Untersuchungen zum Einfluss des Wurzelendophyten Piriformospora indica auf das Wachstum von Hordeum vulgare, die Resistenz gegen Blumeria graminis f.sp. hordei und die Genexpression in den BlÀttern
Der Wurzelendophyt Piriformospora indica wurde 1997 von der Spore eines AM-Pilzes in der indischen HalbwĂŒste Thar isoliert. Wachstumsfördernde Eigenschaften dieses Endophyten konnten bei verschiedenen Pflanzenarten beobachtet werden. Ziel der vorliegenden Arbeit war es, die Wechselwirkungen von P. indica mit Hordeum vulgare nĂ€her zu untersuchen und systemische Wirkungen auf den Spross zu charakterisieren.
Die Besiedelung von Wurzeln mit P. indica fĂŒhrte im GewĂ€chshaus zu einer signifikanten Förderung des Pflanzenwachstums. In FreilandgefĂ€Ăexperimenten zeigte sich ein frĂŒheres Schieben der Ăhren, eine erhöhte Ăhrenanzahl, sowie eine Ertragsteigerung von drei verschiedenen Gerstensorten zwischen 1,9 und 11 Prozent. Hinweise auf eine erhöhte Photosyntheserate und einen erhöhten Chlorophyllgehalt in den BlĂ€ttern besiedelter Pflanzen, die sich aus der Analyse einer subtraktiven cDNA Bank ergaben, konnten bestĂ€tigt werden.
In weiteren Versuchen wurde der Einfluss der Besiedelung auf die Resistenz der Pflanzen gegen das biotrophe Blattpathogen Blumeria graminis f. sp. hordei (Bgh) untersucht. Makroskopisch zeigte sich ein erhöhter Resistenzstatus der besiedelten Pflanzen, der auf der Basis von zytologischen Experimenten auf eine gröĂere Penetrationsresistenz zurĂŒckgefĂŒhrt werden konnte. Die beobachtete induzierte Resistenz war mit einer erhöhten Bereitschaft fĂŒr die schnelle und effektive Aktivierung der zellulĂ€ren Verteidigungsantwort assoziiert. Erste Genexpressionsanalysen zeigten, dass bekannte Wege systemisch induzierter Resistenzen hierbei nicht beteiligt zu sein scheinen, und dass die Aktivierung des antioxidativen Systems aber eine wichtige Rolle spielen könnte. Durch Hybridisierungen eines cDNA-Makroarrays und des 22K Barley1 GeneChipÂź (Affymetrix, Santa Clara, USA) wurden in BlĂ€ttern P. indica besiedelter Pflanzen nur verhĂ€ltnismĂ€Ăig wenig differentiell exprimierte Gene identifiziert. AnschlieĂende RealTime-PCR-Expressionsstudien bestĂ€tigten eine stĂ€rkere Induktion von zwei putativen Rezeptorkinasen in BlĂ€ttern P. indica-besiedelter Pflanzen, 12h nach Bgh-Inokulation, die fĂŒr Komponenten der Signalrezeption und Signaltransduktion kodieren. Dies deutet auf eine frĂŒhere Erkennung des Pathogens als Basis fĂŒr die P. indica-induzierte Resistenz hin
Spatially-Correlated Microstructure and Superconductivity in Polycrystalline Boron-Doped Diamond
Scanning tunneling spectroscopies are performed below 100~mK on
nano-crystalline boron-doped diamond films characterized by Transmission
Electron Microscopy and transport measurements. We demonstrate a strong
correlation between the local superconductivity strength and the granular
structure of the films. The study of the spectral shape, amplitude and
temperature dependence of the superconductivity gap enables us to differentiate
intrinsically superconducting grains that follow the BCS model, from grains
showing a different behavior involving the superconducting proximity effect
Non-adiabatic Kohn Anomaly in Heavily Boron-doped Diamond
We report evidence of a non-adiabatic Kohn anomaly in boron-doped diamond,
using a joint theoretical and experimental analysis of the phonon dispersion
relations. We demonstrate that standard calculations of phonons using density
functional perturbation theory are unable to reproduce the dispersion relations
of the high-energy phonons measured by high-resolution inelastic x-ray
scattering. On the contrary, by taking into account non-adiabatic effects
within a many-body field-theoretic framework, we obtain excellent agreement
with our experimental data. This result indicates a breakdown of the
Born-Oppenheimer approximation in the phonon dispersion relations of
boron-doped diamond.Comment: 6 pages, 2 figure
