    In-vitro anticoccidial potential of Saccharum officinarum extract against Eimeria oocysts

    Present study was conducted to evaluate the effect of aqueous methanolic extract from Saccharum officinarum on the sporulation and morphology of oocysts of four Eimeria species (Eimeria tenella, E. necatrix, E. mitis, E. brunetti) of poultry. Sporulation inhibition bioassay was used to evaluate the activity of Saccharum officinarum extract (SOE) on the sporulation of coccidian oocysts. In this assay, unsporulated oocysts were exposed to six concentrations of S. officinarum in 10% dimethyl sulfoxide solution (w/v; 10, 5, 2.5, 1.25, 0.625 and 0.31%) while DMSO and potassium dichromate solution (K2Cr2O7) served as control groups. The Petri dishes were partially covered to allow the passage of oxygen and incubated at 25-29° C for 48 h, providing 60-80% humidity. The sporulation of the oocyst was confirmed by examining sporocysts under inverted microscope at 40x. Results showed anticoccidial activity of SOE against all Eimeria species as proved by its ability to inhibit the sporulation of the oocysts under laboratory conditions. Inhibition of sporulation was observed in dose dependent manner. S. officinarum extract at higher dose also damaged the normal morphology and shape of oocysts of Eimeria species

    In vitro anticoccidial activity of Trachyspermum ammi (Ajwain) extract on oocysts of Eimeria species of Chicken

    Background: Due to development of synthetic anticoccidial drug resistance there is improvement of anticoccidial medicines due to which exploration of diverse therapeutic agents is attractive now a days. Amongst original agents’ miscellaneous botanicals have shown encouraging properties against coccidiosis. Present study is also a part of probing innovative therapeutic drugs against coccidiosis which can provide replacement solution to treat coccidiosis.Methods: In current experiment in vitro anticoccidial effect of Trachyspermum ammi (seeds) extract was evaluated. For this purpose, an in vitro sporulation inhibition assay was used. Collected oocysts of four Eimeria species were exposed to six different concentrations (w/v) of T. ammi in 10% Dimethyl sulphoxide solution (DMSO), while Dimethyl sulphoxide and Potassium dichromate solution (K2Cr2O7) served as control groups.Results: Results of study revealed that T. ammi extract showed in vitro anticoccidial effect by affecting on sporulation (%) and damaging (%) Eimeria oocysts in dose dependent manner. T. ammi extract also damaged the morphology of oocysts in terms of shape, size and number of sporocysts.Conclusion: The results strongly support the botanicals applications of T. ammi extract and also demonstrate its potential for use in Poultry coccidiosis control strategies.Keywords: Trachyspermum ammi extract; In vitro; Eimeria; Oocyst

    Anticoccidial Effect of Cinnamomum verum Essential Oil and Its Impact on Hematological and Serum Biochemical Parameters in Broilers

    Background: There are various synthetic anticoccidial drugs available in the market for the control and treatment of coccidiosis in broilers. However, their extensive usage has resulted in the development of drug resistance as well as the presence of drug residues in meat, thus urging scientists to find alternatives for coccidiosis control. Hence, the current research was aimed at the evaluation of the anticoccidial potential of Cinnamomum verum essential oil through the application of both the in-vitro and in-vivo methods.Methods: The bark of C. verum procured from the market was subjected to hydro-distillation procedure for extraction of the essential oil. The extracted essential oil was subjected to in-vitro evaluation in terms of percent sporulation and oocysts damage at six different concentrations (0.31, 0.625, 1.25, 2.5, 5 and 10% v/v). Similarly, for the in-vivo trial, 72 broiler chicks were randomly divided into six equal groups (A, B, C, D, E and F). The first five groups were infected with oocysts of mixed Eimeria species while the sixth group was kept as non-infected. When the chicks were 14 days old, the infected groups were orally given 55000 oocysts per bird. On the same day 14, the groups A, B and C were given C. verum essential oil at concentrations of 1, 2 and 3% respectively in feed whereas groups D, E and F served as positive control (Toltrazuril® treated), negative control and the normal control respectively.Result: The results revealed C. verum oil to have an effect on the percent sporulation and oocysts damage. The oil also improved the FCR, lesion score, oocysts score, fecal score and serum biochemical parameters in the treated broilers. However, it had no significant positive effect on the hematological parameters like Hb, PCV and blood cells count, and the weight of internal organs in broilers. For most of the parameters, C. verum essential oil showed a dose-dependent effect.Conclusion: In nutshell, C. verum essential oil possesses significant anticoccidial potential as demonstrated by the results of both the in-vitro and the in-vivo experiments. However, further studies are required for its validation and commercialization in the poultry sector

    Efecto promotor del crecimiento de Pimpinella anisum (semilla de anis) en pollos broiler

