    Lower genetic diversity in the limpet Patella caerulea on urban coastal structures compared to natural rocky habitats

    Human-made structures are increasingly found in marine coastal habitats. The aim of the present study was to explore whether urban coastal structures can affect the genetic variation of hard-bottom species. We conducted a population genetic analysis on the limpet Patella caerulea sampled in both natural and artificial habitats along the Adriatic coast. Five microsatellite loci were used to test for differences in genetic diversity and structure among samples. Three microsatellite loci showed strong Hardy-Weinberg disequilibrium likely linked with the presence of null alleles. Genetic diversity was significantly higher in natural habitat than in artificial habitat. A weak but significant differentiation over all limpet samples was observed, but not related to the type of habitat. While the exact causes of the differences in genetic diversity deserve further investigation, these results clearly point that the expansion of urban structures can lead to genetic diversity loss at regional scales

    Chasing genetic structure in coralligenous reef invertebrates: patterns, criticalities and conservation issues

    Conservation of coastal habitats is a global issue, yet biogenic reefs in temperate regions have received very little attention. They have a broad geographic distribution and are a key habitat in marine ecosystems impacted by human activities. In the Mediterranean Sea coralligenous reefs are biodiversity hot spots and are classified as sensitive habitats deserving conservation. Genetic diversity and structure influence demographic, ecological and evolutionary processes in populations and play a crucial role in conservation strategies. Nevertheless, a comprehensive view of population genetic structure of coralligenous species is lacking. Here, we reviewed the literature on the genetic structure of sessile and sedentary invertebrates of the Mediterranean coralligenous reefs. Linear regression models and meta-analytic approaches are used to assess the contributions of genetic markers, phylum, pelagic larval duration (PLD) and geographical distance to the population genetic structure. Our quantitative approach highlight that 1) most species show a significant genetic structure, 2) structuring differs between phyla, and 3) PLD does not appear to be a major driver of the structuring. We discuss the implication of these finding for the management and conservation, suggesting research areas that deserve attention, and providing recommendations for broad assessment and monitoring of genetic diversity in biogenic reefs specie

    Genetic structure of Norway lobster, <i>Nephrops norvegicus</i> (L.) (Crustacea: Nephropidae), from the Mediterranean Sea

    La estructura genética poblacional de la cigala (Nephrops norvegicus) se estudió mediante electroforesis alocímica en acetato de celulosa. Las muestras analizadas provienen de ocho localidades del Mediterráneo y una del Atlántico adyacente. En un total de 15 sistemas enzimáticos se detectaron 23 loci, de los cuales 11 resultaron polimórficos. La heterozigosis media detectada varía desde 0,052 (se 0,025) a 0,142 (se 0,040) y es similar a la observada para otros decápodos. Las distancias de Nei muestran una moderada diferenciación genética, sin embargo el análisis de agrupamiento UPGMA no revela ningún patrón geográfico. No se detectaron clinas en las frecuencias alélicas, la variabilidad genética parece presentar una distribución al azar y la estructura genética de la cigala parece seguir el modelo de insularidad. El valor promedio de FST = 0,122 confirma un cierto grado de subdivisión genética e indica un flujo génico estimado de Nm = 1,80

    Evidence of genetic isolation between two Mediterranean morphotypes of Parazoanthus axinellae

    Coralligenous assemblages are among the most species-rich and vulnerable habitats of the Mediterranean Sea. Nevertheless, data on connectivity patterns on species inhabiting these habitats, crucial to define management and protection priorities, are largely lacking. Moreover, unreliable species-level taxonomy can confound ecological studies and mislead management strategies. In the northwestern Mediterranean two Parazoanthus axinellae morphotypes differing in size, color and preferred substrate are found in sympatry. In this study, we used COI and ITS sequence polymorphism to assess (1) the genetic divergence between the two morphotypes, (2) their connectivity patterns and (3) their phylogenetic position within the Parazoanthidae. Specimens of P. axinellae were sampled in 11 locations along the northwestern Mediterranean; in 6 locations, samples of the two morphotypes were collected in sympatry. Small genetic diversity and structure were found within morphotypes, while marked and consistent differentiation was detected between them. Moreover, the less widespread morphotype appeared to be closer to Pacific species as P. juanfernandezii and P. elongatus. Our findings confirmed the limited knowledge on Parazoanthus species complex, and how this gap can have important implication for the conservation strategies of this widespread and valuable genus in the Mediterranean Sea

