    The reported use and effectiveness of Hypericum (St John’s wort) on affective symptoms in a depression self-help group

    A recent meta-analysis suggested that Hypericum perforatum (St John’s wort) is an effective treatment for mild to moderate depression and may have a superior side-effect profile to some antidepressant drugs. The aim of this study was to assess the use of herbal remedies in treating depressive and anxiety symptoms, as reported by members of the UK self-help organization Depression Alliance using self-completed questionnaires. More than 50% of the 452 respondents reported using Hypericum, onequarter of whom also reported concurrent use of traditional antidepressants. Most of the sample reported sufficient symptoms for warranting a diagnosis of major depression, with the majority also describing symptoms suggestive of co-morbid psychiatric conditions. One-half of the Hypericum users experienced symptom improvement, which for most occurred within the first 4 weeks of use. Response was better for those with mild as compared to severe symptoms and poorer for those taking Hypericum alongside other antidepressants. The responders were generally older than non-responders. Adverse effects were reported by one-quarter of users and were mostly psychological in nature. This retrospective survey indicated that use of herbal remedies was common in this population. Although often helpful in relieving symptoms, particularly in those with mild depression, there is a risk of adverse events and drug interaction

    Amelioration of root disease of subterranean clover (Trifolium subterraneum) by mineral nutrients

    Subterranean clover (Trifolium subterraneum) is a key pasture legume across southern Australia and elsewhere. Decline in subterranean clover pastures was first recognised in Australia during the 1960s and manifests as an increase in weeds and a decrease in desirable legume species. While both root disease and poor nutrition contribute to subterranean clover pasture decline, the relationships between root disease and nutrition have not been determined. The objective of this study was to define these relationships. Field experiments were undertaken to determine the nutritional and pathogen status of soils and subterranean clover from three Western Australian field sites. Subsequently, controlled environment experiments were undertaken to determine the relative severities of tap and lateral root disease and growth of plants when soil cores taken from these three field sites were amended with a complete nutrient solution or a range of individual macro- or micronutrient treatments. Application of a ‘Hoaglands’ complete nutrient solution decreased the severity of tap root disease by an average of 45% and lateral root disease by 32%. Amendment with K alone reduced the severity of tap root disease an average of 32%; while the application of N alone reduced the severity of tap root disease by 33% and lateral root disease by 27%. Application of Hoaglands, K, N or Zn increased shoot and root dry weight, while Mo only increased shoot dry weight. This is the first report to show that mineral nutrients can substantially ameliorate root disease in subterranean clover. The results demonstrate that while root disease limits plant growth, improvement in the nutritional status of nutrient-impoverished soils can significantly reduce root disease. There is significant potential to incorporate nutrient amendments into an integrated and more sustainable approach to better manage root disease and to increase productivity of pasture legumes where soils are inherently nutrient deficient in one or more nutrients

    Short-chain fatty acid level and field cancerization show opposing associations with enteroendocrine cell number and neuropilin expression in patients with colorectal adenoma

    BACKGROUND: Previous reports have suggested that the VEGF receptor neuropilin-1 (NRP-1) is expressed in a singly dispersed subpopulation of cells in the normal colonic epithelium, but that expression becomes dysregulated during colorectal carcinogenesis, with higher levels in tumour suggestive of a poor prognosis. We noted that the spatial distribution and morphology if NRP-1 expressing cells resembles that of enteroendocrine cells (EEC) which are altered in response to disease state including cancer and irritable bowel syndrome (IBS). We have shown that NRP-1 is down-regulated by butyrate in colon cancer cell lines in vitro and we hypothesized that butyrate produced in the lumen would have an analogous effect on the colon mucosa in vivo. Therefore we sought to investigate whether NRP-1 is expressed in EEC and how NRP-1 and EEC respond to butyrate and other short-chain fatty acids (SCFA - principally acetate and propionate). Additionally we sought to assess whether there is a field effect around adenomas. METHODOLOGY: Biopsies were collected at the mid-sigmoid, at the adenoma and at the contralateral wall (field) of 28 subjects during endoscopy. Samples were fixed for IHC and stained for either NRP-1 or for chromogranin A (CgA), a marker of EEC. Stool sampling was undertaken to assess individuals' butyrate, acetate and propionate levels. RESULT: NRP-1 expression was inversely related to SCFA concentration at the colon landmark (mid-sigmoid), but expression was lower and not related to SCFA concentration at the field. Likewise CgA+ cell number was also inversely related to SCFA at the landmark, but was lower and unresponsive at the field. Crypt cellularity was unaltered by field effect. A colocalisation analysis showed only a small subset of NRP-1 localised with CgA. Adenomas showed extensive, weaker staining for NRP-1 which contrastingly correlated positively with butyrate level. Field effects cause this relationship to be lost. Adenoma tissue shows dissociation of the co-regulation of NRP-1 and EEC. CONCLUSION: NRP-1 is inversely associated with levels of butyrate and other SCFA in vivo and is expressed in a subset of CgA expressing cells. EEC number is related to butyrate level in the same way

