    Immunotherapy of Peritoneal Carcinomatosis with the Antibody Catumaxomab in Colon, Gastric, or Pancreatic Cancer: An Open-Label, Multicenter, Phase I/II Trial

    Background: Peritoneal carcinomatosis (PC) is common in gastrointestinal (GI) cancer and there is no effective standard treatment. We investigated the tolerability and maximum tolerated dose (MTD) of the trifunctional antibody catumaxomab in patients with PC. Methods: In this open-label, phase I/II clinical trial, patients with epithelial cell adhesion molecule (EpCAM)-positive PC from GI cancer received 4 sequential intraperitoneal catumaxomab infusions: day 0: 10 mu g; day 3: 10 or 20 mu g; day 7: 30, 50, or 100 mu g; and day 10: 50, 100, or 200 mu g. Dose escalation was guided by dose-limiting toxicities. Results: The MTD was 10, 20, 50, and 200 mu g on days 0, 3, 7, and 10, respectively. Catumaxomab had an acceptable safety profile: Most common treatment-related adverse events (at the MTD) were fever, vomiting, and abdominal pain. At final examination, 11/17 evaluable patients (65%) were progression free: 1 patient had a complete and 3 a partial response. Median overall survival from the time of diagnosis of PC was 502 days. Conclusions: Intraperitoneal catumaxomab is a promising option for the treatment of PC from GI cancer

    Normal values of distal radioulnar translation assessed by three-dimensional C-arm scans: a cadaveric study

    We investigated whether mobile C-arm cone beam computer tomography (CBCT) could be used to measure radioulnar translation. The study was conducted on 31 Thiel-fixed intact cadaver arms. Three-dimensional scans of each wrist were carried out in pronation and supination. Four established measurement methods were used (radioulnar line, subluxation ratio, epicentre and radioulnar ratio methods) to measure radioulnar translation. The intraclass correlation coefficient for inter-observer and intra-observer reliability were excellent in three of four methods (>0.94). The reference ranges for physiological radioulnar translation were between -30% and 91% (radioulnar line method), -32% and 87% (subluxation ratio method), -40% and 23% (epicentre method), and 2% and 73% (radioulnar ratio method). Our results indicate that radioulnar translation in the distal radioulnar joint can be determined reliably using mobile C-arm CBCT. The normal values provide a basis for further experimental and clinical studies

    Differential cohesin loading marks paired and unpaired regions of platypus sex chromosomes at prophase i

    Cohesins are vital for chromosome organisation during meiosis and mitosis. In addition to the important function in sister chromatid cohesion, these complexes play key roles in meiotic recombination, DSB repair, homologous chromosome pairing and segregation. Egg-laying mammals (monotremes) feature an unusually complex sex chromosome system, which raises fundamental questions about organisation and segregation during meiosis. We discovered a dynamic and differential accumulation of cohesins on sex chromosomes during platypus prophase I and specific reorganisation of the sex chromosome complex around a large nucleolar body. Detailed analysis revealed a differential loading of SMC3 on the chromatin and chromosomal axis of XY shared regions compared with the chromatin and chromosomal axes of asynapsed X and Y regions during prophase I. At late prophase I, SMC3 accumulation is lost from both the chromatin and chromosome axes of the asynaptic regions of the chain and resolves into subnuclear compartments. This is the first report detailing unpaired DNA specific SMC3 accumulation during meiosis in any species and allows speculation on roles for cohesin in monotreme sex chromosome organisation and segregation.Aaron E.     Evaluation of ma-N 2400 Series DUV Photoresist for Electron Beam Exposure

    Several DUV positive and negative tone resists were developed for the photolithography at wavelengths of 248 nm and shorter. Most of these DUV photoresists are sensitive to electron beam exposure, too. In this paper the results of electron beam exposure of the negative tone ma-N 2400 series DUV photoresist will be presented. This resist has no chemical amplification and no critical process steps. Further, the resist provides a good resistivity to plasma etching processes, so that it can be used successfully for etch and lift-off processes. The necessary exposure doses for the resist layers spincoated with thicknesses between 0.3 and 0.8 µm on silicon wafers were in the range between 60 and 120 µC/cm². The exposed resist was developed with the metal ion free developer MIF 726. Patterns down to the low submicrometer region were generated.     EchidnaCSI – improving monitoring of a cryptic species at continental scale using Citizen Science

    Short-beaked echidna (Tachyglossus aculeatus) are a cryptic and iconic monotreme found throughout the continent of Australia. Despite observational records spanning many years aggregated in national and state biodiversity databases, the spatial and temporal intensity of sightings is limited. Although the species is of least conservation concern at the global level, a subspecies has been declared endangered on Kangaroo Island in South Australia. We need better population data over the whole continent to inform this species’ conservation management. To increase the temporal and spatial resolution of observations which may be used for more accurate population assessments, we developed a mobile app for citizen scientists to easily record echidna sightings and improve the quantity, quality and distribution of data collected for monitoring this species. EchidnaCSI is a free, cross-platform (Android & iOS), open-source app that we developed to collect echidna observational data around Australia. EchidnaCSI has been in use since September 2017 and uses mobile phone sensors to transparently and automatically record metadata, such as species observation location and time and GPS location precision. We examine differences in spatial coverage between these observations and those in existing data repositories in the Atlas of Living Australia and state biodiversity databases, especially in relation to observations in protected areas and to an index of remoteness and accessibility. EchidnaCSI has contributed over 8000 echidna observations from around Australia, more than recorded in all state systems combined, with similar spatial distribution. Although coverage was more limited in some protected areas than the reference data sources, numbers of observations in all remote areas were greater than the reference scientific data except for very remote regions. EchidnaCSI has improved the spatial and temporal intensity of observations for this iconic species and provides a complement to scientific surveys, which might usefully focus on highly protected areas and very remote regions.Alan Stenhouse, Tahlia Perry, Frank Grützner, Megan Lewis, Lian Pin Ko

    IODP workshop: developing scientific drilling proposals for the Argentina Passive Volcanic Continental Margin (APVCM) – basin evolution, deep biosphere, hydrates, sediment dynamics and ocean evolution

    The Argentine margin contains important sedimentological, paleontological and chemical records of regional and local tectonic evolution, sea level, climate evolution and ocean circulation since the opening of the South Atlantic in the Late Jurassic–Early Cretaceous as well as the present-day results of post-depositional chemical and biological alteration. Despite its important location, which underlies the exchange of southern- and northern-sourced water masses, the Argentine margin has not been investigated in detail using scientific drilling techniques, perhaps because the margin has the reputation of being erosional. However, a number of papers published since 2009 have reported new high-resolution and/or multichannel seismic surveys, often combined with multi-beam bathymetric data, which show the common occurrence of layered sediments and prominent sediment drifts on the Argentine and adjacent Uruguayan margins. There has also been significant progress in studying the climatic records in surficial and near-surface sediments recovered in sediment cores from the Argentine margin. Encouraged by these recent results, our 3.5-day IODP (International Ocean Discovery Program) workshop in Buenos Aires (8–11 September 2015) focused on opportunities for scientific drilling on the Atlantic margin of Argentina, which lies beneath a key portion of the global ocean conveyor belt of thermohaline circulation. Significant opportunities exist to study the tectonic evolution, paleoceanography and stratigraphy, sedimentology, and biosphere and geochemistry of this margin.Facultad de Ciencias Naturales y Muse
