Ulnar dimelia variant: a case report
We report a case of ulnar dimelia, commonly called mirror hand, in a 2-month-old female child who had restriction of elbow flexion and forearm rotation. There was no facial or other internal organ malformation. Radiographs revealed seven triphalangeal digits with double ulnae (one following the other) and absent radius. To the best of the authors’ knowledge, this is the first report of this mirror hand deformity in which fingers are symmetrical while duplicated ulnae are not
Pyothorax in a cat due to Acinetobacter spp. infection : a case report
A 3-month-old kitten weighing 1.05 kg was presented to the University veterinary hospital at Kokkalai in Thrissur district of Kerala, India, with the complaint of open-mouth breathing, lethargy and complete anorexia for the past three days, and weight loss. Detailed clinical examination revealed inspiratory dyspnoea without stridor, tachypnoea, muffled lung and heart sounds on auscultation, pale mucous membrane, and weak pulse. Haematology revealed leukocytosis with granulocytosis and monocytosis. Thoracic radiography, ultrasonography and thoracocentesis confirmed pyothorax. Microbial culture of thoracic aspirate revealed heavy growth of Acinetobacte spp. The cat responded to parenteral therapy with ceftiofur followed by cefixime orally for 21 days. Doxycycline was prescribed for concurrent mycoplasmosis. Thoracic radiograph 10 days later revealed a significant reduction in pleural fluid. A review of the case two months later found that the kitten recovered uneventfully. A rare case of Acinetobacter spp. associated pyothorax and its medical management in a cat is reported. Burnout among surgeons before and during the SARS-CoV-2 pandemic: an international survey
Background: SARS-CoV-2 pandemic has had many significant impacts within the surgical realm, and surgeons have been obligated to reconsider almost every aspect of daily clinical practice. Methods: This is a cross-sectional study reported in compliance with the CHERRIES guidelines and conducted through an online platform from June 14th to July 15th, 2020. The primary outcome was the burden of burnout during the pandemic indicated by the validated Shirom-Melamed Burnout Measure. Results: Nine hundred fifty-four surgeons completed the survey. The median length of practice was 10 years; 78.2% included were male with a median age of 37 years old, 39.5% were consultants, 68.9% were general surgeons, and 55.7% were affiliated with an academic institution. Overall, there was a significant increase in the mean burnout score during the pandemic; longer years of practice and older age were significantly associated with less burnout. There were significant reductions in the median number of outpatient visits, operated cases, on-call hours, emergency visits, and research work, so, 48.2% of respondents felt that the training resources were insufficient. The majority (81.3%) of respondents reported that their hospitals were included in the management of COVID-19, 66.5% felt their roles had been minimized; 41% were asked to assist in non-surgical medical practices, and 37.6% of respondents were included in COVID-19 management. Conclusions: There was a significant burnout among trainees. Almost all aspects of clinical and research activities were affected with a significant reduction in the volume of research, outpatient clinic visits, surgical procedures, on-call hours, and emergency cases hindering the training. Emphysematous spontaneous bacterial peritonitis and renal azotemia due to Escherichia coli infection in a Saint Bernard dog
A two-and-a-half-year-old intact male Saint Bernard dog weighing 44 kg was presented to the University Veterinary Hospital (KVASU), Kokkalai, Thrissur, with a history of vomiting, diarrhoea, and inappetence for the past two days. Clinical examination revealed a distended abdomen, with presence of fluid suspected on ballottement. Abdominal ultrasonography revealed mixed echogenic fluid in the peritoneal cavity, gas shadowing, comet-tail artefact and hyperechoic foci with dirty shadowing. Peritoneal fluid was creamy-white in colour on gross examination. Peritoneal fluid triglyceride was normal whereas serum creatinine and ALP were elevated. Microbial culture yielded growth of organisms which was confirmed as Escherichia coli. This article reports a case of emphysematous spontaneous bacterial peritonitis and renal azotaemia due to E. coli infection in a dog. The animal had an eventful recovery
Cryopreserved ovine spermatogonial stem cells maintain stemness and colony forming ability in vitro
Objective: To assess the effect of cryopreservation on stemness and proliferation potential of sheep spermatogonial stem cells (SSCs) in vitro.
Methods: Sheep testicular cells were isolated and putative SSCs were enriched by the laminin-based differential plating method. Putative SSCs were co-cultured with the Sertoli cell feeder prepared by the Datura Stramonium Agglutinin (DSA-lectin)-based method. The cultured putative SSCs were cryopreserved in Dulbecco's Modified Eagle Medium-10% fetal bovine serum mixture (DMEM-10% FBS) media containing 10% dimethyl sulfoxide (DMSO) alone or 10% DMSO plus 200 mM trehalose. Cryopreserved putative SSCs were evaluated for their proliferation potential using in vitro culture and stemness by immunocytochemistry. Finally, the transfection ability of cryopreserved putative SSCs was analyzed.
Results: We isolated 91% viable testicular cells from sheep testes. The majority of the laminin enriched cells expressed the SSC related marker, ITGA6. Co-culture of sheep putative SSCs with Sertoli cell feeder resulted in the generation of stable colonies, and the expression of SSC marker was maintained after several passages. A significantly higher number of viable putative SSCs was recovered from SSCs cryopreserved in media containing 10% DMSO and 200 mM trehalose compared to 10% DMSO alone (P<0.01). Cryopreserved putative SSCs formed colonies and showed SSC marker expression similar to the non-cryopreserved putative SSCs. The appearance of green fluorescent colonies over the Sertoli cell feeder indicated that cryopreserved sheep SSCs were successfully transfected.
Conclusions: Cryopreserved putative SSCs can retain their stemness, colony forming ability, and transfection efficiency in vitro. Our research may help in the effective preservation of germplasm and the generation of transgenic ovine species
Biofabrication of Zinc Oxide Nanoparticle from Ochradenus baccatus
Biofilms are complex aggregation of cells that are embedded in EPS matrix. These microcolonies are highly resistant to drugs and are associated with various diseases. Biofilms have greatly affected the food safety by causing severe losses due to food contamination and spoilage. Therefore, novel antibiofilm agents are needed. This study investigates the antibiofilm and protein binding activity of zinc nanoparticles (ZnNPs) synthesized from leaf extract of Ochradenus baccatus. Standard physical techniques, including UV-visible spectroscopy Fourier transform infrared spectroscopy and X-ray diffraction and transmission electron microscopy, were used to characterize the synthesized OB-ZnNPs. Synthesized OB-ZnNPs demonstrated significant biofilm inhibition in human and food-borne pathogens (Chromobacterium violaceum, Escherichia coli, P. aeruginosa, Klebsiella pneumoniae, Serratia marcescens, and Listeria monocytogenes) at subinhibitory concentrations. OB-ZnNPs significantly reduced the virulence factors like violacein, prodigiosin, and alginate and impaired swarming migration and EPS production. OB-ZnNPs demonstrated efficient binding with HSA protein and no change in their structure or stability was observed. In addition, in vivo toxicity evaluation confirmed that OB-ZnNPs possessed no serious toxic effect even at higher doses. Moreover, they were found to have excellent antioxidant properties that can be employed in the fields of food safety and medicine. Hence, it is envisaged that the OB-ZnNPs can be used as potential nanomaterials to combat drug resistant bacterial infections and prevent contamination/spoilage of food
