    A general approach to high-yield biosynthesis of chimeric RNAs bearing various types of functional small RNAs for broad applications.

    RNA research and therapy relies primarily on synthetic RNAs. We employed recombinant RNA technology toward large-scale production of pre-miRNA agents in bacteria, but found the majority of target RNAs were not or negligibly expressed. We thus developed a novel strategy to achieve consistent high-yield biosynthesis of chimeric RNAs carrying various small RNAs (e.g. miRNAs, siRNAs and RNA aptamers), which was based upon an optimal noncoding RNA scaffold (OnRS) derived from tRNA fusion pre-miR-34a (tRNA/mir-34a). Multi-milligrams of chimeric RNAs (e.g. OnRS/miR-124, OnRS/GFP-siRNA, OnRS/Neg (scrambled RNA) and OnRS/MGA (malachite green aptamer)) were readily obtained from 1 l bacterial culture. Deep sequencing analyses revealed that mature miR-124 and target GFP-siRNA were selectively released from chimeric RNAs in human cells. Consequently, OnRS/miR-124 was active in suppressing miR-124 target gene expression and controlling cellular processes, and OnRS/GFP-siRNA was effective in knocking down GFP mRNA levels and fluorescent intensity in ES-2/GFP cells and GFP-transgenic mice. Furthermore, the OnRS/MGA sensor offered a specific strong fluorescence upon binding MG, which was utilized as label-free substrate to accurately determine serum RNase activities in pancreatic cancer patients. These results demonstrate that OnRS-based bioengineering is a common, robust and versatile strategy to assemble various types of small RNAs for broad applications

    Statistical regularities across trials bias attentional selection

    Previous studies have shown that attentional selection can be biased toward locations that are likely to contain a target and away from locations that are likely to contain a distractor. It is assumed that through statistical learning, participants are able to extract the regularities in the display, which in turn biases attentional selection. The present study employed the additional singleton task to examine the ability of participants to extract regularities that occurred across trials. In four experiments, we found that participants were capable of picking up statistical regularities concerning target positions across trials both in the absence and presence of distracting information. It is concluded that through statistical learning, participants are able to extract intertrial statistical associations regarding subsequent target location, which in turn biases attentional selection. We argue here that the weights within the spatial priority map can be dynamically adapted from trial to trial such that the selection of a target at a particular location increases the weights of the upcoming target location within the spatial priority map, giving rise to a more efficient target selection

    A Statistical Study on Force-Freeness of Solar Magnetic Fields in the Photosphere

    It is an indisputable fact that solar magnetic fields are force-free in the corona, where force free fields means that current and magnetic fields are parallel and there is no Lorentz force in the fields. While the force-free extent of photospheric magnetic fields remains open. In this paper, the statistical results about it is given. The vector magnetograms (namely, BxB_{x}, ByB_{y} and BzB_{z} in heliocentric coordinates) are employed, which are deduced and calibrated from Stokes spectra, observed by Solar Magnetic Field Telescope (SMFT) at Huairou Solar Observing Station (HSOS) are used. We study and calibrated 925 magnetograms calibrated by two sets of calibration coefficients, that indicate the relations between magnetic fields and the strength of Stokes spectrum and can be calculated either theoretically or empirically. The statistical results show that the majority of active region magnetic fields are not consistent with the force-free model.Comment: 10 pages, 5 figures, has been accepted by PAS

    The authors have obtained the following results: (1) the definition of uniformly closed countable family of nonlinear mappings, (2) strong convergence theorem by the monotone hybrid algorithm for two countable families of hemirelatively nonexpansive mappings in a Banach space with new method of proof, (3) two examples of uniformly closed countable families of nonlinear mappings and applications, (4) an example which is hemirelatively nonexpansive mapping but not weak relatively nonexpansive mapping, and (5) an example which is weak relatively nonexpansive mapping but not relatively nonexpansive mapping. Therefore, the results of this paper improve and extend the results of Plubtieng and Ungchittrakool (2010) and many others
