    Quantifying Isoniazid Levels in Small Hair Samples: A Novel Method for Assessing Adherence during the Treatment of Latent and Active Tuberculosis.

    BackgroundTuberculosis (TB) is the leading cause of death from an infectious pathogen worldwide and the most prevalent opportunistic infection in people living with HIV. Isoniazid preventive therapy (IPT) reduces the incidence of active TB and reduces morbidity and mortality in HIV-infected patients independently of antiretroviral therapy. However, treatment of latent or active TB is lengthy and inter-patient variability in pharmacokinetics and adherence common. Current methods of assessing adherence to TB treatment using drug levels in plasma or urine assess short-term exposure and pose logistical challenges. Drug concentrations in hair assess long-term exposure and have demonstrated pharmacodynamic relevance in HIV.MethodsA large hair sample from a patient with active TB was obtained for assay development. Methods to pulverize hair and extract isoniazid were optimized and then the drug detected by liquid chromatography/ tandem mass spectrometry (LC/MS-MS). The method was validated for specificity, accuracy, precision, recovery, linearity and stability to establish the assay's suitability for therapeutic drug monitoring (TDM). Hair samples from patients on directly-observe isoniazid-based latent or active TB therapy from the San Francisco Department of Public Health TB clinic were then tested.ResultsOur LC/MS-MS-based assay detected isoniazid in quantities as low as 0.02ng/mg using 10-25 strands hair. Concentrations in spiked samples demonstrated linearity from 0.05-50ng/mg. Assay precision and accuracy for spiked quality-control samples were high, with an overall recovery rate of 79.5%. In 18 patients with latent or active TB on treatment, isoniazid was detected across a wide linear dynamic range.ConclusionsAn LC-MS/MS-based assay to quantify isoniazid levels in hair with performance characteristics suitable for TDM was developed and validated. Hair concentrations of isoniazid assess long-term exposure and may be useful for monitoring adherence to latent or active TB treatment in the setting of HIV

    Scaup Depredation on Arkansas Baitfish and Sportfish Aquaculture

    Lesser scaup (Aythya affinis) and greater scaup (A. marila), hereafter scaup, consume a variety of aquatic invertebrates, plants, and occasionally small fish. Scaup have foraged on commercial aquaculture farms in the southern United States for decades. However, the types, abundance, and rate of fish exploitation by scaup on baitfish and sportfish farms are not well documented. Thus, information is needed to understand how fish and other foods influence scaup use of aquatic resources, and any potential economic effects of depredation of fish. From November–March in winters 2016–2017 and 2017–2018, we conducted 1,458 pond surveys to estimate the abundance and distribution of scaup on Arkansas baitfish and sportfish farms that commercially produce species such as golden shiners (Notemigonus crysoleucas), fathead minnows (Pimephales promelas), goldfish (Carassius auratus), and sunfish (Lepomis spp.). We also collected and processed 531 foraging scaup and quantified the proportion of scaup consuming fish and the proportion of their diet obtained from fish. Fish consumption was highly variable between years. In our survey area, we estimated total fish consumption at 1,400 kg and 60,500 kg for winters 2016–2017 and 2017–2018, respectively. Sunfish ponds experienced the maximum loss (18,000 fish/ha) during winter 2017–2018, while goldfish ponds experienced a loss of just 2,600 fish/ha during the same winter. The estimates of baitfish and sportfish loss to scaup revealed potential management strategies for minimizing fish loss and can inform economic analysis of the financial impact of scaup on producers

    Foraging Ecology and Distribution of Scaup (Aythya spp.) on Arkansas Commercial Baitfish and Sportfish Farms

