    Metabolomic analysis by UAE-GC MS and antioxidant activity of <i>Salvia hispanica </i>(L.) seeds grown under different irrigation regimes

    Chia (Salvia hispanica L.) is an emerging crop with a high content of α-linolenic acid and metabolites of industrial and pharmaceutical interest but information on metabolome variations in response to agricultural management is scarce. We investigated the yield and metabolic profile of the seeds of two chia populations, one commercial black (B) and one long-day flowering genotype (G8), in response to two irrigation levels: replacement of 100% ET⁠0 (I) or rainfed (NI). Seed yield was higher in irrigated plots in G8 only (0.255kgm⁠−2 for I vs 0.184kgm⁠−2 for NI) while it was very low regardless of irrigation in B due to late flowering. Ultrasound assisted extraction (UAE) of seeds followed by gas chromatography-mass spectrometry (GC/MS) analysis showed differences in fatty acids and the major classes of organic compounds due to both genotype and irrigation, especially in the non-polar phase where irrigated samples showed a higher content of α-linolenic and other fatty acids and a lower oleic/linoleic ratio (47.4 in NI vs. 39.6 in I). The antioxidant activity, expressed as trolox equivalent antioxidant capacity (TEAC), ranged from 1.317±0.027 to 2.174±0.010mmol TEAC/g of defatted chia seed after 2 and 40min respectively, and was negatively affected by irrigation. The total polyphenolic content (TPC) measured with the Folin-Ciocalteu method, also decreased with irrigation. According to our results irrigation can affect chia yield, metabolome and antioxidant behavior but some of the effects are genotype-dependent

    Chia is a food plant producing seeds which have seen increasing interest owing to their health benefits. This work is the first report on the metabolite profile, total polyphenols and antioxidant activity of chia seeds, determined by ultrasound-assisted extraction, coupled with gas chromatography-mass spectrometry (UAE GC-MS). Different chia sources were compared: two commercial (black and white) and three early flowering (G3, G8 and G17) mutant genotypes. Organic extracts were mainly composed of mono- and polyunsaturated fatty acids with alpha-linolenic being the most abundant. Polar extracts contained sucrose, methylgalactoside and glucose as main sugars. Antioxidant activity and total polyphenolic content were correlated. Chemical composition and yield potential of early flowering genotypes were different from commercial chia, and while white chia showed the highest content of omega-3 fatty acids, the high content of nutraceuticals in G17 and G8 suggests them as a potential source of raw materials for the food/feed industry

    Bayesian Measurement Error Correction in Structured Additive Distributional Regression with an Application to the Analysis of Sensor Data on Soil-Plant Variability

    The flexibility of the Bayesian approach to account for covariates with measurement error is combined with semiparametric regression models for a class of continuous, discrete and mixed univariate response distributions with potentially all parameters depending on a structured additive predictor. Markov chain Monte Carlo enables a modular and numerically efficient implementation of Bayesian measurement error correction based on the imputation of unobserved error-free covariate values. We allow for very general measurement errors, including correlated replicates with heterogeneous variances. The proposal is first assessed by a simulation trial, then it is applied to the assessment of a soil-plant relationship crucial for implementing efficient agricultural management practices. Observations on multi-depth soil information forage ground-cover for a seven hectares Alfalfa stand in South Italy were obtained using sensors with very refined spatial resolution. Estimating a functional relation between ground-cover and soil with these data involves addressing issues linked to the spatial and temporal misalignment and the large data size. We propose a preliminary spatial interpolation on a lattice covering the field and subsequent analysis by a structured additive distributional regression model accounting for measurement error in the soil covariate. Results are interpreted and commented in connection to possible Alfalfa management strategies