    Blood immune transcriptome analysis of artificially fed dairy calves and naturally suckled beef calves from birth to 7 days of age

    peer-reviewedNeonatal calves possess a very immature and naïve immune system and are reliant on the intake of maternal colostrum for passive transfer of immunoglobulins. Variation in colostrum management of beef and dairy calves is thought to affect early immune development. Therefore, the objective of this study was to examine changes in gene expression and investigate molecular pathways involved in the immune-competence development of neonatal Holstein dairy calves and naturally suckled beef calves using next generation RNA-sequencing during the first week of life. Jugular whole blood samples were collected from Holstein (H) dairy calves (n = 8) artificially fed 5% B.W. colostrum, and from beef calves which were the progenies of Charolais-Limousin (CL; n = 7) and Limousin-Friesian beef suckler cows (LF; n = 7), for subsequent RNA isolation. In dairy calves, there was a surge in pro-inflammatory cytokine gene expression possibly due to the stress of separation from the dam. LF calves exhibited early signs of humoral immune development with observed increases in the expression genes coding for Ig receptors, which was not evident in the other breeds by 7 days of age. Immune and health related DEGs identified as upregulated in beef calves are prospective contender genes for the classification of biomarkers for immune-competence development, and will contribute towards a greater understanding of the development of an immune response in neonatal calves

    A time-dependent model for improved biogalvanic tissue characterisation

    Measurement of the passive electrical resistance of biological tissues through biogalvanic characterisation has been proposed as a simple means of distinguishing healthy from diseased tissue. This method has the potential to provide valuable real-time information when integrated into surgical tools. Characterised tissue resistance values have been shown to be particularly sensitive to external load switching direction and rate, bringing into question the stability and efficacy of the technique. These errors are due to transient variations observed in measurement data that are not accounted for in current electrical models. The presented research proposes the addition of a time-dependent element to the characterisation model to account for losses associated with this transient behaviour. Influence of switching rate has been examined, with the inclusion of transient elements improving the repeatability of the characterised tissue resistance. Application of this model to repeat biogalvanic measurements on a single ex vivo human colon tissue sample with healthy and cancerous (adenocarcinoma) regions showed a statistically significant difference (p 0.05) between tissue types was found when measurements were subjected to the current model, suggesting that the proposed model may allow for improved biogalvanic tissue characterisation

    Supercritical water oxidation of dioxins and furans in waste incinerator fly ash, sewage sludge and industrial soil

    Three environmental samples containing dioxins and furans have been oxidized in the presence of hydrogen peroxide under supercritical water oxidation conditions. The samples consisted of a waste incinerator fly ash, sewage sludge and contaminated industrial soil. The reactor system was a batch, autoclave reactor operated at temperatures between 350°C and 450°C, corresponding to pressures of ~20-33.5 MPa and with hydrogen peroxide concentrations from 0.0 to 11.25 vol%. Hydrogen peroxide concentration and temperature/pressure had a strong positive effect on the oxidation of dioxins and furans. At the highest temperatures and pressure of supercritical water oxidation of 450°C and 33.5 MPa and with 11.25 vol% of hydrogen peroxide, the destruction efficiencies of the individual polychlorinated dibenzo-ρ-dioxins/polychlorinated dibenzofurans (PCDD/PCDF) isomers were between 90% and 99%. There did not appear to be any significant differences in the PCDD/PCDF destruction efficiencies in relation to the different sample matrices of the waste incinerator fly ash, sewage sludge and contaminated industrial soil

    Abstract In science education, the question about using technology to teach the content and concepts is enduring. The complexity of teaching content, concepts and practices is expressed at the same time that technology has "advanced" to multiple devices beyond the desktop or laptop forms of computers to devices that are hand-held and range from e-readers to iPads and beyond. Each device has applications (Apps) that range from transmission of content to learning tools. Because no formal evaluation of Ipad Apps exists, a method was developed and implemented for our research. Researchers in this study developed a method for reviewing science and mathematics Apps and provided a list of the "top" Apps in an alphabetical topic list found in a newly developed website
