    Global disparities in surgeons’ workloads, academic engagement and rest periods: the on-calL shIft fOr geNEral SurgeonS (LIONESS) study

    : The workload of general surgeons is multifaceted, encompassing not only surgical procedures but also a myriad of other responsibilities. From April to May 2023, we conducted a CHERRIES-compliant internet-based survey analyzing clinical practice, academic engagement, and post-on-call rest. The questionnaire featured six sections with 35 questions. Statistical analysis used Chi-square tests, ANOVA, and logistic regression (SPSS® v. 28). The survey received a total of 1.046 responses (65.4%). Over 78.0% of responders came from Europe, 65.1% came from a general surgery unit; 92.8% of European and 87.5% of North American respondents were involved in research, compared to 71.7% in Africa. Europe led in publishing research studies (6.6 ± 8.6 yearly). Teaching involvement was high in North America (100%) and Africa (91.7%). Surgeons reported an average of 6.7 ± 4.9 on-call shifts per month, with European and North American surgeons experiencing 6.5 ± 4.9 and 7.8 ± 4.1 on-calls monthly, respectively. African surgeons had the highest on-call frequency (8.7 ± 6.1). Post-on-call, only 35.1% of respondents received a day off. Europeans were most likely (40%) to have a day off, while African surgeons were least likely (6.7%). On the adjusted multivariable analysis HDI (Human Development Index) (aOR 1.993) hospital capacity > 400 beds (aOR 2.423), working in a specialty surgery unit (aOR 2.087), and making the on-call in-house (aOR 5.446), significantly predicted the likelihood of having a day off after an on-call shift.     The technique of Fragment‐Based Drug Design (FBDD) considers the interactions of different moieties of molecules with biological targets for the rational construction of potential drugs. One basic assumption of FBDD is that the different functional groups of a ligand interact with a biological target in an approximately additive, that is, independent manner. We investigated the interactions of different fragments of ligands and Interleukin‐1 Receptor‐Associated Kinase 4 (IRAK‐4) throughout the FBDD design of Zimlovisertib, a promising anti‐inflammatory, currently in trials to be used for the treatment of COVID‐19 pneumonia. We utilised state‐of‐the‐art methods of wave function analyses mainly the Interacting Quantum Atoms (IQA) energy partition for this purpose. By means of IQA, we assessed the suitability of every change to the ligand in the five stages of FBDD which led to Zimlovisertib on a quantitative basis. We determined the energetics of the interaction of different functional groups in the ligands with the IRAK‐4 protein target and thereby demonstrated the adequacy (or lack thereof) of the changes made across the design of this drug. This analysis permits to verify whether a given alteration of a prospective drug leads to the intended tuning of non‐covalent interactions with its protein objective.  Overall, we expect that the  methods exploited in this paper will prove valuable in the understanding and control of chemical modifications across FBDD processes

    Son muchos los factores que influyen en la producción de larvas y alevinos de peces, destacándose la temperatura, la calidad del agua, la especie, el estado nutricional, la calidad y cantidad del alimento disponible durante la primera alimentación, entre otros. Dicho alimento debe sostener los diferentes cambios fisiológicos y estructurales en el desarrollo de órganos y sistemas. El sistema digestivo es uno de los más importantes durante las fases tempranas de peces, debido a que es donde se da la principal absorción de nutrientes suficientes para el crecimiento y el desarrollo. No obstante, existen diferencias en el desarrollo entre las especies, considerando así, el estudio ontogénico del tracto digestivo una herramienta útil para determinar el momento oportuno de la formación de los diferentes órganos. En este sentido, conocer y entender el manejo de la primera alimentación y cómo influye en el desarrollo del sistema digestivo de larvas es un aspecto fundamental en el éxito de la producción de larvas de calidad y resistencia. Por lo tanto, el objetivo de esta revisión es ofrecer información actualizada sobre el desarrollo ontogénico y principales enzimas del sistema digestivo, así como la influencia del tipo de alimento en el desarrollo de las larvas de peces.There are many factors that influence the production of larvae and fingerlings of fish, among which the temperature, the quality of the water, the species, the nutritional status, the quality and quantity of the food available during the first feeding stand out, among others. This food must support the different physiological and structural changes in the development of organs and systems. The digestive system is one of the most important during the early stages of fish, because it is where the main absorption of sufficient nutrients for growth and development occurs. However, there are differences in development between species, thus considering the ontogenetic study of the digestive tract a useful tool to determine the opportune moment for the formation of the different organs. In this sense, knowing and understanding the management of the first feeding and how it influences the development of the digestive system of larvae is a fundamental aspect that influences the success of the production of quality and resistance larvae. Therefore, the objective of this review is to offer updated information on the ontogenetic development and main enzymes of the digestive system, as well as the influence of the type of food on the development of fish larvae

    Comprehensive virtual screening of 4.8 k flavonoids reveals novel insights into allosteric inhibition of SARS-CoV-2 M<sup>PRO</sup>

Since flavonoids show antiviral activity, several in silico works have proposed them as potential SARS-CoV-2 main protease inhibitors. Nonetheless, there is reason to doubt certain results given the lack of consideration for flavonoid promiscuity or main protease plasticity, usage of short library sizes, absence of control molecules and/or the limitation of the methodology to a single target site. Here, we report a virtual screening study where dorsilurin E, euchrenone a11, sanggenol O and CHEMBL2171598 are proposed to inhibit main protease through different pathways. Remarkably, novel structural mechanisms were observed after sanggenol O and CHEMBL2171598 bound to experimentally proven allosteric sites. The former drastically affected the active site, while the latter triggered a hinge movement which has been previously reported for an inactive SARS-CoV main protease mutant. The use of a curated database of 4.8 k flavonoids, combining two well-known docking software (AutoDock Vina and AutoDock4.2), molecular dynamics and MMPBSA, guaranteed an adequate analysis and robust interpretation.     Naturaleza Urbana presenta experiencias autogestionadas que, con el tiempo, se han posicionado como ejercicios alternativos de identificación, monitoreo y recuperación de la biodiversidad urbana. En otros casos, el modelo comunidad-gobierno ha permitido desarrollar diagnósticos y propuestas de gestión corresponsables y sistémicas, entendiendo por esto último iniciativas que nacen desde los valores mismos que cada comunidad le atribuye a su biodiversidad. Del mismo modo, se presentan esfuerzos gubernamentales que han enriquecido la visión ambiental de los principales instrumentos de planificación urbana, por ejemplo, integrando la condición propiamente urbana como oportunidad para aumentar la oferta ambiental de la ciudad, fortaleciendo las funciones y procesos de la biodiversidad y revitalizando, con ello, la calidad de vida del entorno urbano. Por su parte, las universidades y los centros de investigación se han sumado a la ola emergente de generación de conocimiento en biodiversidad urbana (fenómeno nacional e internacional), han brindado evidencia científica de su valor para el bienestar humano y han propuesto reflexiones y lineamientos cualitativos de biodiversidad, con miras a hacer del ordenamiento un ejercicio más coherente con cada contexto territorial en particular.Bogotá, D.