    Rhizosheath: Roles, Formation Processes and Investigation Methods

    Climate change negatively affects crop productivity, threatening the survival of entire populations from many vulnerable hotspot regions of the world with the risk of exacerbating hunger, malnutrition and international inequality. Selecting plant species manifesting abiotic stress-tolerant adaptive traits represents a challenge towards ensuring that crops are more resistant and resilient to environmental perturbations. The rhizosheath, defined as the complex of root hair, exudates and soil that strongly adheres to plant roots, is a promising root adaptive trait in facing conditions of water and nutrient deficits, as well as acidic soil. Several beneficial ecological functions are attributed to the rhizosheath, such as enhancing water and nutrient uptake; protecting from dehydration, heat and acid stresses; and stimulating microbial activities. It has been described in several Angiosperm species, including crops grown in severe habitats. The aim of this review was to collect the relevant literature produced to date regarding rhizosheaths, focusing on (i) the various processes involved in its formation, including both physicochemical and biological ones; (ii) the evolutionary and ecological role of rhizosheaths; and (iii) the most frequently used methodologies for its investigation and characterization. The present work provides a comprehensive overview of this revolutionary root’s great agronomic importance in order to address future research aiming to fill the existing knowledge gaps and define a common and shared methodology

    Metabolomic analysis and antioxidant activity of wild type and mutant Chia (Salvia hispanica L.) stem and flower grown under different irrigation regimes

    BACKGROUND: Chia (Salvia hispanica L.) is a functional food from Central America. Interest in it is growing rapidly due to the many health benefits from the seed. However, when chia is grown at high latitudes, seed yield may be low whereas a high stem biomass and immature inflorescences are produced. Little is known about the chemical composition and the properties of stems and flowers. In this work, the metabolite profile, the antioxidant activity, and the total polyphenol content of stems and inflorescences were evaluated in a factorial experiment with different chia populations (commercial black chia and long-day flowering mutants G3, G8, and G17) and irrigation (100% and 50% of evapotranspiration).RESULTS: The results show the influence of irrigation and seed source on the antioxidant activity and total polyphenol content of chia flower and stem. Inflorescences exhibit higher antioxidant activity, suggesting their potential use as natural antioxidant. The mutants G3 and G8, at 50% irrigation, contained the highest amounts of compounds with nutraceutical value, especially within the flower. The mutant G17 showed lower antioxidant activity and polyphenol content compared to other seed sources but exhibited high omega 3 content in flowers but low in stems. This indicates that chia varieties should be chosen according to the objective of cultivation.CONCLUSION: These findings, indicating a close relation of metabolite content with irrigation and seed source, may provide the basis for the use of chia flower and stem for their nutraceutical value in the food, feed, and supplement industries.</p

    Use of plant growth promoting bacteria (PGDB) for promoting tomato growth and its evaluation as biological control agent.

    The plant growth promoting effect of some bacteria usable to control plant diseases has a potential for resolving technical problems of plant production. The effects of Burkholderia gladioli pv. agaricicola ICMP11096 on development of tomato seedlings development and control of their infection by Fusarium oxysporum and Sclerotinia sclerotiorum were evaluated in the present study. Bacterized plants showed a higher shoot and root weight and a more extended root system in terms of total length and root density than non-bacterized plants. The bacter-izing treatments gave rise to significant reduction of disease symptoms such as leaf wilting induced by F. oxysporum infection and/or root ne-crosis and leaf yellowing by S. sclerotiorum. The potential use of Burkholderia gladioli pv. agaricicola ICMP11096 in biocontrol of fungal patho-gens and to enhance seedling growth parameters is underlined as a tool to achieve a high commercial quality of nursery tomato plants

    Follow-up of the air pollution and the human male-to-female ratio analysis in São Paulo, Brazil: a times series study

    Objectives in order to assess if ambient air pollution in urban areas could be related to alterations in male/female ratio this study objectives to evaluate changes in ambient particulate matter (PM10) concentrations after implementation of pollution control programmes in São Paulo city and the secondary sex ratio (SRR).Design and methods A time series study was conducted. São Paulo's districts were stratified according to the PM10 concentrations levels and were used as a marker of overall air pollution. the male ratio was chosen to represent the secondary sex ratio (SSR=total male birth/total births). the SSR data from each area was analysed according to the time variation and PM10 concentration areas using descriptive statistics. the strength association between annual average of PM10 concentration and SSR was performed through exponential regression, and it was adopted as a statistical significance level of p<0.05.Results the exponential regression showed a negative and significant association between PM10 and SSR. SSR varied from 51.4% to 50.7% in São Paulo in the analysed period (2000-2007). Considering the PM10 average concentration in São Paulo city of 44.72g/m(3) in the study period, the SSR decline reached almost 4.37%, equivalent to 30934 less male births.Conclusions Ambient levels of PM10 are negatively associated with changes in the SSR.     Chia (Salvia hispanica L.) as a novel forage and feed source: A review

