The human silent information regulator (Sir)2 homologue hSIRT3 is a mitochondrial nicotinamide adenine dinucleotide-dependent deacetylase.
The yeast silent information regulator (Sir)2 protein links cellular metabolism and transcriptional silencing through its nicotinamide adenine dinucleotide (NAD)-dependent histone deacetylase activity. We report that mitochondria from mammalian cells contain intrinsic NAD-dependent deacetylase activity. This activity is inhibited by the NAD hydrolysis product nicotinamide, but not by trichostatin A, consistent with a class III deacetylase. We identify this deacetylase as the nuclear-encoded human Sir2 homologue hSIRT3, and show that hSIRT3 is located within the mitochondrial matrix. Mitochondrial import of hSIRT3 is dependent on an NH2-terminal amphipathic alpha-helix rich in basic residues. hSIRT3 is proteolytically processed in the mitochondrial matrix to a 28-kD product. This processing can be reconstituted in vitro with recombinant mitochondrial matrix processing peptidase (MPP) and is inhibited by mutation of arginines 99 and 100. The unprocessed form of hSIRT3 is enzymatically inactive and becomes fully activated in vitro after cleavage by MPP. These observations demonstrate the existence of a latent class III deacetylase that becomes catalytically activated upon import into the human mitochondria
SIRT1 and SIRT3 deacetylate homologous substrates: AceCS1,2 and HMGCS1,2.
SIRT1 and SIRT3 are NAD+-dependent protein deacetylases that are evolutionarily conserved across mammals. These proteins are located in the cytoplasm/nucleus and mitochondria, respectively. Previous reports demonstrated that human SIRT1 deacetylates Acetyl-CoA Synthase 1 (AceCS1) in the cytoplasm, whereas SIRT3 deacetylates the homologous Acetyl-CoA Synthase 2 (AceCS2) in the mitochondria. We recently showed that 3-hydroxy-3-methylglutaryl CoA synthase 2 (HMGCS2) is deacetylated by SIRT3 in mitochondria, and we demonstrate here that SIRT1 deacetylates the homologous 3-hydroxy-3-methylglutaryl CoA synthase 1 (HMGCS1) in the cytoplasm. This novel pattern of substrate homology between cytoplasmic SIRT1 and mitochondrial SIRT3 suggests that considering evolutionary relationships between the sirtuins and their substrates may help to identify and understand the functions and interactions of this gene family. Fast 2.5D Finite Element Simulations of Borehole Resistivity Measurements
We develop a rapid 2.5-dimensional (2.5D) finite element method for simulation of borehole resistivity measurements in transversely isotropic (TI) media. The method combines arbitrary high-order - and (curl)-conforming spatial discretizations. It solves problems where material properties remain constant along one spatial direction, over which we consider a Fourier series expansion and each Fourier mode is solved independently. We propose a novel a priori method to construct quasi-optimal discretizations in physical and Fourier space. This construction is based on examining the analytical (fundamental) solution of the 2.5D formulation over multiple homogeneous spaces and assuming that some of its properties still hold for the 2.5D problem over a spatially heterogeneous formation. In addition, a simple parallelization scheme over multiple measurement positions provides efficient scalability. Our method yields accurate borehole logging simulations for realistic synthetic examples, delivering simulations of borehole resistivity measurements at a rate faster than 0.05 s per measurement location along the well trajectory on a 96-core computer
Long-term calorie restriction in humans is not associated with indices of delayed immunologic aging: A descriptive study.