    Integrative taxonomy approach to detect spatial and temporal variability of the Mediterranean benthic communities through artificial substrate units (ASUs)

    Monitoring spatial and temporal changes of marine benthic communities using standardized procedures is essential to take necessary steps towards conservation of marine ecosystems. In this study we combined Artificial Substrate Units (ASUs) for sampling of benthic communities, with integrative taxonomy approach that incorporated morphological identification of organisms and COI DNA metabarcoding, to characterize the diversity of communities at three locations across the Central Mediterranean Sea (Livorno and Palinuro, Italy; Rovinj, Croatia) in 2019 and 2020. Significant differences in the communities’ structure were observed both at large spatial scale between sampled locations, and at small spatial scale (less than ten kilometers) between sites. Moreover, significant temporal variability in species richness and structure of benthic assemblages was detected, with higher richness in 2020. Revealed variability can probably be attributed to the peculiar geomorphological, oceanographical, and ecological features of locations, but also to the influence of interplaying local chemical and physical factors and biological processes such as species settlement, competition, and migration that can act at small spatial and temporal scales. The similarity in the species composition and community structure accessed by morphological and metabarcoding approaches was low, with only 16% of the species (out of 133 species identified overall) commonly detected by both approaches. This is mostly both due to a lack of data on COI sequences of numerous benthic invertebrates in the public barcoding databases on the one hand, and difficulties in detecting small and cryptic taxa through morphological analyses on the other. This suggests that combining of two approaches is required to fully describe the biodiversity of benthic assemblages. Finally, comparison of the spatial variability of the benthic communities’ structure with two approaches at different taxonomic levels, indicated that genus and family levels give results that are consistent to those obtained by the species level. This suggests that family level might be satisfactory in monitoring the spatial-temporal variability of Mediterranean hard bottom benthic communities

    Opadanje stoka školjkaša Ruditapes philippinarum u uvalama sjevernog Jadrana:anketa o ekološkim i društveno-ekonomskim aspektima

    The Manila clam, Ruditapes philippinarum, introduced in the Venice lagoons in 1983 and rapidly spread to the nearby coastal lagoons represents one of the most important commercially- exploited resources of this area. Abundance, size-class and biomass distribution of the wild population living in the Pialassa Baiona lagoon were assessed and related to the hydrological and sediment characteristics. Despite the lagoon being affected by eutrophication, chemical and thermal pollution, the clams were harvested by about thirty professional fishermen until 2003. The commercially available stock was estimated at 36.8 10 3 kg in July 2002, 29.3 10 3 kg in April 2003, and 10.3 10 3 kg in October 2003. Stock estimations and observed mortality were in good accordance with the fishermen data. The decline of the available stock could be due to both overfishing and the extraordinary summer heat wave occurred in 2003, which may have reduced larval recruitment and increased the mortality. Overall, the juvenile recruitment appeared insufficient to annually restore the natural stock. Although the national and regional high relevance of clams market, local harvesting of wild populations appeared marginal and inadequate to support a remunerative commercial activity, due to the variability and unpredictability of the annual yield and the lack of a sustainable management based on a production chain’s approach.Školjkaš Ruditapes philippinarum, zabilježena u Venecijanskoj laguni 1983. godine gdje se brzo proširila i na obližnje obalne lagune te predstavlja jedan od najvažnijih komercijalno iskorištenih resursa ovog područja. U laguni Pialassa Baiona procjenjene su abundancija, veličina klase i raspodjela biomase prirodnih populacija u odnosu na hidrološke i sedimentne karakteristike. Unatoč tome što je laguna bila pogođena eutrofikacijom, kemijskim i toplinskim zagađenjima, oko trideset profesionalnih ribara sakupljalo je školjke do 2003. godine. Dostupne zalihe stoka komercijalnih vrsta procijenjene su na 36,8 10, 3kg u srpnju 2002. godine, zatim na 29.3 10 3kg u travnju 2003. godine i na 10,3 10 3 kg u listopadu 2003. godine. Procjene zaliha i smrtnosti bile su u skladu s podacima ribara. Pad raspoloživih zaliha mogao bi biti posljedica prekomjernog izlova i izvanrednog ljetnoga toplinskog vala koji se dogodio 2003. godine, što je utjecalo na smanjenje novačenja ličinki i povećanje smrtnosti. Sveukupno gledano novačenje juvenilnih primjeraka nije bilo dovoljno za godišnje obnavljanje prirodnih zaliha. Iako je na nacionalnoj i regionalnoj razini velika važnost tržišta školjki, lokalna berba divljih populacija jest marginalna i neadekvatna za podupiranje profitabilne komercijalne aktivnosti, zbog varijabilnosti i nepredvidljivosti godišnjeg prinosa i nedostatka održivog upravljanja temeljenog na pristupu proizvodnog lanca

