    Assessment of a discontinuous galerkin method for the simulation of the turbulent flow around the drivaer car model

    none7noThe turbulent flow over the DrivAer fastback model is here investigated with an orderadaptive discontinuous Galerkin (DG) method. The growing need of high-fidelity flow simulations for the accurate determination of problems, e.g., vehicle aerodynamics, promoted research on models and methods to improve the computational efficiency and to bring the practice of Scale Resolving Simulations (SRS), like the large-eddy simulation (LES), to an industrial level. An appealing choice for SRS is the Implicit LES (ILES) via a high-order DG method, where the favourable numerical dissipation of the space discretization scheme plays directly the role of a subgrid-scale model. Implicit time integration and the p-adaptive algorithm reduce the computational cost allowing a high-fidelity description of the physical phenomenon with very coarse mesh and moderate number of degrees of freedom. Two different models have been considered: (i) a simplified DrivAer fastback model, without the rear-view mirrors and the wheels, and a smooth underbody; (ii) the DrivAer fastback model, without rear-view mirrors and a smooth underbody.     Ship graffiti at the Zanzibar Gereza (Old Fort), Stone Town, Unguja, Tanzania

Available from Taylor & Francis via the DOI in this record. A number of graffiti of ships are to be found engraved into the plaster of the Gereza (Old Fort) of Stone Town on Unguja, the main island of the Zanzibar Archipelago, Tanzania. Most of those reported here appear on the ramparts of the southwestern tower, while some are on the western face of the main partition wall separating the western and eastern wards. Although sometimes sketchy, the images suggest a number of vessel types, including a frigate or frigate-built vessel and a number of settee-rigged ocean-going vessels referred to exonymically as ‘dhows’. Some appear to have transom sterns, hinting at particular vessel types, such as the baghla, ghanja, sanbūq or kotia. Two graffiti might also depict the stem heads of the East African mtepe. The graffiti are documented and interpreted in the context of the fort, Oman’s East African empire, and the Indian Ocean dhow trade. The construction history of the building and the vessel types depicted date the graffiti to the mid-late nineteenth century.British Academ

    An Innovative Lipidomic Workflow to Investigate the Lipid Profile in a Cystic Fibrosis Cell Line

    Altered lipid metabolism has been associated to cystic fibrosis disease, which is characterized by chronic lung inflammation and various organs dysfunction. Here, we present the validation of an untargeted lipidomics approach based on high-resolution mass spectrometry aimed at identifying those lipid species that unequivocally sign CF pathophysiology. Of n.13375 mass spectra recorded on cystic fibrosis bronchial epithelial airways epithelial cells IB3, n.7787 presented the MS/MS data, and, after software and manual validation, the final number of annotated lipids was restricted to n.1159. On these lipids, univariate and multivariate statistical approaches were employed in order to select relevant lipids for cellular phenotype discrimination between cystic fibrosis and HBE healthy cells. In cystic fibrosis IB3 cells, a pervasive alteration in the lipid metabolism revealed changes in the classes of ether-linked phospholipids, cholesterol esters, and glycosylated sphingolipids. Through functions association, it was evidenced that lipids variation involves the moiety implicated in membrane composition, endoplasmic reticulum, mitochondria compartments, and chemical and biophysical lipids properties. This study provides a new perspective in understanding the pathogenesis of cystic fibrosis and strengthens the need to use a validated mass spectrometry-based lipidomics approach for the discovery of potential biomarkers and perturbed metabolism

    Real-time marker-less multi-person 3D pose estimation in RGB-Depth camera networks

    This paper proposes a novel system to estimate and track the 3D poses of multiple persons in calibrated RGB-Depth camera networks. The multi-view 3D pose of each person is computed by a central node which receives the single-view outcomes from each camera of the network. Each single-view outcome is computed by using a CNN for 2D pose estimation and extending the resulting skeletons to 3D by means of the sensor depth. The proposed system is marker-less, multi-person, independent of background and does not make any assumption on people appearance and initial pose. The system provides real-time outcomes, thus being perfectly suited for applications requiring user interaction. Experimental results show the effectiveness of this work with respect to a baseline multi-view approach in different scenarios.     Inhibition of Sphingolipid Synthesis as a Phenotype-Modifying Therapy in Cystic Fibrosis

    Cystic Fibrosis (CF) is an inherited disease associated with a variety of mutations affecting the CFTR gene. A deletion of phenylalanine 508 (F508) affects more than 70% of patients and results in unfolded proteins accumulation, originating a proteinopathy responsible for inflammation, impaired trafficking, altered metabolism, cholesterol and lipids accumulation, impaired autophagy at the cellular level. Lung inflammation has been extensively related to the accumulation of the lipotoxin ceramide. We recently proved that inhibition of ceramide synthesis by Myriocin reduces inflammation and ameliorates the defence response against pathogens infection, which is downregulated in CF.     Prodromal frontotemporal dementia: clinical features and predictors of progression

    Background: The prodromal phase of frontotemporal dementia (FTD) is still not well characterized, and conversion rates to dementia and predictors of progression at 1-year follow-up are currently unknown. // Methods: In this retrospective study, disease severity was assessed using the global CDR plus NACC FTLD. Prodromal FTD was defined to reflect mild cognitive or behavioural impairment with relatively preserved functional independence (global CDR plus NACC = 0.5) as well as mild, moderate and severe dementia (classified as global CDR plus NACC = 1, 2, 3, respectively). Disease progression at 1-year follow-up and serum NfL measurements were acquired in a subgroup of patients. // Results: Of 563 participants, 138 were classified as prodromal FTD, 130 as mild, 175 as moderate and 120 as severe FTD. In the prodromal and mild phases, we observed an early increase in serum NfL levels followed by behavioural disturbances and deficits in executive functions. Negative symptoms, such as apathy, inflexibility and loss of insight, predominated in the prodromal phase. Serum NfL levels were significantly increased in the prodromal phase compared with healthy controls (average difference 14.5, 95% CI 2.9 to 26.1 pg/mL), but lower than in patients with mild FTD (average difference -15.5, 95% CI -28.4 to -2.7 pg/mL). At 1-year follow-up, 51.2% of patients in the prodromal phase had converted to dementia. Serum NfL measurements at baseline were the strongest predictors of disease progression at 1-year follow-up (OR 1.07, 95% CI 1.03 to 1.11, p < 0.001). // Conclusions: Prodromal FTD is a mutable stage with high rate of progression to fully symptomatic disease at 1-year follow-up. High serum NfL levels may support prodromal FTD diagnosis and represent a helpful marker to assess disease progression