    Chia (Salvia hispanica L.), is a traditional pre-Colombian food crop from Central America. Being considered the richest botanical source of omega-3 fatty acids, it has recently been rediscovered as a functional food and feed. A growing body of literature indicates that dietary chia seeds greatly improve animal products quality without compromising growth, productivity and organoleptic quality. Chia is mainly cultivated as a seed crop but recently interest has been raised on biomass production as a potential forage source opening alleys toward the integration of chia in crop-livestock systems. Literature on chia is flourishing, up to now reviews addressed botany, agronomy phytochemical and medicinal uses, this article reviews the main findings on chia use in animal nutrition and includes an overview on both seed and biomass yield and quality as affected by environment, agronomy, and genetic background. Chia is a short-day flowering crop, seed yields of commercial varieties can be as high as 2999 kg ha–1 in areas of origin while at European latitudes seed production is severely hampered by photoperiod sensitivity (max 518 kg ha–1). The viable growing of chia for seeds worldwide relies on the availability of genotypes flowering at longer days than in the areas of origin, while for whole plant a relatively high forage yield can be expected. In southern Italy commercial short-day flowering varieties up to 2.07 t ha–1 of leaf dry biomass and in Greece chia yielded up to 15 T ha−1 dry biomass. Chia seeds supplement in livestock diet are administered with the main objective to increase the content of omega-3 and improve animal health. The majority of work has been done on poultry and rabbits where rewarding results have been obtained in terms of improvement of products lipids profile. Only one work was published on pig but the first results are encouraging. Published data on ruminants are few but in agreement with findings on other species these works demonstrate chia has no adverse effects health performances, and sizeable improvement of milk fatty acid profile. A qualitative improvement of freshwater cultivated fish fillets was also obtained with a partial replacement of soybean oil with chia. Finally an innovative study tested the effect of total or partial replacement of wheat bran in the diets of two edible insects that can be considered the new frontier of food and feed production chains

    Peritumoral adipose tissue promotes lipolysis and white adipocytes browning by paracrine action

    Background: Stromal adipocytes and tumor breast epithelial cells undergo a mutual metabolic adaptation within tumor microenvironment. Therefore, browning and lipolysis occur in cancer associated adipocytes (CAA). However, the paracrine effects of CAA on lipid metabolism and microenvironment remodeling remain poorly understood. Methods: To analyze these changes, we evaluated the effects of factors in conditioned media (CM) derived from explants of human breast adipose tissue from tumor (hATT) or normal (hATN) on morphology, degree of browning, the levels of adiposity, maturity, and lipolytic-related markers in 3T3-L1 white adipocytes by Western blot, indirect immunofluorescence and lipolytic assay. We analyzed subcellular localization of UCP1, perilipin 1 (Plin1), HSL and ATGL in adipocytes incubated with different CM by indirect immunofluorescence. Additionally, we evaluated changes in adipocyte intracellular signal pathways. Results: We found that adipocytes incubated with hATT-CM displayed characteristics that morphologically resembled beige/brown adipocytes with smaller cell size and higher number of small and micro lipid droplets (LDs), with less triglyceride content. Both, hATT-CM and hATN-CM, increased Pref-1, C/EBPβ LIP/LAP ratio, PPARγ, and caveolin 1 expression in white adipocytes. UCP1, PGC1α and TOMM20 increased only in adipocytes that were treated with hATT-CM. Also, hATT-CM increased the levels of Plin1 and HSL, while decreased ATGL. hATT-CM modified the subcellular localization of the lipolytic markers, favoring their relative content around micro-LDs and induced Plin1 segregation. Furthermore, the levels of p-HSL, p-ERK and p-AKT increased in white adipocytes after incubation with hATT-CM. Conclusions: In summary, these findings allow us to conclude that adipocytes attached to the tumor could induce white adipocyte browning and increase lipolysis as a means for endocrine/paracrine signaling. Thus, adipocytes from the tumor microenvironment exhibit an activated phenotype that could have been induced not only by secreted soluble factors from tumor cells but also by paracrine action from other adipocytes present in this microenvironment, suggesting a “domino effect”.Fil: Pagnotta, Priscila Ayelén.