    Targeting neurosteroid synthesis as a therapy for schizophrenia-related alterations induced by early psychosocial stress

    Background Cogent evidence has shown that schizophrenia vulnerability is enhanced by psychosocial stress in adolescence, yet the underpinnings of this phenomenon remain elusive. One of the animal models that best capture the relationship between juvenile stress and schizophrenia is isolation rearing (IR). This manipulation, which consists in subjecting rats to social isolation from weaning through adulthood, results in neurobehavioral alterations akin to those observed in schizophrenia patients. In particular, IR-subjected rats display a marked reduction of the prepulse inhibition (PPI) of the startle reflex, which are posited to reflect imbalances in dopamine neurotransmission in the nucleus accumbens (NAcc). We recently documented that the key neurosteroidogenic enzyme 5α-reductase (5αR) plays an important role in the dopaminergic regulation of PPI; given that IR leads to a marked down-regulation of this enzyme in the NAcc, the present study was designed to further elucidate the functional role of 5αR in the regulation of PPI of IR-subjected rats. Methods We studied the impact of the prototypical 5αR inhibitor finasteride (FIN) on the PPI deficits and NAcc steroid profile of IR-subjected male rats, in comparison with socially reared (SR) controls. Results FIN (25–100 mg/kg, i.p.) dose-dependently countered IR-induced PPI reduction, without affecting gating integrity in SR rats. The NAcc and striatum of IR-subjected rats displayed several changes in neuroactive steroid profile, including a reduction in pregnenolone in both SR and IR-subjected groups, as well as a decrease in allopregnanolone content in the latter group; both effects were significantly opposed by FIN. Conclusions These results show that 5αR inhibition counters the PPI deficits induced by IR, possibly through limbic changes in pregnenolone and/or allopregnanolone concentrations

    The Environmental Effects of the Innovative Ejectors Plant Technology for the Eco-Friendly Sediment Management in Harbors

    A sediment bypassing plant based on innovative jet pump, ejectors, has been tested in the first-of-a-kind demo application at the harbor of Cervia (Italy, Northern Adriatic Sea). The ejector is a jet pump aimed to reduce sediment accumulation in navigation channels and coastal areas. Herein we present results of the first study assessing the potential ecological effects of the ejectors plant. Sediment characteristics, benthic, and fish assemblages before and after the plant activation have been analyzed in the putatively impacted (the sediment removal and discharge) areas and four control locations, one time before and two times after plant activation. Ejectors plant operation resulted in a reduction of the mud and organic matter content in the sediment, as well as in changes in shell debris amount in the impacted areas. Abundance and species richness of benthic macroinvertebrates, initially reduced in the impacted areas, probably due to the previous repeated dredging, returned to higher values during demo plant continuous operation. Higher diversity of fish fauna was observed in the study area during plant operation period. Observed dynamics of the ecological status of the marine habitat suggest that an ejectors plant could represent an eco-friendly solution alternative to dredging operations to solve harbor siltation problems
