    Six antigen detection-based rapid influenza point-of-care tests were compared for their ability to detect avian influenza A(H7N9) virus. The sensitivity of at least four tests, standardised by viral infectivity (TCID50) or RNA copy number, was lower for the influenza A(H7N9) virus than for seasonal A(H3N2), A(H1N1)pdm09 or other recent avian A(H7) viruses. Comparing detection limits of A(H7N9) virus with Ct values of A(H7N9) clinical specimens suggests the tests would not have detected most clinical specimens

    The influenza surveillance system in Victoria is comprised of several components, including a general practitioner sentinel surveillance system, surveillance for influenza-like illness (ILI) in consultations made by the Melbourne Medical Deputising Service, laboratory confirmed influenza notified to the Victorian Department of Health and strain typing performed by the World Health Organization Collaborating Centre for Reference and Research on Influenza. As measured by ILI from both the MMDS and GPSS, the 2011 influenza season in Victoria was mild compared to previous seasons and was not dominated by any type or subtype of influenza. There were 13 laboratory confirmed influenza outbreaks in 2011, nearly all of which were in aged care facilities. GPs continue to swab more patients, a trend started in 2009, with a significantly lower percent of these testing positive for influenza than previous years. The proportion of ILI and swabbed patients who were vaccinated was also significantly lower in 2011 than previously. Strain analysis undertaken by the WHO Collaborating Centre indicated a good antigenic match between the 2011 vaccine and circulating strains. The Victorian influenza surveillance system continues to provide a reliable, consistent system for monitoring the epidemiology of ILI and laboratory confirmed influenza in Victoria.VIDRL receives support for its influenza surveillance program from the Victorian Government Department of Health.     5D seismic data completion and denoising using a novel class of tensor decompositions

    We have developed a novel strategy for simultaneous interpolation and denoising of prestack seismic data. Most seismic surveys fail to cover all possible source-receiver combinations, leading to missing data especially in the midpoint-offset domain. This undersampling can complicate certain data processing steps such as amplitude-variation-with-offset analysis and migration. Data interpolation can mitigate the impact of missing traces. We considered the prestack data as a 5D multidimensional array or otherwise referred to as a 5D tensor. Using synthetic data sets, we first found that prestack data can be well approximated by a low-rank tensor under a recently proposed framework for tensor singular value decomposition (tSVD). Under this low-rank assumption, we proposed a complexity-penalized algorithm for the recovery of missing traces and data denoising. In this algorithm, the complexity regularization was controlled by tuning a single regularization parameter using a statistical test. We tested the performance of the proposed algorithm on synthetic and real data to show that missing data can be reliably recovered under heavy downsampling. In addition, we demonstrated that compressibility, i.e., approximation of the data by a low-rank tensor, of seismic data under tSVD depended on the velocity model complexity and shot and receiver spacing. We further found that compressibility correlated with the recovery of missing data because high compressibility implied good recovery and vice versa.National Science Foundation (U.S.).     TaqMan real time RT-PCR assays for detecting ferret innate and adaptive immune responses

    AbstractThe ferret is an excellent model for many human infectious diseases including influenza, SARS-CoV, henipavirus and pneumococcal infections. The ferret is also used to study cystic fibrosis and various cancers, as well as reproductive biology and physiology. However, the range of reagents available to measure the ferret immune response is very limited. To address this deficiency, high-throughput real time RT-PCR TaqMan assays were developed to measure the expression of fifteen immune mediators associated with the innate and adaptive immune responses (IFNα, IFNβ, IFNγ, IL1α, IL1β, IL2, IL4, IL6, IL8, IL10, IL12p40, IL17, Granzyme A, MCP1, TNFα), as well as four endogenous housekeeping genes (ATF4, HPRT, GAPDH, L32). These assays have been optimized to maximize reaction efficiency, reduce the amount of sample required (down to 1ng RNA per real time RT-PCR reaction) and to select the most appropriate housekeeping genes. Using these assays, the expression of each of the tested genes could be detected in ferret lymph node cells stimulated with mitogens or infected with influenza virus in vitro. These new tools will allow a more comprehensive analysis of the ferret immune responses following infection or in other disease states

    Oseltamivir-resistant influenza A(H1N1)pdm09 virus in Dutch travellers returning from Spain, August 2012

    Two Dutch travellers were infected with oseltamivirresistant influenza A(H1N1)pdm09 viruses with an H275Y neuraminidase substitution in early August 2012. Both cases were probably infected during separate holidays at the Catalonian coast (Spain). No epidemiological connection between the two cases was found, and neither of them was treated with oseltamivir before specimen collection. Genetic analysis of the neuraminidase gene revealed the presence of previously described permissive mutations that may increase the likelihood of such strains emerging and spreading widely
