    Evaluating Sentiment Classification to Specify Polarity by Lexicon-Based and Machine Learning Approaches for COVID-19 Twitter Data Sets

    As part of data science, sentiment analysis (SA) applied to social media data is a trending research topic. Identifying positive, negative, or neutral opinions or feelings in the text is the attention of sentiment analysis. In the past few years, Social media platforms have become increasingly popular. In this research, natural language processing (NLP) will be employed to extract useful data and information from unstructured text. .The two methods for sentiment analysis covered in this research are the machine-learning method and the lexicon-based method. The paper examines several lexicon approaches to demonstrate the sentiments from Twitter. To increase classification accuracy, it is necessary to use a reliable method with the highest performance. In this study, classifiers such as Support Vector Machine (SVM) and Naive Bayes (NB) were used together with techniques such as TF-IDF (Term Frequency-Inverse Document Frequency) and BOW (Bag of Words). Each algorithm produces a unique outcome. In order to measure the accuracy of classification, metrics such as Precision, Recall, and F-score are considered. This research shows Support Vector Machine (SVM) with TF-IDF is better than other classifiers with an accuracy of 88%

    Synthesis, Characterisation and Biological Evaluation of Dalfampridine Genotoxic Impurities

    From the planned reaction mechanism and chemistry of pyridine nucleus was understood made an attempt to synthesize different pyridine derivatives, that is fampridine genotoxic impurities from waste application of chemical reactions like N-oxidation, Azo formation, N- deoxygenation, selective deoxigenation, Selective diazo Reduction, Nitro to amine reduction of pyridine containing N- oxide, Nitro to amine reduction, Diamide formation. All the targeted molecules synthesized with good yield and high purity. To achieve high purity attention was drawn forwards automated combiflash and it was performed by developing different analytical methods for separation of synthesized genotoxic impurities. The synthesized genotoxic impurities were characterized using hyponated analytical instruments like NMR, LCMS-MS, HPLC Combiflash. The HPLC purity of genotoxic impurities listed below: 1. IMP-DAL 3 - 99.93%, 2. IMP-DAL 6 - 99.70%, 3. IMP-DAL 4 - 99.05%, 4. IMP-DAL 5 - 99.05%, 5. IMP-DAL 7 - 96.62%, 6. IMP-DAL 2 - 94.42%, 7. IMP-DAL 1 - 92.15%. By the above achievement and all the characterization data’s of genotoxic impurities can be used as reference standards in pharmaceutical industry to evaluate the drug substance. Novel produced compounds, assessed for its in-vitro anti-bacterial activity compared to both gram-positive and gram-negative bacteria using the standard drug Gentamycin. It was concluded as IMP-DAL 1, IMP-DAL 2, IMP-DAL 7 exhibited substantial antibacterial activity against its direction Escherichia-coli ˃ Pseudomonas-aeruginosa ˃ Bacillus-subtilis ˃ Staphylococcus-aureus. IMP-DAL 3, IMP-DAL 4, IMP-DAL 5, IMP-DAL 6 showed mild antibacterial activity contrary to gram +ve bacteria and gram -ve bacteria test organisms. 100 μg/ml of synthesized molecules showed more activity compared to other concentrations

    EFFECT OF HYDROALCOHOLIC EXTRACT OF ACHYRANTHES ASPERA ON HALOPERIDOLINDUCED PARKINSON'S DISEASE IN WISTAR RATS

      Objective: Prolonged usage of neuroleptics in psychotic disorders such as schizophrenia provokes extrapyramidal symptoms that are also seen in Parkinson's disease. An attempt has been made to study the neuroprotective role of Achyranthes aspera hydroalcoholic (HA) extract on haloperidol-induced Parkinson's symptoms in Wistar rats.Methods: The present study deals with the antiparkinson effect of HA extract of A. aspera on haloperidol (2 mg/kg intraperitoneal administration)- induced catatonia in Wistar rats. The motor coordination in case of haloperidol-treated animals was studied by performing rotarod test and hang test. Dopamine and 3,4-dihydroxyphenylacetic acid were estimated using an electrochemical detector and high-performance liquid chromatography. The antioxidant status was also assessed to know the neurotoxicity of haloperidol by estimating the levels of lipid peroxidation, superoxide dismutase, glutathione (GSH) peroxidase, and reduced GSH by performing individual assays.Results: All these assessments were done on 24 Wistar rats which were divided into four groups (n=6). HA was administered at 200 and 400 mg/kg doses, 30 minutes before haloperidol treatment for 20 days. HA significantly (*p<0.05, **p<0.01) improved the antioxidant status.Conclusion: The results shown that HA shows a protective role in haloperidol catalepsy and also possess antioxidant property

