    CLON: overlay networks and gossip protocols for cloud environments

    Although epidemic or gossip-based multicast is a robust and scalable approach to reliable data dissemination, its inherent redundancy results in high resource consumption on both links and nodes. This problem is aggravated in settings that have costlier or resource constrained links as happens in Cloud Computing infrastructures composed by several interconnected data centers across the globe. The goal of this work is therefore to improve the efficiency of gossip-based reliable multicast by reducing the load imposed on those constrained links. In detail, the proposed clon protocol combines an overlay that gives preference to local links and a dissemination strategy that takes into account locality. Extensive experimental evaluation using a very large number of simulated nodes shows that this results in a reduction of traffic in constrained links by an order of magnitude, while at the same time preserving the resilience properties that make gossip-based protocols so attractive.HP Labs Innovation Research Award, project DC2MS (IRA/CW118736

    First Impact Assessment of Genotoxic Components in the Qatari Marine Environment

    The Arabian Gulf is a semi-enclosed sea with very high evaporation and low discharge rates resulting in extreme saline and thermal conditions. Additionally the system is characterized by a weak hydrodynamic flushing resulting in pollutant build-up over time. As a result, compared to open marine systems, added stress imposed by pollutants is likely to have severe consequences. Qatar has witnessed a rapid expansion in coastal development, linked to its industrial and population growth in recent decades. While economically and socially valuable, the growth comes with an associated environmental cost and Qatar's marine environment now faces many pressures including eutrophication, inputs of domestic sewage, discharge of industrial waste and the resuspension of sediment due to coastal construction. Although the threats pose to biota inhabiting Qatar's marine environment are evident, their extent has yet to be fully assessed. A large percentage of contaminants in the aquatic environment consist of potentially (directly or indirectly) genotoxic, carcinogenic and mutagenic substances. A genotoxin can modify the genetic material at non lethal and non cytotoxic concentrations and has often belated effects which are significantly important at the population and community levels. Genotoxins have particularly high ecotoxicological relevance in situations of chronic exposure to low doses and to multiple contaminants (e.g. in case of PAHs rich tarballs arriving in the shorelines), raising he need to establish genotoxicological profiles of the ecosystems. Indeed several regulatory developments such as: EU - Marine Strategy Framework Directive, the or US - Environmental Protection Agency - Integrated Risk Information System have stressed explicitly on the need of the detection and assessment of potential carcinogenic and mutagenic toxicants using genotoxicity endpoints. The analysis of cytogenetic endpoints in organisms exposed to contaminants in their natural environment contributes significantly to the early detection of genotoxic damage. The relevance of cytogenetic parameters and atypical cytogenetic features, such as numerical chromosomal abnormalities, i.e. aneuploidy, have largely proven their relevance as alerting indicators of poor environmental health and relevant biomarkers for the early detection of environmental stressors. The use of marine invertebrates for in situ environmental assessment is a widely accepted method for identifying risks to the ecosystems. Moreover, at the DNA and chromosome levels they express qualitatively similar types of induced damage to that found in higher organism (e.g. numerical and structural chromosomal aberrations). In this study, we aimed to take a step towards Qatar's marine sustainability by assessing the health status of the marine environment, and providing early alerting symptoms of degradation, by having as specific objectives: i) to measure the levels, in abiotic (water, sediments) and a marine invertebrate model species, of various anthropogenic contaminants (metals, polycyclic aromatic hydrocarbons, (PAHs) and Total polyaromatic hydrocarbons (TPH) at 3 selected sites around the Qatari coast, ii) measure the biological response at the chromosome level, and iii) determine the main drivers of genotoxicity through a multivariate analysis in order to establish a first partial genotoxicological profile of the Qatar Marine Zone. The 3 selected sampling sites, with expected different levels and sources of pollution were: South of Al Khor, Al Wakra harbor and South of Doha harbor. Two sampling campaigns were performed, one in summer and one in winter, to evaluate the role of the abiotic parameters, among others, on the bioavailability of the studied contaminants. The native pearl oyster Pinctada radiata was selected as model and surrogate species due to, its wide distribution along the Qatari coast, filter feeder and sessile mode of life and to its ability as a bivalve to bioaccumulate pollutants. Chemical analyses of the main trace metals and hydrocarbons were performed in water, sediment and P. radiata samples. The evaluation of the aneuploidy levels in P. radiata was estimated in 25-30 animals from each sampling site and season, by counting the total number of aneuploid metaphases over 30 metaphases counted per individual. The evaluation of the aneuploidy level on Pinctada radiata from the three sampling sites revealed an occurrence of significantly higher levels in Al-Wakra harbor (17% in summer and 20% in winter) and South of Doha harbor (19% in summer and 17% in winter), when compared to Al Khor (5% in summer cand 7% in winter). No statistically significant differences were observed between seasons in each location. In order to investigate the discrepancy between sampling sites and seasons with respect to all estimated descriptors and to evaluate the relationship between all the studied parameters, a principal component analysis (PCA) was performed. Aneuploidy levels were highly correlated to mercury and PAHs levels in the bivalve tissue. Moreover, the higher aneuploidy levels registered at Al Wakra harbor (both seasons) and Doha harbor (summer) showed a high correlation with the contaminants levels in P. radiata tissues. South of Al Khor (in both sampling seasons) was highly positively correlated with Cadmium (Cd), although this contamination was not responsible for a significant increase of the aneuploidy levels. The studied genotoxic contaminants were found to be highly variable among considered locations and between sampling seasons. Indeed, the 6 observations (contaminants levels among three sampling sites at two sampling seasons) differ substantially, no site or sampling season grouping being observed, which suggests an important spatial and temporal variability of the bioaccumulation of pollutants into P. radiata tissues. The aneuploidy levels, however, were consistently different among sampled locations, but did not differ between the two sampling seasons, suggesting that aneuploidy is the consequence of a local chronic contamination, and not a direct response to the temporal variability of the contaminants in P. radiata tissues. The results of this study confirm the suitability of the cytogenetic endpoints to discriminate, categorize the studied sites as regards to their level of contamination, underlining the added value of the detection of the genotoxicity levels in the marine environment to environmental health assessment and mitigation research programs. Further studies should be developed, under the specific hydrological and toxicological conditions of the Qatar Marine Zone (QMZ), to better explain the underlining mechanisms of such genotoxicity in the local filter feeders. The establishment of a Genotoxicological profile of the QMZ would be a valuable contribution to a wider approach on environmental diagnosis or prognosis, contributing to the protection and sustainability of the QMZ natural habitats and resources.qscienc

