    CA 125 in seminal plasma: correlation with semen parameters

    Ovarian cancer marker CA 125 was measured in human seminal plasma, and the concentrations ranged between 22 and 1149 U/ml, and between 39 and 4711 U/ejaculate. This very high patient-to-patient variability was in contrast to a much lower within-patient variability, which was comparable to that of other semen parameters. No significant differences in CA 125 concentration were found in seminal plasma from normospermic patients, patients with male factors, vasectomized men, and in aliquots of samples which led to a pregnancy, via artificial insemination or in-vitro fertilization. The seminal plasma CA 125 concentration was not correlated with sperm count, motility and morphology. In contrast, seminal plasma CA 125 concentrations correlated with the age of the patient (P < 0.001) and inversely with the volume of the ejaculate (P < 0.001). These correlations were independent of each other. CA 125 did not correlate with the prostatic marker zinc, but did do so with the seminal vesicle marker fructose and the epididymal marker carnitin

    Effects of tumour necrosis factor-α, interleukin-1 α, macrophage colony stimulating factor and transforming growth factor β on trophoblastic matrix metalloproteinases

    The aim of this study was to determine the effects of tumour necrosis factor α (TNF), interleukin-1 α (IL-1α), macrophage colony-stimulating factor (MCSF) and transforming growth factor β (TGFβ) on the secretion of matrix metalloproteinases (MMP), human chorionic gonadotrophin (HCG) and fetal fibronectin (fFN) by purified first trimester cytotrophoblastic cells (CTB) in vitro. CTB were obtained from legal abortions and cultured in vitro in the presence or absence of the different cytokines. Secreted gelatinases were analysed in the culture supernatants by zymography, by measurements of the total gelatinolytic activity and by enzyme immunoassays. HCG and fFN were measured by commercially available immunoassays. TNF increased the total gelatinolytic activity by increasing MMP-9 activity (P = 0.025-0.0177) but decreased MMP-2 activity (P < 0.03) and immunoreactivity (P < 0.05), fFN (P < 0.02) and HCG (P < 0.01). IL-1α significantly increased the secretion of fFN (P < 0.02), the activity (P < 0.02) and immunoreactivity (P < 0.05) of MMP-9 but had no effect on the other parameters. MCSF increased MMP-9 immunoreactivity (P < 0.05) and moderately decreased HCG. TGFβ inhibited total gelatinolytic activity, MMP-9 activity and immunoreactivity, but was without effect on MMP-2 concentrations and activity.     It has been shown that and human endometria have the capacity to produce complement component 3 (C3). In rats, endometrial C3 is an oestrogen-dependent protein produced and secreted by glandular cells. The cell responsible for the synthesis and secretion of human endometrial C3 has not been clearly defined. Our study was aimed at answering this question. Samples of endometrium obtained from hysterectomies were either immunostained for C3 or digested with collagenase; then the stromal and glandular cells were separated and immunopurified (or not) with an antibody to CD45 coupled to magnetic beads to eliminate the endometrial lymphomyeloid cells. Cells were cultured for 2 weeks and C3 measured in the medium by an in-house radioimmunoassay. Glandular as well as stromal cells stained positively for C3 and released C3 in vitro. The release of C3 from both cell types could be inhibited by cycloheximide. Epithelial cells produced significantly more C3 than stromal cells, and endometrial C3 production was higher for both cell types when these were obtained from secretory as compared to proliferative endometria. Lymphomyeloid cells were possibly a source of C3 since after immunoadsorption of these cells, the remaining stromal or glandular cells produced significantly less C3.     Status of p53 in first-trimester cytotrophoblastic cells