    Se realizó el presente estudio para evaluar el efecto promotor del crecimiento de Pimpinella anisum L. (anís) en pollos de  engorde en términos de ganancia de peso corporal, peso de los órganos, el consumo de alimento y conversión alimenticia (FCR). Para este  propósito, cuatro niveles (0,5, 1,0, 1,5 y 2,0 gramos por kg de alimento) de anís en polvo se administraron en el pienso desde el día 0 hasta  el final del experimento (42 días de edad). La ganancia de peso corporal, consumo de alimento y FCR fueron registrados en forma semanal.  Las aves mostraron un buen rendimiento (mejor ganancia de peso, un mejor consumo de alimento y FCR) cuando el anís se alimentó en  dosis más bajas, sin embargo, las aves mostraron malos resultados en los niveles más altos de anís. Del mismo modo, se observó casi el  mismo patrón de peso de los órganos. Este estudio sugiere que el buen efecto del anís en el rendimiento de pollos de engorde en dosis más  bajas que sugiere la necesidad adicional de experimentos de la serie para conocer los niveles de seguridad mínimos de anís que se  incorporarán en la ración de pollos de engorde para obtener los resultados deseados

    Arsenic Induced Toxicity in Broiler Chicks and Its Amelioration with Ascorbic Acid: Clinical, Hematological and Pathological Study

    This study was conducted to observe the arsenic (As) toxicity lesions in birds and to know either Vit C ameliorates these toxic effects or not. One-day-old broilers chicks (n=72) procured from a local hatchery were randomly divided into four equal groups. First group was kept as control and second group was given As (50 mg/kg BW) via crop tubing. Third group received in addition to As, Vit C (250 mg/kg BW) whereas fourth group received only Vit C. Killing by neck dislocation of randomly selected six birds from each group was carried out on experimental days 0, 16 and 32 for collection of blood and tissues specimens. Arsenic treated birds showed clinical signs of toxicity throughout the experiment than all other groups. These clinical signs included decreased body weight and feed intake, dullness, open mouth breathing, increased thirst, ruffled feathers, pale comb, skin irritation and watery diarrhea which were not significant in any other group. As treated group showed a significant (P<0.05) decrease in hematological parameters. Severe gross and histopathological changes were observed in intestines, spleen and lungs of birds fed with As than all other groups. Decreased height of villi of middle portion of small intestines was also observed in As treated birds. Villi height in Vit C treated group increased as compared to control group. It was concluded that As induces severe toxic effects in broiler birds; however, these toxic effects can be partially ameliorated by Vit C

    Babesia bovis in Large Ruminants in Pakistan - Molecular Detection and Haemato-Biochemical Alterations

    Background: Babesiosis is endemic in Pakistan and is one of the most important bovine diseases that causes huge economic losses and high mortality in young animals. A hematobiochemical study was conducted to unveil the difference between diseased and healthy animals in selected districts i.e., Faisalabad (31° 25' 7.3740'' N and 73° 4' 44.7924'' E), Toba Tek Singh (30° 58' 9.7392'' N and 72° 27' 40.7484'' E) and Jhang (31° 16' 40.9656'' N and 72° 18' 42.3360'' E) of Punjab, Pakistan. Materials, Methods &amp; Results: A total of 518 (Cattle = 360, Buffalo = 158) blood samples were collected. The samples were analyzed by polymerase chain reaction (PCR) targeting apocytochrome b-gene (Babesia bovis-gene) (CYTb) followed by haemato-biochemical analysis. Chi-square test for univariate analysis was used to analyze the data. In summer the PCR-based prevalence was 29.4 (53/180) and 24.05% (19/79) in cows and buffaloes, respectively. On the other hand, in winter results showed that 12.7 (23/180), 13.92 % (11/79) samples positive for Babesia genus from cows and buffaloes, respectively. The positive samples were further investigated for hematological and biochemical analysis. The results revealed that, the mean value of hematological parameters like RBCs, Hb, PCV, MCV and MCHC was significantly (P &lt; 0.05) decreased in infected animals (cows and buffaloes) as compared to the non-infected ones. While the biochemical parameters like Alanine aminotransferase (ALT), Aspartate aminotransferase (AST), cholesterol and Lactate dehydrogenase were significantly (P &lt; 0.05) increased in infected animals as compared to healthy animals. This study is the first molecular and hematobiochemical evidence of Babesia bovis in dairy herds of Punjab province, Pakistan. Discussion: Bovine babesiosis is one of the important tick-borne diseases (TBD) affecting dairy industry. In bovines, among 3 Babesia species that cause the disease B. bovis is more pathogenic with high mortality and morbidity. Pakistan is situated in tropical and sub-tropical region where the humidity is high in some part of countries. This high humidity mostly favors the reproduction of the ticks thus higher prevalence of TBDs in this region. Initially the babesiosis was diagnosed by light microscopy using thin blood smear stained with Giemsa stain. Many studies verified that PCR is a more specific and sensitive tool than conventional techniques for the detection of carrier / asymptomatic ruminants. The haemato-biochemical profile is another valuable footprint to track the disease. Keeping in view the above-mentioned fact the present project has been planned to evaluate the haemato-biochemical alteration between health and Babesia infected cattle along with the molecular detection of Babesia species involved in bovine babesiosis. The mean values of haemato-biochemical parameters in clinically ill and healthy animals revealed that the mean values of hematological parameters like RBCs, Hb, PCV, and HCT were significantly decreased in diseased animals as compared to the healthy ones. All these might be due the fact that the parasite is intra-erythrocytic in nature and destruction of red blood cells results in significant (P &lt; 0.05) decrease level of all the hematological parameters. The mean value of ALT in babesiosis infected cattle was significantly higher as compared to healthy cattle. The mean values of AST and LDH in babesiosis infected cows was significantly higher as compared to that in healthy cows. The elevation in liver enzymes in babesiosis may be due to the hepatic damage and lesions induced by the parasite during its multiplication in the blood followed by disturbed liver function. These enzymes are present in high concentrations in the muscles and liver. High level of these enzymes in the blood is indicator of organ necrosis or damage.     Chemo-Modulatory Potential of Flaxseed Oil as Natural Anticancer Therapeutic