    Methylmercury, Trace Metals, Organotins and Their Effects on the Qatari Mangrove Shrimp, Palaemon khori

    The Qatari mangroves of Al-Khor are being increasingly exposed to a wide variety of anthropogenic pollutants due to land reclamation and urban expansion. In this study, we evaluated the lethal and genotoxic effects of methylmercury, trace metals, and organotins, assessing mortality and aneuploidy levels (abnormal number of chromosomes) in the endemic shrimp Palaemon khori under laboratory conditions. In the experimental design, two different concentrations were used for each family of contaminant (single or combined): an environmental concentration equivalent to the maximum value reported in the environment and a value ten times higher, for a period of eight weeks. Survival decreased significantly when pollutants were administrated in combination, even at environmental concentrations (as shown by Cox proportional hazards ratios): similar levels of mortality would be reached by individual type of pollutants only at ten times the environmental concentration. This critical result, under controlled lab conditions, highlights the importance of monitoring mixtures of contaminant types over single ones in the marine environment. Aneuploidy was reported in all treatments and control ranging from 5% to 19% at week four and from 7% to 21% at week eight. All treatments presented significantly higher aneuploidy levels when compared to the control. However, no significant difference was observed between the two time periods, even though 30% of the treatments could not be assessed at week eight, as not enough animals were still alive. In conclusion, the use of endemic species should be considered a valuable tool to determine local perturbations, representing a regional bioindicator of multiple environmental stressors from the initial stages of contamination

    Prescribing of Non-Steroidal Anti-Inflammatory Drugs to Patients with Diabetes Mellitus in Portugal

    INTRODUCTION: Portugal presents the highest incidence of stage 5 chronic kidney disease in Europe. It is speculated that a high consumption of non-steroidal anti-inflammatory drugs (NSAIDS) may contribute to this high incidence. Our aim was to characterize the prescription of non-steroidal anti-inflammatory drugs to patients with diabetes mellitus in Portugal. MATERIAL AND METHODS: We analyzed the national prescription database in triennium 2015 - 2017. In patients with diabetes mellitus, we evaluated the prescription of non-steroidal anti-inflammatory drugs according to age, gender and region of the patient and specialty of the prescribing physician. We evaluated the prescription of non-steroidal anti-inflammatory drugs in all patients with diabetes mellitus, in patients with presumed renal impairment, and in those with concomitant prescription of angiotensin converting enzyme inhibitors or angiotensin receptor antagonists. RESULTS: We analyzed 23 320 620 prescriptions, corresponding to 610 157 adults, including 104 306 patients with diabetes mellitus. The most prescribed non-steroidal anti-inflammatory drugs were ibuprofen (20.1%), metamizole (14.7%), and diclofenac (11.4%). The prescription of non-steroidal anti-inflammatory drugs was higher in females, in patients aged 51 - 70 years and in the Alentejo region. Non-steroidal anti-inflammatory drugs were prescribed to 70.6% of patients with diabetes mellitus, from which 10.6% were prescribed ≥ 10 packages during the three years. Among patients with diabetes mellitus on angiotensin converting enzyme inhibitors/angiotensin receptor antagonists and with presumed reduction in kidney function, 69.3% were prescribed non-steroidal anti-inflammatory drugs and 11.5% were prescribed ≥ 10 packages during the three years. DISCUSSION: The level of prescribing of non-steroidal anti-inflammatory drugs to patients with diabetes mellitus is high. The concern of reducing non-steroidal anti-inflammatory drugs prescription to patients already on angiotensin converting enzyme inhibitors/angiotensin receptor antagonists and/or decreased renal function does not seem to exist. CONCLUSION: In Portugal, the level of prescribing of non-steroidal anti-inflammatory drugs to patients with diabetes mellitus should be reduced, particularly in the subgroups identified with higher prescription and with higher risk of progression to stage 5 chronic kidney disease.info:eu-repo/semantics/publishedVersio

