solo se referencia el/la que aparece en primer lugar y el/la autor/a perteneciente a la UC3M.Nitro-substituted 1,3-benzothiazinones (nitro-BTZs) are mechanism-based covalent inhibitors of Mycobacterium tuberculosis decaprenylphosphoryl ...-D-ribose-2-oxidase (DprE1) with strong antimycobacterial properties. We prepared a number of oxidized and reduced forms of nitro-BTZs to probe the mechanism of inactivation of the enzyme and to identify opportunities for further chemistry. The kinetics of inactivation of DprE1 was examined using an enzymatic assay that monitored reaction progress up to 100 min, permitting compound ranking according to kinact/Ki values. The side-chain at the 2-position and heteroatom identity at the 1-position of the BTZs were found to be important for inhibitory activity. We obtained crystal structures with several compounds covalently bound. The data suggest that steps upstream from the covalent end-points are likely the key determinants of potency and reactivity. The results of protein mass spectrometry using a 7-chloro-nitro-BTZ suggest that nucleophilic reactions at the 7-position do not operate and support a previously proposed mechanism in which BTZ activation by a reduced flavin intermediate is required. Unexpectedly, a hydroxylamino-BTZ showed time-dependent inhibition and mass spectrometry corroborated that this hydroxylamino-BTZ is a mechanism-based suicide inhibitor of DprE1. With this BTZ derivative, we propose a new covalent mechanism of inhibition of DprE1 that takes advantage of the oxidation cycle of the enzyme

    Sensitivity of Bunker Cave to climatic forcings highlighted through multi-annual monitoring of rain-, soil-, and dripwaters

    The last two decades have seen a considerable increase in studies using speleothems as archives of past climate variability. Caves under study are now monitored for a wide range of environmental parameters and results placed in context with speleothem data. The present study investigates trends from a seven year long monitoring of Bunker Cave, northwestern Germany, in order to assess the hydraulic response and transfer time of meteoric water from the surface to the cave. Rain-, soil-, and dripwater were collected from August 2006 to August 2013 at a monthly to bimonthly resolution and their oxygen and hydrogen isotope composition was measured. Furthermore, drip rates were quantified. Due to different drip characteristics, annual mean values were calculated for the drip rates of each drip site. Correlations of the annual mean drip rate of each site with precipitation and infiltration demonstrate that the annual infiltration, and thus the annual precipitation control the inter-annual drip-rate variability for all except one site. The hydraulic response is not delayed on an annual basis. All drip sites display identical long-term trends, which suggests a draining of a common karst reservoir over these seven years of monitoring. Correlations of soil- and dripwater monthly δ18O and δD values with atmospheric temperature data reveal water transfer times of 3 months to reach a depth of 40 cm (soilwater at site BW 2) and 4 months for 70 cm depth (soilwater at site BW 1). Finally, the water reaches the cave chambers (15 to 30 m below land surface) after ca. 2.5 years. Consequently, a temporal offset of 29 to 31 months (ca. 2.5 years) between the hydraulic response time (no time lag on annual basis) and the water transfer time (time lag of 29 to 31 months) was found, which is negligible with regard to Bunker Cave speleothems because of their slow growth rates. Here, proxies recording precipitation/infiltration and temperature are registered on a decadal scale. Variations in drip rate and thus precipitation and infiltration are recorded by δ13C and Mg/Ca ratios in speleothem calcite. Speleothem δ18O values reflect both temperature and precipitation signals due to drip rate-related fractionation processes. We document that long-term patterns in temperature and precipitation are recorded in dripwater patterns of Bunker Cave and that these are linked to the North Atlantic Oscillation (NAO)

    Individual and spousal education, mortality and life expectancy in Switzerland: a national cohort study.

    BACKGROUND Household measures of socioeconomic position may better account for the shared nature of material resources, lifestyle, and social position of cohabiting persons, but household measures of education are rarely used. We aimed to evaluate the association of combined educational attainment of married couples on mortality and life expectancy in Switzerland. METHODS The study included 3 496 163 ever-married persons aged ≥30 years. The 2000 census was linked to mortality records through 2008. Mortality by combined educational attainment was assessed by gender-age-specific HRs, with 95% CIs from adjusted models, life expectancy was derived using abridged life tables. RESULTS Having a less educated partner was associated with increased mortality. For example, the HR comparing men aged 50-64 years with tertiary education married to women with tertiary education to men with compulsory education married to women with compulsory education was 2.05 (1.92-2.18). The estimated remaining life expectancy in tertiary educated men aged 30 years married to women with tertiary education was 4.6 years longer than in men with compulsory education married to women with compulsory education. The gradient based on individual education was less steep: the HR comparing men aged 50-64 years with tertiary education with men with compulsory education was 1.74 (1.67-1.81). CONCLUSIONS Using individual educational attainment of married persons is common in epidemiological research, but may underestimate the combined effect of education on mortality and life expectancy. These findings are relevant to epidemiologic studies examining socio-demographic characteristics or aiming to adjust results for these characteristics

    Body mass index distribution in rheumatoid arthritis: a collaborative analysis from three large German rheumatoid arthritis databases

    Background METARTHROS (Metabolic impact on joint and bone disease) is a nationwide German network to investigate the overlap between inflammatory and metabolic diseases. The objective of this study was to compare the body mass index (BMI) distribution in patients with early and established rheumatoid arthritis (RA) with data from the general population, and to evaluate the association of BMI with patient characteristics and clinical markers. Methods The BMI distribution was examined with data collected at inclusion of patients in the early arthritis cohort CAPEA, the biologics register RABBIT, and the National database of the German Collaborative Arthritis Centers. A data source with a representative sample of the German population (German Ageing Survey) was used as a comparator. BMI categories of <18.5 kg/m2 (underweight), 18.5 to <25 kg/m2 (normal weight), 25 to <30 kg/m2 (overweight), and ≥30 kg/m2 (obese) were used. Patients were stratified by age and sex, and compared to controls from the German Ageing Survey. Associations between BMI and markers of disease activity were analysed with non-parametric tests and linear models. Results Data from 1207 (CAPEA), 12,230 (RABBIT), and 3424 (National database) RA patients and 6202 population controls were evaluated. The mean age was 56, 56, 62, and 62 years, respectively, the mean disease duration was 13 weeks, 9.9 years, and 13.5 years, respectively, and the mean disease activity score (DAS28) was 5.1, 5.2, and 3.1, respectively. In all RA cohorts, obesity was more frequent (23.8 %, 23.4 %, 21.4 %, respectively) than in controls (18.2 %). This applied to all age groups <70 years, was independent of disease duration, and was more pronounced in females. In all cohorts, the age at RA onset was associated with BMI, being higher in overweight/obese patients compared to normal-weight patients. Current smoking was negatively associated with BMI. Linear analyses revealed increased erythrocyte sedimentation rate (ESR) values in underweight and obese females, and an increasing disparity between tender joint counts (TJCs) and swollen joint counts (SJCs) in higher BMI categories. Conclusions Compared to the general population, a higher prevalence of obesity was observed in all RA cohorts. The dominance of obesity in females and the different behaviour of disease activity markers in relation to the BMI in females indicate that additional parameters need to be considered when analysing the impact of obesity on inflammation in RA
