    Toxoplasma ceramide synthases: Gene duplication, functional divergence, and roles in parasite fitness.

    Toxoplasma gondii is an obligate, intracellular apicomplexan protozoan parasite of both humans and animals that can cause fetal damage and abortion and severe disease in the immunosuppressed. Sphingolipids have indispensable functions as signaling molecules and are essential and ubiquitous components of eukaryotic membranes that are both synthesized and scavenged by the Apicomplexa. Ceramide is the precursor for all sphingolipids, and here we report the identification, localization and analyses of the Toxoplasma ceramide synthases TgCerS1 and TgCerS2. Interestingly, we observed that while TgCerS1 was a fully functional orthologue of the yeast ceramide synthase (Lag1p) capable of catalyzing the conversion of sphinganine to ceramide, in contrast TgCerS2 was catalytically inactive. Furthermore, genomic deletion of TgCerS1 using CRISPR/Cas-9 led to viable but slow-growing parasites indicating its importance but not indispensability. In contrast, genomic knock out of TgCerS2 was only accessible utilizing the rapamycin-inducible Cre recombinase system. Surprisingly, the results demonstrated that this "pseudo" ceramide synthase, TgCerS2, has a considerably greater role in parasite fitness than its catalytically active orthologue (TgCerS1). Phylogenetic analyses indicated that, as in humans and plants, the ceramide synthase isoforms found in Toxoplasma and other Apicomplexa may have arisen through gene duplication. However, in the Apicomplexa the duplicated copy is hypothesized to have subsequently evolved into a non-functional "pseudo" ceramide synthase. This arrangement is unique to the Apicomplexa and further illustrates the unusual biology that characterize these protozoan parasites. [Abstract copyright: © 2023 The Authors.     Direct epitaxial growth of polar Hf0.5 Zr0.5 O2 films on corundum

    Single-phase epitaxial Hf0.5 Zr0.5 O2 films with non-centrosymmetric orthorhombic structure have been grown directly on electrode-free corundum (a-Al2 O3) substrates by pulsed laser deposition. A combination of high-resolution X-ray diffraction and X-ray absorption spectroscopy confirms the epitaxial growth of high-quality films belonging to the Pca21 space group, with 111] out-of-plane orientation. The surface of a 7-nm-thick sample exhibits an atomic step-terrace structure with a corrugation of the order of one atomic layer, as proved by atomic force microscopy. Scanning transmission electron microscopy reveals that it consists of grains with around 10 nm lateral size. The polar nature of this film has been corroborated by pyroelectric measurements. These results shed light on the mechanisms of the epitaxial stabilization of the ferroelectric phase of hafnia. © 2022 by the authors. Licensee MDPI, Basel, Switzerland

    Heart echinococcus cyst as an incidental finding: early detection might be life-saving

    We present a 46-year-old female smoker who was admitted to the emergency department of our hospital due to cough with blood-tinged sputum for the last four days before admission. Using echocardiography and Multi-Detector Computed Tomography (MDCT) heart Echinococcosis was diagnosed. Echinococcosis is a severe health issue in some geographical regions of the world. Hydatid infection of the heart is rare and the clinical presentation is usually insidious but there is always the lethal hazard of cyst perforation. Early diagnosis and an integrated treatment strategy are crucial. The results of surgical treatment of heart echinococcosis are better than the conservative strategy only. Extraction of the cyst combined with chemotherapy peri or post operative aiming to decrease the recurrences, consists the lege artis method of encountering this medical entity. Surgical excision was performed and the patient had an uneventful recovery and follow up at six and twelve months

    Rational dynamical systems

    We introduce the notion of a rational dynamical system extending the classical notion of a topological dynamical system and we prove (multiple) recurrence results for such systems via a partition theorem for the rational numbers proved by Farmaki and Koutsogiannis (2010) in [3]. In particular, we extend classical recurrence results developed by Furstenberg and Weiss. Also, we give some applications of these topological recurrence results to topology, to combinatorics, to Diophantine approximations and to number theory. © 2011 Elsevier B.V

    Topological dynamics indexed by words

    Starting with a combinatorial partition theorem for words over an infinite alphabet dominated by a fixed sequence, established recently by the authors, we prove recurrence results for topological dynamical systems indexed by such words. In this way we extend the classical theory developed by Furstenberg and Weiss of dynamical systems indexed by the natural numbers to systems indexed by words. Moreover, applying this theory to topological systems indexed by semigroups that can be represented as words we get analogous recurrence results for such systems. © 2011 Elsevier B.V

    Addressing missing environmental data via a machine learning scheme

    An important aspect in environmental sciences is the study of air quality, using statistical methods (environmental statistics) which utilize large datasets of climatic parameters. The air-quality-monitoring networks that operate in urban areas provide data on the most important pollutants, which, via environmental statistics, can be used for the development of continuous surfaces of pollutants’ concentrations. Generating ambient air-quality maps can help guide policy makers and researchers to formulate measures to minimize the adverse effects. The information needed for a mapping application can be obtained by employing spatial interpolation methods to the available data, for generating estimations of air-quality distributions. This study used point-monitoring data from the network of stations that operates in Athens, Greece. A machine-learning scheme was applied as a method to spatially estimate pollutants’ concentrations, and the results can be effectively used to implement missing values and provide representative data for statistical analyses purposes. © 2021 by the authors.
