    Тенденции развития комплексных транспортных схем городов

    The article discusses the process of planning the work of urban transport operations in the form of an integrated transport scheme (ITS), which is the transport part of the master plan for development of a city, and studies organization of urban transport. The objective of the article is to formulate the main directions of developments in the field of transport planning based on modern domestic and foreign experience and knowledge, and on retrospective analysis of changes in the field of urban transport planning.The methodology is presented for the conditions prevailing in the Russian Federation, the peculiarity of which comprise a relatively late (compared to European countries and the USA), but rapidly developing motorization of the country, a change in qualifications of specialists working in the field of urban transport planning, a certain loss of experience and skills in development of ITS in recent years.The suggested conclusions represent a brief description of the concept of a common toolkit, a scheme and the most promising methods for developing approaches to development of integrated transport schemes and require further in-depth research in each specific segment.В статье рассматривается процесс планирования работы городского транспорта в форме комплексной транспортной схемы (КТС), являющейся транспортной частью генерального плана развития города и рассматривающей вопросы организации городского транспорта. Цель статьи – сформулировать основные направления разработок в сфере транспортного планирования на основе современного отечественного и зарубежного опыта и знаний, в том числе с использованием ретроспективного анализа изменений в сфере планирования городского транспорта.Анализ представлен для условий Российской Федерации, особенностями которых являются относительно поздняя (по сравнению со странами Европы и США), но быстро протекающая автомобилизация страны, изменение квалификационного состава специалистов, работающих в сфере планирования городского транспорта, определённая потеря опыта и умения разработки КТС за последние годы.Предложенные выводы представляют собой краткое описание концепта общего инструментария, схем и наиболее перспективных методов разработки подходов к развитию комплексных транспортных схем и требуют дальнейших углублённых исследований по каждому направлению

    Remote visual detection of impacts on the lunar surface

    We propose a novel method of remotely observing impacts on the airless Moon that may extend the present data base on meteoroids down to 1 m in diameter. Meteorites or comets of radius approximately 1-100 m are burnt away or dispersed in the atmospheres of the Earth and Venus. However, when such objects strike the Moon they deposit their energy in a small initial volume, forming a plasma plume whose visible and infrared radiation may be visible from the Earth.     Mapping of heterologous expressed sequence tags as an alternative to microarrays for study of defense responses in plants

    BACKGROUND: Microarray technology helped to accumulate an immense pool of data on gene expression changes in response to different environmental factors. Yet, computer- generated gene profiling using expressed sequence tags (EST) represents a valuable alternative to microarrays, which allows efficient discovery of homologous sequences in evolutionarily different species and comparison of gene sets on the whole genome scale. In this study, we used publicly available EST database derived from different plant species infected with a variety of pathogens, to generate an expression profile of homologous genes involved in defense response of a model organism, Arabidopsis thaliana. RESULTS: EST-driven prediction identified 4,935 genes (16% of the total Arabidopsis genome) which, according to the origin of EST sets, were associated with defense responses in the reference genome. Profiles of defense-related genes, obtained by mapping of heterologous EST, represent putative Arabidopsis homologs of the corresponding species. Comparison of these profiles in pairs and locating common genes allowed estimating similarity between defense-related gene sets of different plant species. To experimentally support computer data, we arbitrarily selected a number of transcription factor genes (TF) detected by EST mapping. Their expression levels were examined by real-time polymerase chain reaction during infection with yellow strain of Cucumber mosaic virus, a compatible virus systemically infecting Arabidopsis. We observed that 65% of the designated TF were upregulated in accordance with the EST-generated profile. CONCLUSION: We demonstrated that heterologous EST mapping may be efficiently used to reveal genes involved in host defense responses to pathogens. Upregulated genes identified in this study substantially overlap with those previously obtained by microarrays

    The influence of different anticoagulants and sample preparation methods on measurement of mCD14 on bovine monocytes and polymorphonuclear neutrophil leukocytes

    <p>Abstract</p> <p>Background</p> <p>Membrane-CD14 (mCD14) is expressed on the surface of monocytes, macrophages and polymorphonuclear neutrophil leukocytes (PMN). mCD14 acts as a co-receptor along with Toll like receptor 4 (TLR 4) and MD-2 for the detection of lipopolysaccharide (LPS). However, studies using different sample preparation methods and anticoagulants have reported different levels of mCD14 on the surface of monocytes and neutrophils. In this study, the influence of various anticoagulants and processing methods on measurement of mCD14 on monocytes and neutrophils was examined.</p> <p>Results</p> <p>Whole blood samples were collected in vacutainer tubes containing either sodium heparin (HEPARIN), ethylenediaminetetraacetic acid (EDTA) or sodium citrate (CITRATE). mCD14 on neutrophils and monocytes in whole blood samples or isolated cells was measured by the method of flow cytometry using fluorescein isothiocyanate (FITC)-labeled monoclonal antibody. There was a significant difference (<it>p </it>< 0.05) in the mean channel fluorescence intensity (MFI) of mCD14 on neutrophils in whole blood samples anticoagulated with HEPARIN (MFI = 64.77) in comparison with those in whole blood samples anticoagulated with either EDTA (MFI = 38.25) or CITRATE (MFI = 43.7). The MFI of mCD14 on monocytes in whole blood samples anticoagulted with HEPARIN (MFI = 206.90) was significantly higher than the MFI in whole blood samples anticoagulated with EDTA (MFI = 149.37) but similar to that with CITRATE (MFI = 162.55). There was no significant difference in the percentage of whole blood neutrophils or monocytes expressing mCD14 irrespective of type of anticoagulant used. However, MFI of mCD14 on monocytes was about 3.2-folds (HEPARIN), 3.9-folds (EDTA) or 3.7 folds (CITRATE) higher than those on neutrophils. Furthermore, there was no significant difference in mCD14 levels between unprocessed whole blood monocytes and monocytes in peripheral blood mononuclear cell preparation. Conversely, a highly significant difference was observed in mCD14 between unprocessed whole blood neutrophils and isolated neutrophils (<it>p </it>< 0.05).</p> <p>Conclusion</p> <p>From these results, it is suggested that sodium heparin should be the preferred anticoagulant for use in the reliable quantification of the surface expression of mCD14. Furthermore, measurement of mCD14 is best carried out in whole blood samples, both for neutrophils and monocytes.</p