BACKGROUND: Delayed immunologic aging is purported to be a major mechanism through which calorie restriction (CR) exerts its anti-aging effects in non-human species. However, in non-obese humans, the effect of CR on the immune system has been understudied relative to its effects on the cardiometabolic system. OBJECTIVE: To examine whether CR is associated with delayed immunologic aging in non-obese humans. METHODS: We tested whether long-term CR practitioners (average 10.03 years of CR) evidenced decreased expression of T cell immunosenescence markers and longer immune cell telomeres compared to gender-, race/ethnicity-, age-, and education-matched "healthy" Body Mass Index (BMI) and "overweight"/"obese" BMI groups. RESULTS: Long-term human CR practitioners had lower BMI (p < 0.001) and fasting glucose (p < 0.001), as expected. They showed similar frequencies of pre-senescent cells (CD8+CD28- T cells and CD57 and PD-1 expressing T cells) to the comparison groups. Even after adjusting for covariates, including cytomegalovirus status, we observed shorter peripheral blood mononuclear cell telomeres in the CR group (p = 0.012) and no difference in granulocyte telomeres between groups (p = 0.42). CONCLUSIONS: We observed no clear evidence that CR as it is currently practiced in humans delays immune aging related to telomere length or T cell immunosenescent markers
EDUCAÇÃO, Formação Humana e Tecnologia: Diálogos Com o Homem Unidimensional de Marcuse
Esta dissertação se insere no âmbito da pesquisa teórica, no campo dos fundamentos filosóficos da educação crítica. O problema da pesquisa diz respeito aos desafios que os conceitos de tecnologia e cultura, presentes nas reflexões de Herbert Marcuse, oferecem para a teoria crítica da educação na atualidade. A reflexão gira em torno dos processos formativos a partir de sua relação com a tecnologia, especificamente com a Internet. Parte da fundamentação filosófica de Herbert Marcuse, sobre os conceitos de cultura e tecnologia presentes no livro One-Dimensional Man e nos ensaios Sobre o caráter afirmativo da cultura e Comentários para uma redefinição do conceito de cultura. A hipótese é que os conceitos desenvolvidos pelo autor em diálogo com fenômenos socialmente constituídos bem como com os emergentes caminham a contrapelo da lógica da racionalidade técnico-instrumental hegemônica. Toma como ponto de partida a revisão teórica cujo resultado revela a atualidade e interesse pela temática no campo educacional e também constata a incipiência de trabalhos a partir da filosofia crítica de Marcuse, tanto em fontes nacionais quanto em internacionais. Pela análise crítica dos trabalhos acessados infere-se que mediada pela Internet a tecnologia também pode causar a utilização de formas empobrecidas e reduzidas dos modos de perceber e significar acontecimentos, relações, projeções e possibilidades, além de poder ser alterado o caráter da subjetivação por meio de processos de apropriação de indícios danificados de memória e cultura. Combinados às técnicas de propaganda e autopromoção, esses elementos corroboram a configuração e aplicação do projeto cognitivo para o sujeito unidimensional, confortavelmente rendido à lógica da sociedade que o produz. A potencialidade do uso de ferramentas tecnológicas livres do projeto da racionalidade técnico-instrumental que se sobrepõe à faceta sensível e amorosa da vida humana, típico do capitalismo tardio das sociedades industriais avançadas, pode reverter os movimentos de organização tecnológica intelectual e política, de conquista e manipulação do pensamento negativo, da consciência infeliz, de liberdades, necessidades, desejos, modos de administrar o tempo livre, de projeção de padrões de vida que em conjunto cerceiam movimentos para a transformação social

Metabolic reprogramming of human CD8+ memory T cells through loss of SIRT1.
