    Low b-wave amplitudes in a strain of rabbits with a pigment epithelium defect

    AbstractWhen preparing isolated rabbit retinas we found in some animals fundi which were not uniformly dark but had abnormal areas of red coloration. The in situ electroretinograms (ERG) of 82 rabbits recorded after 1 h of dark adaptation were checked for abnormalities indicative of a degenerative disorder. The ERGs of eight rabbits with small dark adapted b-waves (≤250 μV) were re-recorded and their b-waves found to decline with time. The greatest reduction, in three rabbits, was ≥150 μV over 2.5 years. After 1 year, however, the light adapted b-waves were similar to those of rabbits with normal dark adapted b-waves. The majority of the progeny of these rabbits also had small b-waves, which became still smaller in 2 years. Ultrastructural studies of two rabbit retinas of the first generation showed pathological changes of the pigment epithelium (Wrigstad, Hanitzsch & Nilsson, Ultrastructural and electrophysiological studies of the retina and the retinal pigment epithelium in rabbits with low b-wave amplitudes, in preparation). Evidently there is an inheritable defect in the pigment epithelium which first impairs the rod pathway

    vPIF-1 is an insulin-like antiferroptotic viral peptide

    Iridoviridae, such as the lymphocystis disease virus-1 (LCDV-1) and other viruses, encode viral insulin-like peptides (VILPs) which are capable of triggering insulin receptors (IRs) and insulin-like growth factor receptors. The homology of VILPs includes highly conserved disulfide bridges. However, the binding affinities to IRs were reported to be 200- to 500-fold less effective compared to the endogenous ligands. We therefore speculated that these peptides also have noninsulin functions. Here, we report that the LCDV-1 VILP can function as a potent and highly specific inhibitor of ferroptosis. Induction of cell death by the ferroptosis inducers erastin, RSL3, FIN56, and FINO2 and nonferroptotic necrosis produced by the thioredoxin-reductase inhibitor ferroptocide were potently prevented by LCDV-1, while human insulin had no effect. Fas-induced apoptosis, necroptosis, mitotane-induced cell death and growth hormone-releasing hormone antagonist-induced necrosis were unaffected, suggesting the specificity to ferroptosis inhibition by the LCDV-1 VILP. Mechanistically, we identified the viral C-peptide to be required for inhibition of lipid peroxidation and ferroptosis inhibition, while the human C-peptide exhibited no antiferroptotic properties. In addition, the deletion of the viral C-peptide abolishes radical trapping activity in cell-free systems. We conclude that iridoviridae, through the expression of insulin-like viral peptides, are capable of preventing ferroptosis. In analogy to the viral mitochondrial inhibitor of apoptosis and the viral inhibitor of RIP activation (vIRA) that prevents necroptosis, we rename the LCDV-1 VILP a viral peptide inhibitor of ferroptosis-1. Finally, our findings indicate that ferroptosis may function as a viral defense mechanism in lower organisms

    Affective responses as guides to category-based inferences

    Initial nonconscious affective reactions to a target individual may influence a perceiver's selection from among descriptively plausible categories with which to organize his impression of the target. Specifically, a perceiver may be more likely to employ a category that is consistent, in affective tone, with the tone of his affective reaction. Subjects in two studies were exposed to photographs of faces of target individuals. Degree of preference for the faces was manipulated, outside of subjects' awareness, by varying the state of pupillary dilation. Participants in Study One reported that verbal descriptions that characterized positively (compared to negatively) evaluated category prototypes were more likely to be descriptive of targets with dilated pupils. Similarly, participants judged descriptions that characterized negatively (compared to positively) evaluated prototypes as more likely to be descriptive of targets with constricted pupils. In Study Two, subjects' recall of personality descriptions that were (evaluatively) inconsistent with their initial affective response to the target was superior to their recall of descriptions that were (evaluatively) consistent with the tone of their initial response.     Reaction Pathway toward Seven-Atom-Wide Armchair Graphene Nanoribbon Formation and Identification of Intermediate Species on Au(111)

    The prototypical surface reaction of 10,10'-dibromo-9,9'-bianthryl (DBBA) towards the 7-atom-wide armchair graphene nanoribbon (7-AGNR) on the Au(111) surface has been investigated by means of vibrational spectroscopy, thermal desorption spectroscopy and density functional theory. Specifically, a direct correlation between annealing temperature and the formation of various intermediate species is derived. By comparing IR spectra with results from DFT calculations an identification of reaction intermediates has been achieved, allowing for a precise mapping of individual reaction steps. Thereby, we identify a prior unknown partially dehalogenated and strongly tilted DBBA* mono-radical species (DBBA-1Br) after mild annealing (380-450 K). This inclined adsorption geometry stabilizes the second Br atom, preventing full dehalogenation. According to our calculations dimerization of DBBA-1Br reverses this inclination and provides an efficient way to enable abstraction of the second Br atom and initiate polyanthracene chain formation by means of DBBA* addition. As a competitive process to dimerization a minor amount of the DBBA* mono-radicals recombine with previously released bromine atoms leading to associative DBBA desorption. In the course of thermal processing hydrogen release signals planarization of the twisted anthracene units of DBBA by means of cyclodehydrogenation and formation of the final 7-AGNR. Based on the results of this work an in-depth understanding of the on-surface synthesis of 7-AGNR on Au(111), starting with a DBBA molecular precursor, has been attained
