    A Comparison of Staining Techniques for Somatic Chromosomes of Strawberry

    A factorial combination of pretreatments, fixatives, and stains was examined to identify the best available method for staining the chromosomes of strawberry (Fragaria × ananassa Duch. ‘Chandler’) root-tip cells. Three pretreatments ( a -bromonaphthalene, p -dichlorobenzene, and 8-hydroxyquinoline), three fixatives (Farmer’s fluid, Carnoy’s fluid, and Newcomer’s fluid), and five stains (acetocarmine, alcoholic hydrochloric-acid carmine, altered carbol fuchsin, lacto-propionic orcein, and leucobasic fuchsin) were tested. Pretreatment with either a -bromonaphthalene (saturated aqueous) or 8-hydroxyquinoline (2 mM) for 5 hours at 14C, overnight fixation in Farmer’s fluid, hydrolysis in 1 N HCl (15 minutes at 60C), and staining with altered carbol fuchsin produced chromosome preparations superior to other treatment combinations. Treatment with a-bromonaphthalene, Farmer’s fluid, and alcoholic hydrochloric-acid carmine (2 days at 25C) also produced acceptable chromosome preparations

    A study was conducted to maxmuze plant regeneration frequencies from cultured anthers of\u27 Chandler\u27, \u27Honeoye\u27, and \u27Redchief strawberries (Fragaria x ananassa Duch.). A comparison of auxins (IAA, NAA), cytokinins (BA, BPA, KIN) and carbohydrates (sucrose, glucose, maltose) in MS medium showed that the highest shoot regeneration across cultivars (8%) occurred when using a medium containing 2 mgtl IAA, I mgtl BA, and 0.2 M glucose. A comparison ofMS, NN, and HI inorganic medium (a new formulation based on the anther culture literature) solidified with either agar or gellan gum and containing IAA, BA, and glucose, showed the highest shoot regeneration across cultivars (19%) when using HI and gellan gum. Lastly, media containing Fe-EDTAyielded more shoots than media containing Fe-Metalosate, and anthers cultured on Fe-EDT A media in darkness for 30d followed by 30d in white light produced more shoots (16% average regeneration) than those cultured on Fe-EDTA media under white or yellow light (16h photoperiod) for the initial 30d (0.3% and 5% respectively). Plants were acclimated ex vitro where they flowered and set :fruit. Chromosome counts of root tip cells confirmed that haploid plants were obtained from all three cultivar

    Culture medium pH is influenced by basal medium, carbohydrate source, gelling agent, activated charcoal, and medium storage method

    When four carbohydrates were tested against six commonly cited inorganic basal media, post-autoclave pH was highest for carbohydrate-free and sucrose- containing media, and progressively lower for maltose-, P lucose-, and fructose-containing media, respectively, post-autoclave pH for these media without carbohydrates was related to medium buffering capacity. Addition of gelling agents (10 of 11 tested) increased the post¬autoclave pH of MS medium containing sucrose. Neutralized and acid-washed activated charcoal also increased the post-autoclave pH of liquid and agar- solidified MS medium, and the pH changed further during 8 weeks of storage. Changes in medium pH caused by gelling agents, but not charcoal, could be alleviated by adjusting the pH after their addition but prior to autoclaving

    Neural Correlates of Competing Fear Behaviors Evoked by an Innately Aversive Stimulus

    Environment and experience influence defensive behaviors, but the neural circuits mediating such effects are not well understood. We describe a new experimental model in which either flight or freezing reactions can be elicited from mice by innately aversive ultrasound. Flight and freezing are negatively correlated, suggesting a competition between fear motor systems. An unfamiliar environment or a previous aversive event, moreover, can alter the balance between these behaviors. To identify potential circuits controlling this competition, global activity patterns in the whole brain were surveyed in an unbiased manner by c-fos in situ hybridization, using novel experimental and analytical methods. Mice predominantly displaying freezing behavior had preferential neural activity in the lateral septum ventral and several medial and periventricular hypothalamic nuclei, whereas mice predominantly displaying flight had more activity in cortical, amygdalar, and striatal motor areas, the dorsolateral posterior zone of the hypothalamus, and the vertical limb of the diagonal band. These complementary patterns of c-fos induction, taken together with known connections between these structures, suggest ways in which the brain may mediate the balance between these opponent defensive behaviors

    Suzaku Observations of the Black Hole H1743-322 in Outburst

    We observed the Galactic black hole candidate H1743-322 with Suzaku for approximately 32 ksec, while the source was in a low/hard state during its 2008 outburst. We collected and analyzed the data with the HXD/PIN, HXD/GSO and XIS cameras spanning the energy range from 0.7-200 keV. Fits to the spectra with simple models fail to detect narrow Fe XXV and Fe XXVI absorption lines, with 90% confidence upper limits of 3.5 eV and 2.5 eV on the equivalent width, respectively. These limits are commensurate with those in the very high state, but are well below the equivalent widths of lines detected in the high/soft state, suggesting that disk winds are partially state-dependent. We discuss these results in the context of previous detections of ionized Fe absorption lines in H1743-322 and connections to winds and jets in accreting systems. Additionally, we report the possible detection of disk reflection features, including an Fe K emission line.Comment: 16 pages, 4 figures, 4 tables. Accepted for publication in ApJ