Doping-induced metal-insulator transition in aluminum-doped 4H silicon carbide
International audienceWe report an experimental determination of the doping-induced metal-insulator transition in aluminum-doped 4H silicon carbide. Low temperature transport measurements down to 360 mK and temperature dependent Raman experiments down to 5 K, together with secondary ion mass spectroscopy profiling, suggest a critical aluminum concentration lying between 6.4 and 8.7 1020 cmâ3 for the metal-insulator transition in these epilayers grown by the vapor-liquid-solid technique. Preliminary indications of a superconducting transition in the metallic sample are presented
Metal-to-insulator transition and superconductivity in boron-doped diamond
International audienceThe experimental discovery of superconductivity in boron-doped diamond came as a major surprise to both the diamond and the superconducting materials communities. The main experimental results obtained since then on single-crystal diamond epilayers are reviewed and applied to calculations, and some open questions are identified. The critical doping of the metal-to-insulator transition (MIT) was found to coincide with that necessary for superconductivity to occur. Some of the critical exponents of the MIT were determined and superconducting diamond was found to follow a conventional type II behaviour in the dirty limit, with relatively high critical temperature values quite close to the doping-induced insulator-to-metal transition. This could indicate that on the metallic side both the electron-phonon coupling and the screening parameter depend on the boron concentration. In our view, doped diamond is a potential model system for the study of electronic phase transitions and a stimulating example for other semiconductors such as germanium and silicon
Low-temperature transport in highly boron-doped nanocrystalline diamond
International audienceWe studied the transport properties of highly boron-doped nanocrystalline diamond thin films at temperatures down to 50 mK. The system undergoes a doping-induced metal-insulator transition with an interplay between intergranular conductance g and intragranular conductance g0, as expected for a granular system. The conduction mechanism in the case of the low-conductivity films close to the metal-insulator transition has a temperature dependence similar to Efros-Shklovskii type of hopping. On the metallic side of the transition, in the normal state, a logarithmic temperature dependence of the conductivity is observed, as expected for a metallic granular system. Metallic samples far away from the transition show similarities to heavily borondoped single-crystal diamond. Close to the transition, the behavior is richer. Global phase coherence leads in both cases to superconductivity also checked by ac susceptibility , but a peak in the low-temperature magnetoresistance measurements occurs for samples close to the transition. Corrections to the conductance according to superconducting fluctuations account for this negative magnetoresistance

Superconductivity has been discovered in case of heavily boron-doped single crystal cubic silicon (prepared by GILD), the MIT in the case of highly aluminium-doped 4H silicon carbide (prepared by VLS). The study of the MIT and the magnetotransport in highly boron-doped nanocrystalline diamond has shown the importance of the granularity in this system.La transition métal-isolant (TMI) induite par le dopage et la supraconductivité ont été étudiées dans le diamant monocristallin fortement dopé au bore (propriétés structurales, approche en loi d'échelle pour la TMI, substitution isotopique, structure de bande vs effet Hall, mode de vibration des dimÚres de bore) et des matériaux voisins. La supraconductivité a été découverte pour le silicium monocristallin fortement dopé au bore (préparé par la méthode GILD), la TMI dans le cas du carbure de silicium 4H fortement dopé à l'aluminium (préparé par la méthode VLS).
