    Effect of HER2 Expression on NUPL2 Protein in Cervical Cancer Cells

    Recurring cervical cancer patients frequently present overexpressed Human Epidermal Growth Factor Receptor 2 (HER2) protein, which promotes the growth of the cancer. Some HER2-expressing cells and cervical cancer patients have enhanced gene expression of Nucleoporin-like 2 (NUPL2). As part of the nuclear pore complex, the NUPL2 protein serves to selectively export substances from the nucleus to the cytoplasm. In this experiment, HER2 was expressed in cervical cancer cells to examine its effect on NUPL2 protein expression and localization. Interestingly, there was no difference in NUPL2 protein levels between HER2-expressing and non-expressing cells. Importantly, consistent instances of NUPL2 protein localization specific to the nucleus occurred in a large sampling of HER2-expressing cells. This is significant because it suggests a mechanism as to how HER2 promotes excessive cell growth, via changes in NUPL2 localization, potentially impacting its function and ability to selectively export substances that modulate cell growth

    Interpretations of, and orientations to, “understanding mathematics in depth”: students in MEC programmes across institutions

    In this paper we present initial findings from our study of interpretations and orientations to ‘understanding mathematics in depth’ among students in selected Mathematics Enhancement Courses (MEC) in the UK. The MEC is a 26-week pre-Initial Teacher Education (ITE) ‘mathematics subject knowledge for teaching’ course designed for, and undertaken by, graduates wishing to teach mathematics at secondary level, but do not have a Mathematics degree. It is completed before commencing with a PGCE. A common theme running through the MEC documentation is the importance of ‘understanding mathematics in depth’. We are interested in what and how MEC students interpret and orient themselves towards ‘understanding mathematics in depth’. In designing and conducting our empirical work we have drawn upon a related project in South Africa, which is exploring ‘mathematics for teaching’, specifically what and how mathematics and teaching are co-constituted in mathematics teacher education programmes. The MEC is an interesting empirical context for such study, as it is a mathematics course, or set of courses, specifically designed for future teachers. We have collected data through guided, semi-structured interviews with 18 students and 4 lecturing staff at three different institutions. The interpretations and orientations of MEC students towards mathematics and the notion of ‘understanding mathematics in depth’, we contend, provide additional insight into the developing notion of mathematical knowledge in and for teaching

    Early intervention for lactate dehydrogenase elevation improves clinical outcomes in patients with the HeartMate II left ventricular assist device: Insights from the PREVENT study

    BACKGROUND: Hemolysis, assessed by elevated serum lactate dehydrogenase (LDH), is strongly associated with HeartMate II pump thrombosis (PT). However, it is unknown whether early intervention for elevated LDH circumvents the risk of serious PT requiring pump exchange. We sought to evaluate the relationship between elevated LDH and clinical outcomes, the effectiveness of early medical intervention, and risk factors for elevated LDH. METHODS: We studied 268 patients in the prospective, multicenter PREVENT study who had 2 or more LDH measurements at ≥30 days post-implant. Elevated LDH was defined as LDH ≥2.5× upper limit of normal (ULN) for 2 consecutive measurements. RESULTS: Fourteen percent of patients had elevated LDH. Stroke-free survival at 6 months was lower in patients with elevated LDH vs patients with normal LDH (83 ± 6% vs 93 ± 2%, p = 0.035). Elevated LDH resolved without intervention in 19% of patients, with intensified medical therapy in 43% and required surgical intervention in 38%. For patients receiving only medical therapy, survival was 94 ± 6% at 6 months post-treatment. In this subgroup, resolution of symptoms with intensified medical therapy was sustained in 15 of 16 patients, with PT occurring in 1 patient at 171 days after initial treatment for elevated LDH (202 days post-implant). Early medical intervention at moderately elevated LDH (2.5× to 3.2× ULN), as compared with higher levels (>3.2× ULN), led to more sustained resolution of symptoms without subsequent PT or need for surgical intervention (91% vs 26% at 6 months post-treatment, p = 0.002). CONCLUSIONS: Early medical intervention can successfully resolve moderate LDH elevations (2.5× to 3.2× ULN) with a low incidence of death or PT at 6 months post-treatment