The expansion of CD8+CD28- T cells, a population of terminally differentiated memory T cells, is one of the most consistent immunological changes in humans during aging. CD8+CD28- T cells are highly cytotoxic, and their frequency is linked to many age-related diseases. As they do not accumulate in mice, many of the molecular mechanisms regulating their fate and function remain unclear. In this paper, we find that human CD8+CD28- T cells, under resting conditions, have an enhanced capacity to use glycolysis, a function linked to decreased expression of the NAD+-dependent protein deacetylase SIRT1. Global gene expression profiling identified the transcription factor FoxO1 as a SIRT1 target involved in transcriptional reprogramming of CD8+CD28- T cells. FoxO1 is proteasomally degraded in SIRT1-deficient CD8+CD28- T cells, and inhibiting its activity in resting CD8+CD28+ T cells enhanced glycolytic capacity and granzyme B production as in CD8+CD28- T cells. These data identify the evolutionarily conserved SIRT1-FoxO1 axis as a regulator of resting CD8+ memory T cell metabolism and activity in humans
Diversity-Productivity Relationship in the Northeastern Tamaulipan Thornscrub Forest of Mexico
This research examines the diversity-productivity relationship in a semiarid scrubland, initially under late successional conditions and subsequently under early successional conditions created by experimental clearing, to explore the roles that productivity and stochastic mortality play in species exclusion in this environment. A total of fifteen plots were studied by measuring environmental conditions and biomass components of shrubs and seedlings. These stands were distributed along a productivity gradient across five different landforms. A hypothesis about the stochastic self-thinning mortality model along the gradient was evaluated with the diversity-productivity-environment data. The diversity-productivity relationship was linear and reversed between the early and late succession stages. The hypothesis of stochastic mortality of species exclusion was rejected in the early stages of succession and partially accepted in the mature stage of succession. Species exclusion was negatively related to productivity gradients, suggesting that strong interspecific competition occurs in high productivity plots and that a larger number of species can survive in higher abiotic stress landscapes. Further research is needed to understand the temporal and spatial variations of the ecological interactions that shape this plant community
Conilon Vitória 'Incaper 8142': variedade clonal de café desenvolvida para o Estado do Espírito Santo.
O objetivo deste trabalho é apresentar as principais características da mais nova variedade melhorada de café conilon desenvolvida pelo Instituto Capixaba de Pesquisa, Assistência Técnica e Extensão Rural (Incaper), a ?Conilon Vitória ? Incaper 8142?. Foi obtida através da seleção, avaliação e caracterização de clones ao longo de dezoito anos de pesquisa com a espécie no estado. É formada pelo agrupamento de treze clones considerados superiores. Possui uma série de características agronômicas de interesse, destacando-se de forma especial o seu desempenho produtivo médio, da ordem de 70,4 Sc benef./ha, considerando-se um número mínimo de oito safras em condições não irrigadas. A sua correta utilização poderá possibilitar avanços ainda mais expressivos na cafeicultura do conilon do Espírito Santo
Caracterização morfo agronômica de café conilon
O Espírito Santo destaca-se como o maior produtor brasileiro da espécie Coffea canephora, conhecida no Estado como café Conilon. No programa de melhoramento genético de café do Incaper, tem-se diferentes materiais genéticos e populações de melhoramento sendo avaliados para diferentes características agronômicas. Este trabalho foi desenvolvido com o objetivo de estudar a caracterização fenotípica do café conilon, com base na avaliação de uma população heterogênea composta de 150 indivíduos. Os dados são oriundos da avaliação de 24 característica da população por dois anos na Fazenda Experimental de Marilândia, tomados com base na lista oficial dos descritores mínimos estabelecidos para Coffea sp. A população em estudo foi caracterizada para as seguintes características: a) Fruto (tamanho, formato, cor na fase madura, presença ou ausência de sépala, grau de aderência, suculência do mesocarpo, ciclo e uniformidade de maturação; b) semente: cor do endosperma, tonalidade da película de cobertura, grau de aderência da película, comprimento, largura e espessura; d) inflorescência: quantidade de rosetas por ramos e de flores por roseta e; e) planta: porte, vigor, adaptação (escala geral), reação a ferrugem, mancha de cercóspora e bicho mineiro. Verificaram-se diferenças importantes entre as progênies e os progenitores de um ano para o outro, evidenciando a necessidade de avaliação de populações de melhoramento em mais de uma safra e em regiões representativas da cultura para a precisa caracterização. Novos estudos são necessários, para estabelecer de maneira precisa todos os padrões para caracterização de genótipos de café conilon, com base na lista oficial dos descritores mínimos estabelecidos para café