    Background: Cancer is a disease which is characterised by uncontrolled cell proliferation and development. Surgery, chemotherapy, radiotherapy, and photodynamic therapy are the most frequent cancer treatments. On the other hand, there are many negative health impacts of radiation and chemotherapy that limit the efficient use of these therapies.Methods: This scenario needs natural treatments that are cost-effective and has no adverse effects. Flaxseed oil can be used as a nutraceuticals for the management of cancer. The current research was aimed on the exploration of the flaxseed (Linum usitatissimum) oil for in vitro anticancer activity as a natural therapy for the management of cancer.Result: The flaxseed powder contained 36.6±0.04% oil contents with an average yield of 36.6±0.03% by using hexane as solvent for extraction. Moreover, the oil contained polyunsaturated fatty acid with omega 3 fatty acid (alpha-linolenic acid) as a dominant content of the oil. In-vitro anticancer activity of flaxseed oil was observed by Cytotoxic (3T3 cell line) and Prostate Cancer (PC3 cell line) indicating that the oil possessed anticancer activity which was dose-dependent.Conclusion: On the basis of results, it was concluded that the flaxseed or its oil can be used for the management of cancer as a natural therapy by using optimized dose levels for a different types of cancer

    Acaricidal and insecticidal effects of essential oils against ectoparasites of veterinary importance

    Ectoparasitism in animals has become an issue of great concern that needs to be resolved to prevent huge economic losses occurring to livestock industry all over the world. Synthetic adrugs have been playing a major role in controlling ectoparasites, but their frequent and irrational use has resulted in drug resistance to routinely used chemicals and their residual effects on food and environment. Therefore, this approach of using chemical acaricides and insecticides is losing its popularity and effectiveness in controlling ectoparasites. So, the development of alternative approaches in ectoparasite management is currently required. Among alternative protocols, plants and their essential oils have played remarkable role in controlling different ectoparasites (ticks, flies, mites, lice) of veterinary importance.     Human Cryptosporidiosis: An insight into Epidemiology, Modern Diagnostic Tools and Recent Drug Discoveries

    Cryptosporidiosis is an emerging food and water borne zoonotic disease, which is caused by genusCryptosporidium. The first Cryptosporidium spp. was isolated from mice in 1907 and gained importance when it was found in an HIV positive patient. It usually causes self-limiting diarrhea in young children and immunocompetent patients. However, it may lead to chronic diarrhea with life threatening condition in immunocompromised patients. Other complications related to this transmittable infection may include respiratory problems, skin rashes and headache. HIV/AIDS patients are highly susceptible host for this parasite. Cryptosporidium parvum and Cryptosporidium hominis are the known pathogenic species, prevalent among humans and they are being transmitted through contaminated food and water. Usually, the diagnosis of Cryptosporidium spp. is dependent on microscopic technique in many countries, which has a low sensitivity and specificity leading to false positive results. However, for a step forward to successful epidemiological studies, advanced techniques (Serological and DNA-based) provide us the better ways of diagnosis with more sensitivity and specificity. Furthermore, no antiparasitic drug has found to be effective againstCryptosporidium spp. except Nitazoxanide which is FDA-approved and effective only when administered along with antiretroviral therapy. In this regard, present review summarizes the various epidemiological studies conducted around the globe along with modern diagnostic tools and the suitable treatment available now a days. This systemized review will help the scientists to better understand all the aspects of cryptosporidiosis at one platform which may help in designing surveillance studies through selection of sensitive diagnostic techniques. The new drugs mentioned in this review may also help to better control this parasite in humans, especially immunocompromised individuals