    Lesser Scaup (Aythya affinis)and Greater Scaup (Aythya marila) have been reported to consume substantial quantities of golden shiners (Notemigonus crysoleucas), fathead minnows (Pimephales promelas), goldfish (Carassius auratus), and sunfish (Lepomis spp.) produced on Arkansas commercial baitfish and sportfish farms. The goals of this study were to investigate foraging ecology and distribution of Scaup at these facilities, and use this information to assist producers in administering bird harassment efforts more efficiently. During typical wintering period for Scaup in Arkansas (November-March), we conducted approximately 1,400 pond surveys to estimate abundance and distribution of scaup on farms in 2016-2017 and 2017-2018. Information related to pond size, fish species, fish size, and stocking density, were also obtained to enable a more detailed analysis of Scaup use. We also collected 561 Scaup from these facilities to quantify the proportion of diet obtained from fish. There was an increase in Scaup abundance and fish consumption between the first to the second winter, likely attributed to cooler temperatures during the second winter. Our distribution model predicted an increased probability of Scaup use on larger ponds containing high densities of fish, while diet analysis indicated increased fish consumption during colder winter periods. Our results can be used by farm managers to designate resources for bird harassment to particular locations and times of the winter when scaup are more likely to negatively impact the fish crop

    Food habits of wintering double-crested cormorants in the Mississippi Delta

    Given its ubiquity, it is not surprising that agriculture, including fin fish aquaculture, contributes to food webs worldwide and is used by numerous wildlife for foraging and meeting other needs. Double-crested Cormorants (Phalacrocorax auritus) impact United States commercial aquaculture and are considered the primary avian predator in catfish (Ictalurus spp.) aquaculture facilities in the Mississippi Delta. Recent changes in aquaculture practices, regulatory policies, and decreased overall hectares in production prompted this study that assessed cormorant consumption of catfish in relation to their night roosts through surveys and diet analysis. Cormorants were collected from night roosts from October through April 2016–2018 (n=69 collections). On average, catfish constituted 33% of a cormorant\u27s overall diet, which is less than reported in previous studies. There was no statistical difference between consumption of channel (I. punctatus) and hybrid catfish (I. punctatus x I. furcatus) based on biomass estimates, and the greatest consumption of catfish occurred in the months of February and March. The best fit model for predicting catfish consumption was the cubic polynomial function of the area of catfish aquaculture within a 30.6 km forage buffer of a night roost. Our findings will inform wildlife managers about relationships between cormorant night roost locations and consumption of catfish and aid decision making with respect to cormorant management. Despite cormorants having shifted consumption to naturally occurring fish species associated with changes to aquaculture, aquaculture remains an important part of regional food webs

    Depredation Impact of Double-Crested Cormorants (Phalacrocorax auritus) on Commercial Catfish Production in the Mississippi Delta

    Double-crested Cormorants (Phalacrocorax auritus) impact United States commercial aquaculture and are considered the greatest avian predators on catfish (Ictalurus spp.) aquaculture facilities. Cormorants are especially problematic in the Delta region in western Mississippi, where catfish production is concentrated providing ideal wintering and foraging areas. Although cormorant/aquaculture dynamics have been studied, recent changes in aquaculture practices, regulatory policies, and decreased overall hectares in production merit contemporary research. Therefore, we estimated abundance and distribution of cormorants at their night roosts and assessed diet related to catfish consumption. Aerial surveys of cormorant night roosts were flown from October through April, 2016-2018. Following each survey, three active night roosts were randomly selected for harvesting cormorants for later necropsy and stomach contents assessment. We completed 25 total surveys and counted an average of 23,379 cormorants (range 5,026 to 40,535) pooled over years (corrected for observer and method bias). A total of 728 cormorants from 27 different night roosts were collected across years. Survey count models estimated 4.2 and 5 million cormorant forage days in the Delta during winters 2016-2017 and 2017-2018, respectively. Throughout the study, catfish comprised 33% of the prey biomass detected; shad (Dorosoma spp.) also were dominant (58%) prey. Evidence suggests that the area of catfish aquaculture surrounding a night roost within a 30.6-km forage buffer is an important predictor for a bird’s relative amount of catfish consumption. These results will inform wildlife managers regarding relationships between cormorant night roost locations in the Delta and disproportionate consumption of catfish, enhancing techniques to reduce fish losses on aquaculture facilities