    The low-virulent African swine fever virus (ASFV/NH/P68) induces enhanced expression and production of relevant regulatory cytokines (IFNα, TNFα and IL12p40) on porcine macrophages in comparison to the highly virulent ASFV/L60

    The impact of infection by the low-virulent ASFV/NH/P68 (NHV) and the highly virulent ASFV/L60 (L60) isolates on porcine macrophages was assessed through the quantification of IFNα, TNFα, IL12p40, TGFβ and ASFV genes by real-time PCR at 2, 4 and 6 h post-infection. Increased IFNα, TNFα and IL12p40 expression was found in infection with NHV, in which expression of TGFβ was lower than in infection with L60. Principal component analysis showed a positive interaction of cytokines involved in cellular immune mechanisms, namely IFNα and IL12p40 in the NHV infection. Quantification by ELISA confirmed higher production of IFNα, TNFα and IL12p40 in the NHV-infected macrophages. Overall, our studies reinforce and clarify the effect of the NHV infection by targeting cellular and cellular-based immune responses relevant for pig survival against ASFV infection

    Two new sponge species (Demospongiae: Chalinidae and Suberitidae) isolated from hyperarid mangroves of Qatar with notes on their potential antibacterial bioactivity.

    This study presents the taxonomic description of two new sponge species that are intimately associated with the hyperarid mangrove ecosystem of Qatar. The study includes a preliminary evaluation of the sponges' potential bioactivity against pathogens. Chalinula qatari sp. nov. is a fragile thinly encrusting sponge with a vivid maroon colour in life, often with oscular chimneys and commonly recorded on pneumatophores in the intertidal and shallow subtidal zone. Suberites luna sp. nov. is a massive globular-lobate sponge with a greenish-black colour externally and a yellowish orange colour internally, recorded on pneumatophores in the shallow subtidal zone, with large specimens near the seagrass ecosystem that surrounds the mangrove. For both species, a drug extraction protocol and an antibacterial experiment was performed. The extract of Suberites luna sp. nov. was found to be bioactive against recognized pathogens such as Staphylococcus epidermidis, Staphylococcus aureus and Enterococcus faecalis, but no bioactive activity was recorded for Chalinula qatari sp. nov. This study highlights the importance of increasing bioprospecting effort in hyperarid conditions and the importance of combining bioprospecting with taxonomic studies for the identification of novel marine drugs.QNRF/URE

    Expanding ocean protection and peace: a window for science diplomacy in the Gulf.

    The ecological state of the Persian or Arabian Gulf (hereafter 'Gulf') is in sharp decline. Calls for comprehensive ecosystem-based management approaches and transboundary conservation have gone largely unanswered, despite mounting marine threats made worse by climate change. The region's long-standing political tensions add additional complexity, especially now as some Gulf countries will soon adopt ambitious goals to protect their marine environments as part of new global environmental commitments. The recent interest in global commitments comes at a time when diplomatic relations among all Gulf countries are improving. There is a window of opportunity for Gulf countries to meet global marine biodiversity conservation commitments, but only if scientists engage in peer-to-peer diplomacy to build trust, share knowledge and strategize marine conservation options across boundaries. The Gulf region needs more ocean diplomacy and coordination; just as critically, it needs actors at its science-policy interface to find better ways of adapting cooperative models to fit its unique marine environment, political context and culture. We propose a practical agenda for scientist-led diplomacy in the short term and lines of research from which to draw (e.g. co-production, knowledge exchange) to better design future science diplomacy practices and processes suited to the Gulf's setting.We acknowledge support from the Smithson Fellowship (C.M.F.)
