A rebel spirit, or, The Pre-Raphaelite movement in the nineteenth century
As an account of the Pre-Raphaelite Movement of the ninteenth century, this paper comprises two aims. It undertakes, on the one hand, to reveal a unique movement for what it actually was, an attempt to combine the media of painting and literature. It attempts, on the other, to illustrate this union by examining the work of Dante Gabriel Rossetti whose painting and poetry best show the direction which the movement took for over three decades. During the search for some understanding of the movement's aim, however, certain factors underlying its development emerged. The movement's origin, for instance, as a reaction against conventional English art in the tradition of Raphael, the aesthetic principles that grew out of this reaction, the movement's relation to Victorian societyâall became important aspects of the total picture and worthy of discussion
An awkward specimen: deconstructing the body and the mental spaces in which it inhabits
The written portion of the thesis is an analysis of the conceptual and aesthetic concerns of a series of artworks exploring the effects and relationships of medical and technological interventions on the body. My M.F.A. thesis installation includes video, sound, interactive sculpture, and wearable performative prosthesis. A repulsive playfulness is the underlying theme of the artwork. By creating these uncomfortable experiences, I hope to make the viewer more aware of their own body
Optimisation of the Schizosaccharomyces pombe urg1 expression system
The ability to study protein function in vivo often relies on systems that regulate the presence and absence of the protein of interest. Two limitations for previously described transcriptional control systems that are used to regulate protein expression in fission yeast are: the time taken for inducing conditions to initiate transcription and the ability to achieve very low basal transcription in the "OFF-state". In previous work, we described a Cre recombination-mediated system that allows the rapid and efficient regulation of any gene of interest by the urg1 promoter, which has a dynamic range of approximately 75-fold and which is induced within 30-60 minutes of uracil addition. In this report we describe easy-to-use and versatile modules that can be exploited to significantly tune down P urg1 "OFF-levels" while maintaining an equivalent dynamic range. We also provide plasmids and tools for combining P urg1 transcriptional control with the auxin degron tag to help maintain a null-like phenotype. We demonstrate the utility of this system by improved regulation of HO-dependent site-specific DSB formation, by the regulation Rtf1-dependent replication fork arrest and by controlling Rhp18(Rad18)-dependent post replication repair
On-site primary care and mental health services in outpatient drug abuse treatment units
Providing health services to drug abuse treatment clients improves their outcomes. Using data from a 1995 national survey of 597 outpatient drug abuse treatment units, this article examines the relationship between these units' organizational features and the degree to which they provided onsite primary care and mental health services. In two-stage models, Joint Commission on Accreditation of Healthcare Organizations-certified and methadone programs delivered more on-site primary care services. Units affiliated with mental health centers provided more on-site mental health services but less direct medical care. Units with more dual-diagnosis clients provided more on-site mental health but fewer on-site HIV/AIDS treatment services. Organizational features appear to influence the degree to which health services are incorporated into drug abuse treatment. Fully integrated care might be an unattainable ideal for many such organizations, but quality improvement across the treatment system might increase the reliability of clients' access to health services.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/45768/1/11414_2005_Article_BF02287796.pd
PCNA ubiquitylation ensures timely completion of unperturbed DNA replication in fission yeast
PCNA ubiquitylation on lysine 164 is required for DNA damage tolerance. In many organisms PCNA is also ubiquitylated in unchallenged S phase but the significance of this has not been established. Using Schizosaccharomyces pombe, we demonstrate that lysine 164 ubiquitylation of PCNA contributes to efficient DNA replication in the absence of DNA damage. Loss of PCNA ubiquitylation manifests most strongly at late replicating regions and increases the frequency of replication gaps. We show that PCNA ubiquitylation increases the proportion of chromatin associated PCNA and the co-immunoprecipitation of Polymerase ÎŽ with PCNA during unperturbed replication and propose that ubiquitylation acts to prolong the chromatin association of these replication proteins to allow the efficient completion of Okazaki fragment synthesis by mediating gap filling
GSK-3ÎČ is essential for physiological electric field-directed Golgi polarization and optimal electrotaxis
Endogenous electrical fields (EFs) at corneal and skin wounds send a powerful signal that directs cell migration during wound healing. This signal therefore may serve as a fundamental regulator directing cell polarization and migration. Very little is known of the intracellular and molecular mechanisms that mediate EF-induced cell polarization and migration. Here, we report that Chinese hamster ovary (CHO) cells show robust directional polarization and migration in a physiological EF (0.3â1Â V/cm) in both dissociated cell culture and monolayer culture. An EF of 0.6Â V/cm completely abolished cell migration into wounds in monolayer culture. An EF of higher strength (â„1Â V/cm) is an overriding guidance cue for cell migration. Application of EF induced quick phosphorylation of glycogen synthase kinase 3ÎČ (GSK-3ÎČ) which reached a peak as early as 3Â min in an EF. Inhibition of protein kinase C (PKC) significantly reduced EF-induced directedness of cell migration initially (in 1â2Â h). Inhibition of GSK-3ÎČ completely abolished EF-induced GA polarization and significantly inhibited the directional cell migration, but at a later time (2â3Â h in an EF). Those results suggest that GSK-3ÎČ is essential for physiological EF-induced Golgi apparatus (GA) polarization and optimal electrotactic cell migration
Managed care and outpatient substance abuse treatment intensity
This study examines the extent to which managed care behavioral controls are associated with treatment intensity in outpatient substance abuse treatment facilities. Data are from the 1995 National Drug Abuse Treatment System Survey, a nationally representative survey that includes over 600 provider organizations with a response rate of 86%. Treatment intensity is measured in three ways: (1) the number of months clients spend in outpatient drug treatment, (2) the number of individual treatment sessions clients receive over the course of treatment, and (3) the number of group treatment sessions clients receive over the course of treatment. After accounting for selection bias and controlling for market, organization, and client characteristics, there is no significant relationship between the scope of managed care oversight and treatment intensity. However, the stringency of managed care oversight activities is negatively associated with the number of individual and group treatment sessions received over the course of treatment.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/45772/1/11414_2005_Article_BF02287231.pd
