    New Insights into the Bacterial Fitness-Associated Mechanisms Revealed by the Characterization of Large Plasmids of an Avian Pathogenic E. coli

    Extra-intestinal pathogenic E. coli (ExPEC), including avian pathogenic E. coli (APEC), pose a considerable threat to both human and animal health, with illness causing substantial economic loss. APEC strain χ7122 (O78∶K80∶H9), containing three large plasmids [pChi7122-1 (IncFIB/FIIA-FIC), pChi7122-2 (IncFII), and pChi7122-3 (IncI(2))]; and a small plasmid pChi7122-4 (ColE2-like), has been used for many years as a model strain to study the molecular mechanisms of ExPEC pathogenicity and zoonotic potential. We previously sequenced and characterized the plasmid pChi7122-1 and determined its importance in systemic APEC infection; however the roles of the other pChi7122 plasmids were still ambiguous. Herein we present the sequence of the remaining pChi7122 plasmids, confirming that pChi7122-2 and pChi7122-3 encode an ABC iron transport system (eitABCD) and a putative type IV fimbriae respectively, whereas pChi7122-4 is a cryptic plasmid. New features were also identified, including a gene cluster on pChi7122-2 that is not present in other E. coli strains but is found in Salmonella serovars and is predicted to encode the sugars catabolic pathways. In vitro evaluation of the APEC χ7122 derivative strains with the three large plasmids, either individually or in combinations, provided new insights into the role of plasmids in biofilm formation, bile and acid tolerance, and the interaction of E. coli strains with 3-D cultures of intestinal epithelial cells. In this study, we show that the nature and combinations of plasmids, as well as the background of the host strains, have an effect on these phenomena. Our data reveal new insights into the role of extra-chromosomal sequences in fitness and diversity of ExPEC in their phenotypes

    A global experiment on motivating social distancing during the COVID-19 pandemic

    Significance Communicating in ways that motivate engagement in social distancing remains a critical global public health priority during the COVID-19 pandemic. This study tested motivational qualities of messages about social distancing (those that promoted choice and agency vs. those that were forceful and shaming) in 25,718 people in 89 countries. The autonomy-supportive message decreased feelings of defying social distancing recommendations relative to the controlling message, and the controlling message increased controlled motivation, a less effective form of motivation, relative to no message. Message type did not impact intentions to socially distance, but people’s existing motivations were related to intentions. Findings were generalizable across a geographically diverse sample and may inform public health communication strategies in this and future global health emergencies. Abstract Finding communication strategies that effectively motivate social distancing continues to be a global public health priority during the COVID-19 pandemic. This cross-country, preregistered experiment (n = 25,718 from 89 countries) tested hypotheses concerning generalizable positive and negative outcomes of social distancing messages that promoted personal agency and reflective choices (i.e., an autonomy-supportive message) or were restrictive and shaming (i.e., a controlling message) compared with no message at all. Results partially supported experimental hypotheses in that the controlling message increased controlled motivation (a poorly internalized form of motivation relying on shame, guilt, and fear of social consequences) relative to no message. On the other hand, the autonomy-supportive message lowered feelings of defiance compared with the controlling message, but the controlling message did not differ from receiving no message at all. Unexpectedly, messages did not influence autonomous motivation (a highly internalized form of motivation relying on one’s core values) or behavioral intentions. Results supported hypothesized associations between people’s existing autonomous and controlled motivations and self-reported behavioral intentions to engage in social distancing. Controlled motivation was associated with more defiance and less long-term behavioral intention to engage in social distancing, whereas autonomous motivation was associated with less defiance and more short- and long-term intentions to social distance. Overall, this work highlights the potential harm of using shaming and pressuring language in public health communication, with implications for the current and future global health challenges

    A predator has nonconsumptive effects on different life-history stages of a prey

    Through a field experiment, we show that a predator has negative nonconsumptive effects (NCEs) on different life-history stages of the same prey species. Shortly before the recruitment season of the barnacle Semibalanus balanoides (spring), we established experimental cages in rocky intertidal habitats in Nova Scotia, Canada. The cages were used to manipulate the presence and absence of dogwhelks, Nucella lapillus, the main predators of barnacles. At the centre of each cage, we installed a tile where barnacle pelagic larvae could settle and the resulting recruits grow. Mesh prevented caged dogwhelks from accessing the tiles, but allowed waterborne dogwhelk cues to reach the tiles. During the recruitment season, barnacle larvae settled preferentially on tiles from cages without dogwhelks. In the fall, at the end of the dogwhelk activity period and once the barnacle recruits had grown to adult size, barnacle body mass was lower in the presence of dogwhelks. This limitation may have resulted from a lower barnacle feeding activity with nearby dogwhelks, as found in a previous study. The observed larval and adult responses in barnacles are consistent with attempts to decrease predation risk. In the fall, dogwhelk cues also limited barnacle reproductive output, a possible consequence of the limited growth of barnacles. Overall, the results of this study suggest that a predator might influence trait evolution in a prey mediated by NCEs on different life-history stages

    Algal turf consumption by sea urchins and fishes is mediated by fisheries management on coral reefs in Kenya

    Herbivory is a key process that controls the abundance and accumulation of algal turf on tropical coral reefs. The capacity of reefs to prevent algal accumulation hinges on the balance between algal production and consumption (i.e., grazing). In this study, we quantify algal turf biomass accumulation and grazing using experimental substrata and herbivore exclusion cages across sites in Kenya that represent different levels of fisheries management: heavily fished reefs, community marine-protected areas less than 10 yr old, and older government-managed marine-protected areas. These reefs had different assemblages of grazing herbivores with fished reefs being dominated by sea urchins, while government closures had a high abundance of grazing fishes, in particular parrotfishes. The community fisheries closures had an intermediate mix of sea urchins and grazing fishes, with the latter dominated by surgeonfishes. These management regimes mediated algal biomass on experimental substrata such that urchins consumed as much as 90% on fished reefs and fishes as much as 96% at the government marine-protected areas by the end of the 390-d trial. The younger community fisheries closures lacked the herbivory to significantly reduce algal biomass, and consumption was less than 50% of production and never greater than 2 g algae m−2 d−1. These findings point to the importance of recovery dynamics of herbivorous fishes from heavy fishing pressure. They also suggest that while sea urchins might be effective grazers to prevent macroalgal dominance, they are not a functional replacement for fishes due to their ability to reduce reef accretion through bioerosion and prevent settlement of crustose coralline algae in this system