    <sup>18</sup>F-FDG PET/MRI for restaging esophageal cancer after neoadjuvant chemoradiotherapy

    PURPOSE: The purpose of this study was to investigate whether 18F-fluorodeoxyglucose ( 18 F-FDG) PET/MRI may potentially improve tumor detection after neoadjuvant chemoradiotherapy (nCRT) for esophageal cancer. METHODS: This was a prospective, single-center feasibility study. At 6-12 weeks after nCRT, patients underwent standard 18 F-FDG PET/computed tomography (CT) followed by PET/MRI, and completed a questionnaire to evaluate burden. Two teams of readers either assessed the 18 F-FDG PET/CT or the 18 F-FDG PET/MRI first; the other scan was assessed 1 month later. Maximum standardized uptake value corrected for lean body mass (SUL max ) and mean apparent diffusion coefficient (ADC mean ) were measured at the primary tumor location. Histopathology of the surgical resection specimen served as the reference standard for diagnostic accuracy calculations. When patients had a clinically complete response and continued active surveillance, response evaluations until 9 months after nCRT served as a proxy for ypT and ypN (i.e. 'ycT' and 'ycN'). RESULTS: In the 21 included patients [median age 70 (IQR 62-75), 16 males], disease recurrence was found in the primary tumor in 14 (67%) patients (of whom one ypM+, detected on both scans) and in locoregional lymph nodes in six patients (29%). Accuracy (team 1/team 2) to detect yp/ycT+ with 18 F-FDG PET/MRI vs. 18 F-FDG PET/CT was 38/57% vs. 76/61%. For ypN+, accuracy was 63/53% vs. 63/42%, resp. Neither SUL max (both scans) nor ADC mean were discriminatory for yp/ycT+ . Fourteen of 21 (67%) patients were willing to undergo a similar 18 F-FDG PET/MRI examination in the future. CONCLUSION: 18 F-FDG PET/MRI currently performs comparably to 18 F-FDG PET/CT. Improvements in the scanning protocol, increasing reader experience and performing serial scans might contribute to enhancing the accuracy of tumor detection after nCRT using 18 F-FDG PET/MRI.     Supplementation of a western diet with golden kiwifruits (Actinidia chinensis var.'Hort 16A':) effects on biomarkers of oxidation damage and antioxidant protection

    <p>Abstract</p> <p>Background</p> <p>The health positive effects of diets high in fruits and vegetables are generally not replicated in supplementation trials with isolated antioxidants and vitamins, and as a consequence the emphasis of chronic disease prevention has shifted to whole foods and whole food products.</p> <p>Methods</p> <p>We carried out a human intervention trial with the golden kiwifruit, Actinidia chinensis, measuring markers of antioxidant status, DNA stability, plasma lipids, and platelet aggregation. Our hypothesis was that supplementation of a normal diet with kiwifruits would have an effect on biomarkers of oxidative status. Healthy volunteers supplemented a normal diet with either one or two golden kiwifruits per day in a cross-over study lasting 2 × 4 weeks. Plasma levels of vitamin C, and carotenoids, and the ferric reducing activity of plasma (FRAP) were measured. Malondialdehyde was assessed as a biomarker of lipid oxidation. Effects on DNA damage in circulating lymphocytes were estimated using the comet assay with enzyme modification to measure specific lesions; another modification allowed estimation of DNA repair.</p> <p>Results</p> <p>Plasma vitamin C increased after supplementation as did resistance towards H<sub>2</sub>O<sub>2</sub>-induced DNA damage. Purine oxidation in lymphocyte DNA decreased significantly after one kiwifruit per day, pyrimidine oxidation decreased after two fruits per day. Neither DNA base excision nor nucleotide excision repair was influenced by kiwifruit consumption. Malondialdehyde was not affected, but plasma triglycerides decreased. Whole blood platelet aggregation was decreased by kiwifruit supplementation.</p> <p>Conclusion</p> <p>Golden kiwifruit consumption strengthens resistance towards endogenous oxidative damage.</p