    Knowledge of the Human Papillomavirus Vaccine: An Analysis using Together for Health Virginia Population Health Survey

    Purpose: The purpose of this analysis was to identify key predictors which impact knowledge of the Human Papillomavirus vaccine in adults aged 21 to 45 in Virginia. Methods: Data was collected from the Together for Health Virginia Population Surveys administered by Virginia Commonwealth University and the University of Virginia. Logistic regression was performed on data using the variables sex, age, rurality, race, education, income, occupation, and type of health insurance coverage. Results: There was a statistically significant positive relationship between knowledge of the HPV vaccine and part-time occupation (OR: 4.288, CI: 1.492-13.325), younger age (OR: 2.31, CI: 1.088-4.905), and higher education (OR: 2.683, CI: 1.227-5.870). There was a statistically significant negative relationship between knowledge of the vaccine and being male (OR: 0.437, CI: 0.248-0.771), living in an urban area (OR: 0.511, CI: 0.267-0.977), and identifying in the lower income category (OR: 0.246, CI: 0.093-0.651). Conclusion: This study identified 6 key predictors in knowledge of the HPV vaccine among adults in Virginia. Future studies should explore, in particular, the category of students and residents of urban areas. Despite these results, knowledge of the HPV vaccine does not translate to intention to receive the vaccine. Therefore, future studies should additionally study attitudes, behaviors, and potential barriers

    Depolarization of sperm membrane potential is a common feature of men with subfertility and is associated with low fertilization rate at IVF

    STUDY QUESTION. Are significant abnormalities in outward (K+) conductance and resting membrane potential (Vm) present in the spermatozoa of patients undertaking IVF and ICSI and if so, what is their functional effect on fertilization success? SUMMARY ANSWER. Negligible outward conductance (≈5% of patients) or an enhanced inward conductance (≈4% of patients), both of which caused depolarization of Vm, were associated with a low rate of fertilization following IVF. WHAT IS KNOWN ALREADY. Sperm-specific potassium channel knockout mice are infertile with defects in sperm function, suggesting that these channels are essential for fertility. These observations suggest that malfunction of K+ channels in human spermatozoa might contribute significantly to the occurrence of subfertility in men. However, remarkably little is known of the nature of K+ channels in human spermatozoa or the incidence and functional consequences of K+ channel defects. STUDY DESIGN, SIZE AND DURATION. Spermatozoa were obtained from healthy volunteer research donors and subfertile IVF and ICSI patients attending a hospital assisted reproductive techniques clinic between May 2013 and December 2015. In total, 40 IVF patients, 41 ICSI patients and 26 normozoospermic donors took part in the study. PARTICIPANTS/MATERIALS, SETTING, METHODS. Samples were examined using electrophysiology (whole-cell patch clamping). Where abnormal electrophysiological characteristics were identified, spermatozoa were further examined for Ca2+ influx induced by progesterone and penetration into viscous media if sufficient sample was available. Full exome sequencing was performed to specifically evaluate potassium calcium-activated channel subfamily M α 1 (KCNMA1), potassium calcium-activated channel subfamily U member 1 (KCNU1) and leucine-rich repeat containing 52 (LRRC52) genes and others associated with K+ signalling. In IVF patients, comparison with fertilization rates was done to assess the functional significance of the electrophysiological abnormalities. MAIN RESULTS AND THE ROLE OF CHANCE. Patch clamp electrophysiology was used to assess outward (K+) conductance and resting membrane potential (Vm) and signalling/motility assays were used to assess functional characteristics of sperm from IVF and ICSI patient samples. The mean Vm and outward membrane conductance in sperm from IVF and ICSI patients were not significantly different from those of control (donor) sperm prepared under the same conditions, but variation between individuals was significantly greater (P< 0.02) with a large number of outliers (>25%). In particular, in ≈10% of patients (7/81), we observed either a negligible outward conductance (4 patients) or an enhanced inward current (3 patients), both of which caused depolarization of Vm. Analysis of clinical data from the IVF patients showed significant association of depolarized Vm (≥0 mV) with low fertilization rate (P= 0.012). Spermatozoa with electrophysiological abnormities (conductance and Vm) responded normally to progesterone with elevation of [Ca2+]i and penetration of viscous medium, indicating retention of cation channel of sperm (CatSper) channel function. LIMITATIONS, REASONS FOR CAUTION. For practical, technical, ethical and logistical reasons, we could not obtain sufficient additional semen samples from men with conductance abnormalities to establish the cause of the conductance defects. Full exome sequencing was only available in two men with conductance defects. WIDER IMPLICATIONS OF THE FINDINGS. These data add significantly to the understanding of the role of ion channels in human sperm function and its impact on male fertility. Impaired potassium channel conductance (Gm) and/or Vm regulation is both common and complex in human spermatozoa and importantly is associated with impaired fertilization capacity when the Vm of cells is completely depolarized