    p53 has been called the cellular gatekeeper of the genome because it can induce cell-cycle arrest in G1, apoptosis or affect DNA replication in response to DNA damage. As p53 has been observed in first-trimester cytotrophoblastic cells (CTB), but its expression in normal cells is generally not detectable because of its short half-life, p53 could play an important role in cellular differentiation and/or in the control of the invasion of trophoblastic cells; therefore, p53 status was investigated in these cells. Using different antibodies recognizing different epitopes of p53 protein, abundant p53 expression was observed both in nuclear and in cytoplasmic compartments of first-trimester CTB. Whereas p53 was detected in the nuclei of few trophoblastic cells with an antibody recognizing the N-terminal epitope of the protein, high expression level of p53 in the cytoplasm of CTB was detected with an antibody recognizing the middle part of p53. The lack of immunoreactivity of p53 with antibodies recognizing the epitopes located at the N-terminus of p53 and the high level of p53 protein observed in the cytoplasm of CTB suggest that the N-terminus of p53 is involved in the formation of complexes. These cytoplasmic complexes were detected under non-reducing conditions in western blot analysis and had apparent molecular weights (MW) of 195, 167 or 125 kDa. These complexes could prolong the half-life of p53 in the cytoplasm of CTBs. By contrast, in the nuclei of CTBs, p53 seems to be present as a tetrame

    Voice Enabled Interface for Interactive Tour Guide Robots

    This paper considers design methodologies in order to develop voice-enabled interfaces for tour-guide robots to be deployed at the Robotics Exposition of the Swiss National Exhibition (Expo.02). Human-robot voice communication presents new challenges for design of fully autonomous mobile robots, in that interactivity must be robot-initiated in conversation and within a dynamic adverse environment. We approached these general problems for a voice enabled interface, tailored to limited computational resources of one on-board processor, when integrating smart speech signal acquisition, automatic speech recognition and synthesis, as well as dialogue system into the multi-modal, multi-sensor interface for the expo tour-guide robot. We also focus on particular issues that need to be addressed in voice-based interaction when planning specific tasks and research experiments for Expo.02 where tour-guide robots will interact with hundred of thousands of visitors during six months, seven days a week, ten hours per day

    Thermal design optimization of novel modular power converter assembly enabling higher performance, reliability and availability

    An alternative integration scheme for a half-bridge switch using 70 μm thin Si IGBTs and diodes is presented. This flat switch, which is designed for high-frequency application with high power density, exhibits high strength, high toughness, low parasitic inductance and high thermal conductivity. Such a novel assembly approach is suitable to optimize performance, reliability and availability of the power system in which it is used. The paper focuses on the thermal performance of this assembly at normal and extreme operating conditions, studied by means of FEM thermo-fluidynamic simulations of the module integrated with connectors and liquid cooler, and thermal measurement performed on an early prototype. Improved solutions are also investigated by the FE model

    The incommensurately modulated structures of natural natrite at 120 and 293 K from synchrotron X-ray data

    The incommensurately modulated structure of the mineral natrite has been refined for the first time. Two single-crystal grains, Lv and Kh, from two different occurrences [Mt. Karnasurt, Lovozero massif (Lv), Kola peninsula, Russia, and the pegmatite of Mt. Koashva, Khibiny massif (Kh)], have been investigated at 293 and 120 K using synchrotron X-ray data. The average structures of both minerals are identical and the basic features of the structural modulations are similar to the synthetic γ-Na2CO3 phase previously published. The γ (incommensurate) → δ (lock-in) phase transition reported at low temperature for the synthetic compound was not observed down to 120 K in natural natrite. Crystal-chemical aspects, especially about the second coordination sphere for the carbon atoms, are examined to explain the different structural behaviors observed at low temperature. The possible role played by the minor isomorphous substitutions in natural natrite specimens is also discussed

    In this paper we address the task of human-robot interaction in public mass exposition with several autonomous robots at a time. This implies questions regarding multi-robot control and interaction management with respect to social and commercial aspects of such an exposition. Multi-robot and interaction management is addressed with respect to visitor density and visitor flow. Human-robot interaction is modeled using the SOUL environment. Concluding we will present and discuss results from the Swiss national exhibition Expo.02 in the time from 15.05.02 to 20.10.02, with over 10000 hours of total robot operation time and more than 600000 visitors