    IN VITRO ACTIVITY OF VARIOUS POTENCIES OF HOMEOPATHIC DRUG THUJA AGAINST MOLDS INVOLVED IN MYCOTIC KERATITIS

    Objective: Isolation and characterisation of clinically isolated fungi from mycotic keratitis and exploration of the in vitro efficacy of various potencies of homeopathic preparations of Thuja occidentalis on the ocular fungal isolates. Methods: Clinical samples were collected from fungal keratitis patients attending a tertiary eye care hospital in Coimbatore, Tamilnadu state, India. The scrapings were also subjected to Gram staining and 10% KOH mount to detect the presence of fungal hyphae. The fungal isolates were subjected to lacto phenol cotton blue (LCB) mount employing cello tape flag method. Homeopathic drug Thuja occidentalis with various potencies viz., Q, 30 C, 200 C, 1 M, 10 M and 50 M were investigated for the growth inhibition of various fungal isolates by plate assay method. Further, a follow up analyses with varying dilutions of Q and 10 M homeopathic potencies of Thuja was carried out for the determination of minimum inhibitory concentration of both microdilution and minimum fungicidal concentration for the Biopolaris isolates. Results: Out of 35 samples analysed, Fusarium spp. (n=5), Aspergillus flavus (n=6), Bipolaris spp. (n=3), Exserohilum spp. (n=3) and Curvularia spp (n=3) were identified. All the potencies of Thuja had good inhibitory activity against Bipolaris spp., followed by Curvularia spp., Exserohilum spp. and Aspergillus flavus. Statistical analysis revealed significant inhibition of all the test isolates by Thuja Q and 50 M. Significant growth inhibition was exhibited by Thuja 30, 200 C, I M for Bipolaris, Exserohilum & A. flavus isolate and Thuja 10 M for all the isolates tested except Fusaria. It was revealed that for the Biopolaris isolates BS1, BS2, BS3 Thuja 10 M and Thuja Q had MIC and MFC of 0.125/10[20,00]0& 0.0625/10 and 0.25/10[20][00]0 & 0.125/10, respectively. Conclusion: The present investigation concludes that homeopathic drug Thuja has good inhibitory activity against the fungi causing keratitis, irrespective of the potencies. It is evident that no definite co-relation exists between various potencies of the same homeopathic drug with regard to their antimycotic properties

    Lifetime of 19Ne*(4.03 MeV)

    The Doppler-shift attenuation method was applied to measure the lifetime of the 4.03 MeV state in 19Ne. Utilizing a 3He-implanted Au foil as a target, the state was populated using the 20Ne(3He,alpha)19Ne reaction in inverse kinematics at a 20Ne beam energy of 34 MeV. De-excitation gamma rays were detected in coincidence with alpha particles. At the 1 sigma level, the lifetime was determined to be 11 +4, -3 fs and at the 95.45% confidence level the lifetime is 11 +8, -7 fs.Comment: 6 pages, submitted to Phys. Rev.

    Measurement of two-halo neutron transfer reaction p(11^{11}Li,9^{9}Li)t at 3AA MeV

    The p(\nuc{11}{Li},\nuc{9}{Li})t reaction has been studied for the first time at an incident energy of 3AA MeV delivered by the new ISAC-2 facility at TRIUMF. An active target detector MAYA, build at GANIL, was used for the measurement. The differential cross sectionshave been determined for transitions to the \nuc{9}{Li} ground andthe first excited states in a wide range of scattering angles. Multistep transfer calculations using different \nuc{11}{Li} model wave functions, shows that wave functions with strong correlations between the halo neutrons are the most successful in reproducing the observation.Comment: 6 pages, 3 figures, submitted to Physical Review Letter

    Lifetimes of states in 19Ne above the 15 O + alpha breakup threshold

    The 15O(alpha,gamma)19Ne reaction plays a role in the ignition of Type I x-ray bursts on accreting neutron stars. The lifetimes of states in 19Ne above the 15O + alpha threshold of 3.53 MeV are important inputs to calculations of the astrophysical reaction rate. These levels in 19Ne were populated in the 3He(20Ne,alpha)19Ne reaction at a 20Ne beam energy of 34 MeV. The lifetimes of six states above the threshold were measured with the Doppler shift attenuation method (DSAM). The present measurements agree with previous determinations of the lifetimes of these states and in some cases are considerably more precise
